Improvement in the sperm penetration (hamster ova) assay (SPA) results after doxycycline treatment of infertile men.

Thirty-two men with abnormal (less than or equal to 11% penetration) Sperm Penetration (hamster ova) Assay (SPA) results were treated with doxycycline and reevaluated by SPA and seminal fluid analysis. Eighteen of the 32 infertile men with initially abnormal SPAs had normal SPAs (greater than or equal to 15% egg penetration) after treatment. None of 30 untreated infertile men with abnormal SPAs had a normal SPA on repeat testing (P less than 0.001). Improvement in SPA results was associated with a decrease in the number of leukocytes/ml in seminal fluid. Seven of the wives of the treated men conceived, while none of the wives of the untreated infertile men conceived during comparable intervals of time.

Body mass indices and skeletal size in 394 Canadians aged 55-86 years.

Demispan (finger web to midline) is the preferred index of stature in the study of the elderly where height is unreliable because of age-related loss or distortion. As a consequence, two alternative mass indices, Demiquet (male mass. demispan-2) and Mindex (female mass. demispan-1), have been proposed for use in the elderly, in place of body mass index calculated from height, which might be suspect. These gender-specific indices reflect the squared and linear relationship of height to mass in men and women respectively. To assess the independence of Demiquet and Mindex from stature and the reliability of demispan, height, demispan (second fingerweb to midline), and mass were recorded in a random sample of independent Canadian men (182) and women (212) aged 55-86 years. Both Demiquet and Mindex were highly correlated with mass, r = 0.81 (p = 0.001) and r = 0.96 (p = 0.001), respectively. In both gender groups these mass indices were independent of height and demispan. The demispan within- and between-observer errors were small -0.04(-0.16 to 0.08)cm and -0.09(-0.22 to 0.03) cm and the between-observer intraclass correlation estimate of reliability was 0.96 (lower-bound one-sided 95% CI 0.90). Demispan is a reliable and reproducible measure of stature in the elderly. Demiquet and Mindex are useful mass indices, independent of stature and suitable for use in the study of the elderly where the use of mass indices based on height may be questionable.

Aspartyl proteases from the intestinal brush border of Haemonchus contortus as protective antigens for sheep.

A novel pepsin-like aspartyl protease was identified as a component of Haemonchus galactose-containing glycoprotein (H-gal-GP), which is an integral membrane glycoprotein complex located on the intestinal cells of Haemonchus contortus, and a highly protective antigen for sheep. This molecule, designated HcPEP2, showed 50% sequence identity with a previously described aspartyl protease from H-gal-GP known as HcPEP1. Fractions of H-gal-GP, either containing both HcPEP1 and 2 or other lower molecular weight components of the complex, were evaluated as protective antigens in immunization - challenge trials in sheep. When separated from the rest of the complex by gel filtration in 8 m urea, the HcPEP1 and 2 fraction significantly reduced H. contortus egg counts by 48% and worm numbers by 36%, but the lower molecular weight components were not significantly protective. However, the HcPEP1 and 2 fraction did not protect if electro-eluted from SDS-dissociated H-gal-GP, nor did bacterially expressed recombinant HcPEP1, suggesting that conformational epitopes are important for inducing immunity.

Immunization against sheep scab: preliminary identification of fractions of Psoroptes ovis which confer protective effects.

In an attempt to enrich for potentially protective Psoroptes ovis antigens, three separate vaccine trials were conducted in which groups of sheep were immunized three times with various fractions of a soluble extract of P. ovis mites using QuilA as adjuvant. These groups, as well as controls that received adjuvant only, were challenged with P. ovis, and protective immunity was assessed by measuring lesion areas and conducting mite counts 4 and 6 weeks later. All fractions stimulated high titre serum antibodies. Most conferred some protection on sheep with active disease, although there was considerable variation between sheep in all groups, including the controls. Some fractions were more protective than the extract itself, suggesting that the protective components had been concentrated. Indeed the best fraction, obtained by ion exchange chromatography, followed by a gel filtration step, slowed lesion growth to less than a third by 6 weeks after challenge and reduced mite numbers by more than 13 times compared to control sheep vaccinated with QuilA only. However, as judged by polyacrylamide gels, the polypeptide profile of this fraction was still complex, indicating that further work is required to identify the protective components.
