Elevated expression of v-mos is correlated with altered differentiation of carcinoma cells.

Permanent alterations of the epithelial differentiation pattern were investigated after infection of HH-16 cl. 4 adenocarcinoma cells with Moloney murine sarcoma virus (MoMuSV). Transformed cell clones with fibroblastoid morphology were isolated and compared with clones of unchanged epithelioid phenotype. Southern blot analyses showed intact MoMuSV proviral genomes in copy numbers between 4 and 9 in the DNA of the morphologically transformed cell clones as well as in the clones with unaltered morphology. The fibroblastoid cells produced sarcomas after inoculation of newborn rats, whereas MoMuSV-infected cell clones with unaltered epithelioid morphology yielded adenocarcinomas. Immunocytochemical analyses revealed that morphological transformation into the fibroblastoid phenotype was accompanied by loss of cytokeratin expression and appearance of the mesenchymal marker protein vimentin. Proviral DNA was transcribed in the infected cell clones irrespective of their phenotype; however, transcription was significantly higher in cells with fibroblastoid morphology than in epithelioid cells.

[The lithiasis of the gall bladder, a stepchild of the diagnosis of the epigastrium? (author's transl)].

An anamnestic questionnaire project at a hospital for gastroenterological diseases has proved, that in 100 patients cholelithiasis after a cholecystectomy was not diagnosed at first. In more than two thirds of the preliminary diagnoses (69.8 p.c.) stomach was concerned. The therapy required in these cases took 7.9 years on an average. Generally it took 12.6 years of complaints from the first symptom until cholecystectomy was performed. Even when cholelithiasis was diagnosed, the operation was not decided upon until 6.8 years had passed on an average. For the most part the patient himself delayed the operation. According to the patients questioned, chronic pancreatitis was the disease most frequent following cholecystectomy. Reasons for errors in diagnosis of epigastric pain are discussed. The undoubted value of early cholecystectomy is emphasized.

Traditional and emerging molecular markers in neuroblastoma prognosis: the good, the bad and the ugly.

BACKGROUND: Neuroblastomas (NB) are a heterogeneous group of childhood tumours with a wide range of likelihood for tumour progression. As traditional parameters do not ensure completely accurate prognostic grouping, new molecular markers are needed for assessing the individual patient's prognosis more precisely. PATIENTS AND METHODS: 133 NB of all stages were analysed in blind-trial fashion for telomerase activity (TA), expression of surviving, and MYCN status. These data were correlated with other traditional prognostic indicators and disease outcome. RESULTS AND CONCLUSIONS: TA is a powerful independent prognostic marker for all stages and is capable of differentiating between good and poor outcome in putative "favourable" clinical or biological subgroups of NB patients. High surviving expression is associated with an adverse outcome, but is more difficult to interprete than TA because survivin expression needs to be accurately quantified to be of predictive value. We propose an extended progression model for NB including emerging prognostic markers, with emphasis on telomerase activity.
