Immune response in farm animals infected or immunised with bacteria of Chlamydia sp. and Chlamydophila sp. genus.

Bacteria from genera Chlamydia (Ch.) and Chlamydophila (Chl.) are very pathogenic and may infect humans and animals. They also may cause latent infection, especially in animals. In this paper we discuss the non-specific and specific cellular and humoral immunity in farm animals, after infection or immunisation with Chlamydia sp. and Chlamydophila sp. bacteria. It has been shown, that the infection or immunisation with the microorganisms influenced the activity of polimorphonuclear cells (PMN) and mononuclear cells (MN) in the process of phagocytosis. It has also been shown that the bacteria influenced the amount and activity of lysozyme, activities of myeloperoxidase and lysosomal enzymes. Infection or immunisation with the microorganisms was demonstrated to affect numbers of lymphocytes T and B and those of their subpopulation as well as the activity of cytokines and levels of serum and secreted immunoglobulins. The changes were detected just a few hours after infection or immunisation and persisted for a few days to a few decades.

Values of selected immune and haematological parameters in healthy rabbits.

In the paper, values of selected indices of non-specific immunity as well as white and redblood cell patterns were presented in healthy rabbits at the age of 3 to 4,5 months. The purpose of the study was to provide reference values in healthy rabbits being frequent research model in the studies of several diseases in humans and animals.

Potential cidal activity of neutrophilic granulocytes in rabbits immunised with Chlamydia psittaci-Gocaltovo strain.

The potential oxygen-dependent cidal activity of neutrophilic granulocytes was examined in rabbits immunised with Ch. psittaci-Gocaltovo strain. Tests included a nitrotetrazolium blue (NBT) reduction test and estimation of specific antibody titers. The results obtained indicated that immunization of rabbits with Ch. psittaci-Gocaltovo strain induced increased cidal activity of neutrophilic granulocytes four weeks earlier than the augmented titers of specific antibodies.

Non-specific humoral immunity in rabbits immunised with Chlamydia psittaci-Gocaltovo strain.

In this study we monitored parameters of non-specific humoral immunity in rabbits immunised with Ch. psittaci-Gocaltovo strain and looked for specific anti-Chlamydia antibodies. The results obtained indicated that the parameters evaluated, including activity of myeloperoxidase and the level and activity of lysozyme, manifested alterations earlier than positive titers of specific antibodies developed. This may indicate that alterations in parameters of non-specific humoral immunity can be used as accessory tests in the diagnosis of chlamydioses.

Lymphocytes and their subpopulations in peripheral blood of rabbits immunised with Chlamydia psittaci-Gocaltovo strain.

The effects of immunisation with Chlamydia psittaci-Gocaltovo strain were examined on the dynamic alterations in lymphocytes T and B and in their subpopulations in the peripheral blood of rabbits; the titer of specific anti-Chlamydia antibodies was also established. The results showed that the examined strain induced alterations in the form of an increased or decreased number of T and/or B lymphocytes and in their subpopulations, and that immunity conditioned by the cells proved to be a very significant variable in infection with Chlamydia psittaci (at present Chlamydophila abortus). Alterations in the immune pattern developed earlier by 35 days as compared to positive titers of specific antibodies.

Non-specific cell-mediated immunity in rabbits experimentally infected with four different doses of VHD (viral hemorrhagic disease) virus, French strain Fr-2.

Alterations were examined in non-specific cell-mediated immunity in rabbits experimentally infected with four different fractions of lethal dose (25, 50, 75 or 100%) of VHD virus, French strain Fr-2. The results indicated that in groups of rabbits infected with viral doses of VHD distinct patterns of individual parameters studied were observed, and intensity of alterations did not always correspond to the administered fraction of lethal dose of the VHD virus.

Dynamic alterations in peripheral blood lymphocytes in rabbits experimentally infected with VHD (viral hemorrhagic disease) virus--Polish strain Kr-1.

In this study dynamic alterations were examined in the number of lymphocytes T, B, Th, Tc/Ts and activated lymphocytes T and B of rabbits experimentally infected with a 100% lethal dose of VHD virus, Polish strain Kr-1. Results of the studies demonstrated that the Polish strain Kr-1 can be regarded as less immunogenic than the French strain, Fr-2. This may indicate the existence of immunotypes within VHD viruses.

Dynamic alterations in selected indices of non-specific cell-mediated immunity in rabbits experimentally infected with VHD (viral hemorrhagic disease) virus.

Results of studies are presented on non-specific cell-mediated immunity in rabbits experimentally infected with two strains of VHD virus, differing in their biological properties. The results indicate that the applied strains exhibit different immune reactivities, which points to the presence of immunotypes within the virus.

Ingestion capacity of PMN cells in peripheral blood of rabbits experimentally infected with VHD (viral haemorrhagic disease) virus strains originating from various biotopes.

The study presents results of investigations on ingestion capacity in rabbits experimentally infected with two French strains (Fr-1 and Fr-2) and four Polish strains (KGM, SGM, MAL, Kr-1) of VHD virus. The results proved that the strains studied exhibited distinct reactivity even if the VHD virus is thought to be uniform in the biological respect.

Serological response of cattle to Chlamydophila abortus in Slovakia in 1996-2000.

In the Slovak Republic, in 1966-2000, 37,275 blood sera of cattle were investigated for the presence of antibodies against Chlamydophila abortus using the method of complement fixation. The antibody occurrence had following tendency: in 1996--3.72%; 1997--10.02%; 1998--9.15%; 1999--15.99%; 2000--9.51% of the tested sera contained the antibodies. In most cases, antibodies in low titres, 1:32-1:64, were detected. Positive serological reactions at such serum dilutions are not indicative of the clinical disease of cattle; they reflect an immune response of the host organism following contact with the Chlamydophila abortus antigen. The chlamydial antibody titres of 1:256, which were confirmed in 1998-1999, indicate the chlamydial infections.

[Superantigens: new data].

The paper pertains novel data on action of bacterial and viral superantigens on the immune system. Against the background, new morbid conditions have been described, which may result from the action of superantigens. In the context of superantigen action on the immune system, antigen pathways in the system have been described. The review has been based on laboratory results obtained during recent 5 years in various scientific centers in the world.

Activity of neutrophilic granulocytes in rabbits immunized with Chlamydia psittaci.

The studies were performed on rabbits preimmunized with killed Chlamydia psittaci antigen (Czech isolate). In the blood of the animals nitrotetrazolium blue reduction test (spontaneous and stimulated) was performed, coefficient of polymorphonuclear cell metabolic activity in NBT test was calculated and myeloperoxidase activity was estimated, haematologic parameters were established and antibodies to Chlamydia psittaci were searched for using complement fixation test. The animals were subjected to clinical examination and their housing was tested for its zoohygienic standard. Analysis of the results demonstrated decreased activity of polymorphonuclear cells 3 to 4 weeks before appearance of anti-Chlamydia antibodies, as detected by complement fixation test.

[Activity of neutrophilic granulocytes and hematologic picture in rabbits immunized with chlamydia psittaci].

The study was aimed at investigation of selected elements of nonspecific immunity (spontaneous and stimulated NBT reduction test and myeloperoxidase activity of neutrophilic granulocytes) and specific serological response in rabbits immunized twice with suspension of cells of C. psittaci inactivated by formaldehyde. Specific antibodies were detected by complement fixation test and in a titer of 16 or higher were found between 42 and 56 day of the experiment. Statistically significant decrease of myeloperoxidase activity of granulocytes was observed between 14 and 21 day of the experiment, while diminishing values of results of spontaneous NBT reduction test was seen mainly between 35 and 56 day of experiment. The authors conclude that obtained results suggest occurrence of periodical decrease of nonspecific immunity may occur after immunization of animals with inactivated C. psittaci suspension.

Cell-mediated and humoral immunity in bulls infected with IBR/IPV virus (BHV 1).

Time-related changes in specific cell-mediated immunity (CMI) and in humoral immunity were monitored in 20 bulls, aging 12 to 16 months, in four groups, each consisting of 5 animals. Group I was experimentally and group III-naturally infected with BHV 1 virus, groups II and IV serving as control animals. Tests for CMI included delayed type hypersensitivity (DTH), leukocyte migration inhibitory factor (LMIF) and granulocyte migration inhibitor factor (GMIF-LIF-buffy coat leukocyte migration inhibitory factor) in the presence of BHV I antigen while humoral immunity was tested by serum induced neutralization of the virus (SN) and by estimation of serum IgG, IgG1, IgG2, IgM and IgA levels. Immunologic, virologic and clinical studies were performed two days before infection and 17 timed within 91 days after the infection in bulls of group I and II and 7 times within 42 days after developing the disease in bulls of groups III and IV. The results showed that positive CMI reactions appeared 3 to 4 weeks earlier than positive antibody titers in SN test. The antibodies belonged most probably to IgG1 and IgG2 subclasses.

[Phagocytic activity of neutrophils (PMN cells) in cattle infected with IBR/IPV virus (bovine herpesvirus 1--BHV1)].

PMN cell random and directed migration, adherence, aggregation, ingestion of bacteria as well as resting and stimulated NBT reduction activities were studied in two groups of 5, 2-4 months-old bullocks experimentally (group I) or naturally (group III) infected with BHV1. Control groups (II and IV) consisted of 5 healthy calves. Experimental infection of bullous were carried out using Polish field isolate of BHV1; animals were injected intramuscularly, intravenously and into conjunctival sack with 3 ml, 1 ml and 0.5 ml of the virus respectively. PMN cell phagocytic activities and virological examinations were performed in the group I and II control 2 days before, at the day, and at 1, 2, 3, 4, 7, 14, 21, 28, 35, 42, 64 days post infection, while in the groups III and IV at 1, 7, 14, 21, 28, 42 and 52 day of the experiment. Directed migration, ingestion of bacteria and NBT reduction by resting and stimulated PMN cells activities were decreased whereas random migration, adherence and aggregation abilities as well as number phagocytosing cells were increased both in naturally and experimentally infected animals. These changes were noted in experimentally infected animals at 1-7 day post infection, its three weeks before antibody anti BHV1 and lasted for 3-9 weeks, while in naturally infected bullocks they appeared at 1-28 day of the experiment and persisted for 4-8 weeks the virus was easy isolated from nasal swabs especially of naturally infected animals.

Phagocytic activity of peripheral blood leukocytes in bulls experimentally infected with IBR/IPV virus (bovid herpesvirus 1).

Functional status of peripheral blood leukocytes was evaluated in bulls infected experimentally with BHV 1. Results of the studies indicated extensive changes in mechanisms of phagocytosis which play significant role restricting spread of viral infection, particularly at the preliminary stage of the infection. Increase in serum antibody titer appeared as a result of a prolonged contact of BHV 1 with cells of the immune system of the organism. The studies, documenting changes in reactivity of peripheral blood phagocytic cells in BHV 1 - infected bulls, point to new possibilities of evaluating immune status in the cattle infected with the virus.

[Formation of levels of serum immunoglobulins (G1, G2, M and A) in fattened bulls].

The content of serum IgG1, IgG2, IgM and IgA was determined 80 young bulls, black and white aging 2-9 months, originating from individual and large farms. The animals were kept for 40 days in various environments (calf shed with a litter, calf shed of Bisprol 3000, adapted grate calf shed, pasteure) and then transferred into a fattening house of the farm Bisprol 3000 for 160 days. The level of serum immunoglobulins was determined by the method of radial immunodiffusion using specific antisera against bovine immunoglobulins of the own production. It was found that environment influenced the content of serum IgG1, IgM and IgA and depended on age origin of animals. However, the content of IgG2 depended only on age and origin of young bulls. The constant level serum IgG1 reached at the age of 6.5-7.7 months, IgM and IgA at the age of 5.0-5.5 months and IgG2 at the age above 9.0 months. The following mean of serum immunoglobulins in this time were found: IgG1 12.4 milligram, IgM 6.3 milligram and IgA 0.37 milligram.
