Structural relationship between the two small hydrophobic apoproteins in bovine pulmonary surfactant.

Lipid extracts of bovine pulmonary surfactant contain two very hydrophobic surfactant-associated proteins (SP) designated SP-B (15 kDa nonreduced) and SP-C (3.5 kDa). These two low molecular weight apoproteins were delipidated and purified on silica SEP-PAK cartridges using various reagents. Dansylation studies revealed that the 15 kDa apoprotein has three N-termini: Phe, Leu and Ile, while the 3.5 kDa apoprotein has two N-termini: Leu and Ile. In either protein, only a very small amount of N-Ile is present. Quantitative N-terminal dansylation analysis of the 15 kDa protein indicated that Phe and Leu (plus Ile) are present in a 1:1 ratio. Carboxy-terminal analysis showed that the 15 kDa protein contains C-terminal Gly, and the 3.5 kDa protein contains C-terminal Leu. Gas-phase amino terminal sequencing of the 15 kDa protein revealed almost exclusively the Phe-polypeptide (SP-B). These results suggest that the 15 kDa apoprotein is not an oligomer of SP-B and SP-C. The reason that analysis of SP-B reveals N-terminal Leu and Ile by dansylation which cannot be confirmed by amino acid sequencing is not known.

Active immunization against gonadotropin-releasing hormone combined with androgen supplementation is a promising antifertility vaccine for males.

Male rats and rabbits were immunized against gonadotropin-releasing hormone (GnRH) conjugated to tetanus toxoid (GnRH10-TT) using only materials approved for humans. Testosterone (T)-releasing implants or the long-lasting T ester testosterone-17-trans-4-n-butyl-cyclohexane carboxylate (TE) was used as supplemental androgen for maintaining libido. Immunization against GnRH10-TT effectively suppressed fertility (spermatogenesis) in rats and rabbits. Neither T nor TE administration restored fertility. Both androgens were effective in maintaining normal libido in rats. TE, which is not hydrolyzed in rabbits, was less effective in maintaining normal ejaculatory behavior in this species. Active immunization against GnRH could be a convenient and cost-effective method of fertility control in males.

Characterization of the small hydrophobic proteins associated with pulmonary surfactant.

Lipid extracts of bovine pulmonary surfactant, which retain many of the biophysical characteristics of natural surfactant, contain approx. 98% lipid and 2% protein, as determined by amino acid analysis. Polyacrylamide/urea gel electrophoresis reveals that lipid extract surfactant contained a major apoprotein band with apparent Mr 3500 and minor apoprotein bands with apparent Mr 15,000 and 7000. After reduction, the 15 kDa band disappears and is replaced by a prominent band with apparent Mr = 5000. Reduction also results in a relative diminution of the 7 kDa band and a relative increase in the intensity of the 3.5-kDa band. Edman degradation reveals two major peptide sequences which have been designated surfactant-associated peptide (N-terminal Phe) and surfactant-associated peptide (N-terminal Leu) and a minor sequence designated surfactant-associated peptide (N-terminal Ile). The latter surfactant-associated peptide appears to be related to the N-terminal Leu peptide but lacks the terminal Leu. N-Terminal analysis by dansylation demonstrates that the 15 and 5 kDa (reduced) apoprotein species contain N-terminal Phe, Leu and Ile. The 3.5 and 7 kDa bands contain only N-terminal Leu and Ile. Chromatography of lipid extracts on silicic acid columns gives rise to fraction I, which contains protein and phosphatidylglycerol, and fraction II, which contains protein, phosphatidylglycerol and phosphatidylethanolamine. Fraction I was primarily composed of the 15-kDa apoproteins, while fraction II contained mainly the 3.5 and 7 kDa apoproteins. Both fractions exhibited biophysical activity after reconstitution with dipalmitoylphosphatidylcholine. These results indicate that lipid extracts contain an oligomer of 15 kDa containing surfactant-associated peptide (N-terminal Phe) and surfactant-associated peptides (N-terminal Leu or Ile) which interact through sulfhydryl and perhaps other bonds. Lipid extracts also contain 3.5 kDa monomers of surfactant-associated peptides with N-terminal Leu and N-terminal Ile which can dimerize through sulfhydryl and perhaps hydrophobic interactions.

Magnetic resonance imaging of aneurysms of the abdominal aorta.

Experience with magnetic resonance imaging (MRI) of 10 normal control patients and 13 patients with known abdominal aortic aneurysms is presented. The aorta, renal artery origins, and iliac arteries were clearly identified in all control patients. Ten out of 13 aneurysms were optimally visualized. Three cases early in the experience were poorly seen and permitted less confident measurements. Renal artery origins were identified in 12 cases and judged to be uninvolved by the aneurysm in 11. Iliac arteries were identified in 11 cases and judged to be definitely aneurysmal in three. Sonography clearly depicted all 13 aneurysms. Correlation of the actual anteroposterior (AP) and transverse diameter aneurysm measurements with both techniques was excellent. Sonography failed to identify the renal artery origins in all cases, but could infer noninvolvement in two cases. Iliac arteries were seen in eight cases and judged definitely aneurysmal in three. According to these preliminary results, both MRI and sonography provide accurate AP and transverse outer wall measurements. MRI is superior in determining the presence of renal and/or iliac involvement. The authors recommend, however, that sonography continue to be the preliminary screening method.

Characterization of an isozyme of S-adenosylmethionine synthetase from rat liver.

An isozyme of S-adenosylmethionine synthetase has been purified to homogeneity by ammonium sulfate fractionation, DEAE-cellulose column chromatography, and gel filtration on a Sephadex G-200 column. The purified enzyme is very unstable and has a molecular weight of 120 000 consisting of two identical subunits. Amino acid analysis on the purified enzyme showed glycine, glutamate, and aspartate to be the most abundant and the aromatic amino acids to be the least abundant. It possesses tripolyphosphatase activity which can be stimulated five to six times by S-adenosylmethionine (20-40 microM). The findings support the conclusion that an enzyme-bound tripolyphosphate is an obligatory intermediate in the enzymatic synthesis of S-adenosyl-methionine from ATP and methionine.

Regulation of the immune response to tumor antigens. X. Activation of third-order suppressor T cells that abrogate anti-tumor immune responses.

The experiments described further define the suppressor T cell pathway in the S1509a tumor system. We demonstrated previously that S1509a-induced Ts1, TsF1, and Ts2 specifically suppress in vivo Ly1+2- T cell-dependent responses to S1509a and that Ts1 suppress in vivo Ly-1+2- T cell-mediated proliferative responses to S1509a. We have now shown that in vivo administration of either S1509a-induced TsF1 or TsF2 suppresses both in vivo and in vitro Ly-1+2- T cell-mediated responses to S1509a. Furthermore, we revealed the existence of Ts3, which are activated by S1509a tumor antigen and TsF2, in this murine tumor system. Finally, we demonstrated that cyclophosphamide abrogates the suppressive effect of TsF2 but not that of Ts3. These results are discussed with respect to T cell-mediated suppression in other murine tumor systems and the possible pivotal role for a tumor antigen-presenting cell in activating Ts3 in the S1509a tumor system.

Bronchopulmonary moniliasis in the newborn.

Ten instances of bronchopulmonary moniliasis due to C. albicans in neonatal infants, 1 to 11 days of age, are described. The incidence was 1.05% out of a total of 947 consecutive necropsies on neonates. Diagnosis was established by morphological identification of the fungi. Seven cases were associated with inflammatory exudation. In three cases inflammation was absent. Infection secondary to monilial vaginitis in the mother appears to have been over-emphasized. The more common mechanism appears to be aspiration after delivery.

WW and EF hand domains of dystrophin-family proteins mediate dystroglycan binding.

The membrane-spanning dystrophin glycoprotein complex mediates an indirect linkage between the actin-based cytoskeleton and the extracellular matrix. Although expressed by diverse cell types, genetic lesions of members of this complex result in muscular dystrophy phenotypes emphasizing the importance of these interactions in muscle cells. We have characterized interactions between dystrophin family members and dystroglycan: cytoskeletal and transmembrane components of the complex, respectively. Our results demonstrate that both the WW and EF hand domains of dystrophin and utrophin, an autosomal homologue of dystrophin, directly bind the cytoplasmic domain of dystroglycan. Furthermore, alpha-dystrobrevin, a more distantly related dystrophin family member which lacks a WW domain but contains the EF hand domain, binds dystroglycan. This is the first demonstration of a direct interaction between a dystrobrevin or utrophin and dystroglycan, and has implications for the organization of the dystrophin glycoprotein complex and the use of dystrophin homologues in muscular dystrophy therapy.

Does inferior vena cava size predict right atrial pressures in patients receiving mechanical ventilation?

The inferior vena cava diameter and its respiratory response are used to estimate right atrial pressures in spontaneously breathing patients but its value in patients receiving mechanical ventilation is unvalidated. Forty-nine patients undergoing mechanical ventilation were prospectively evaluated in the intensive or coronary care units with two-dimensional echocardiography of the inferior vena cava and simultaneous measurements of mean right atrial pressures by central venous or pulmonary artery catheter. Correlation between inferior vena cava diameter at expiration and mean right atrial pressure was only 0.58. The correlation between inspiratory change in inferior vena cava diameter and mean right atrial pressure was poor (r = 0.13). Despite these correlations, an inferior vena cava diameter of < or = 12 mm predicted a right atrial pressure of 10 mm Hg or less 100% of the time, but sensitivity was only 25%. An inferior vena cava diameter > 12 mm had no predictive value for right atrial pressure.

Design and assembly of an 8 tesla whole-body MR scanner.

PURPOSE: The purpose of this report is to describe the design and construction of an 8 T/80 cm whole-body MRI system operating at 340 MHz. METHOD: The 8 T/80 cm magnet was constructed from 414 km of niobium titanium superconducting wire. The winding of this wire on four aluminum formers resulted in a total inductance of 4,155 H. Gradient subsystems included either a body gradient or a head gradient along with a removable shim insert. The magnet and gradient subsystems were interfaced to two spectrometers. These provided the control of the gradient amplifiers and the two sets of four RF power amplifiers. The latter provide in excess of 8 kW of RF power from 10 to 140 MHz and 10 kW of RF power from 245 to 345 MHz. A dedicated computer-controlled patient table was designed and assembled. The entire system is located in a clinical setting, facilitating patient-based studies. RESULTS: The 8 T/80 cm magnet was energized without complication and achieved persistent operation using 198.9 A of current, thereby storing 81.5 MJ of magnetic energy. Exceptional performance was observed for nearly all components both in isolation and when combined within the complete system. CONCLUSION: An 8 T/80 cm MRI system has been assembled. The magnet subsystem is extremely stable and is characterized by good homogeneity and acceptable boil-off rates.

The hamstring index.

The purpose of this study was to assess the limits of hamstring tightness in 369 children by using three common tests. The straight leg-raise test averaged 100 degrees at birth, increasing to 110 degrees at age 1 year, before decreasing to 80 degrees by age 5-6 years. It then remained constant to skeletal maturity. Hamstring tightness is present if this angle is < 80 degrees in children younger than 2 years. After this age, the limit decreased steadily to plateau at 60 degrees by the age of 6 years. The popliteal angle measured the maximum 180 degrees from birth to age 2 years. This angle then decreased to average 155 degrees by age 6 years and remained steady. An angle < 125 degrees suggests significant hamstring tightness. The touching-toes test showed minimal variation between age groups averaging, 1 +/- 3 cm. Distances reached < -5 cm in children aged up to 6 years and < -15 cm in those older than 6 indicate excessively tight hamstrings. Little difference was seen between sides, although girls demonstrated less hamstring tightness. All tests showed a positive correlation with the grade of ligamentous laxity, but no association with the child's stage of puberty could be detected.
