Response of human endometrium and ovarian carcinoma cell-lines to photodynamic therapy.

The response of the human gynecological carcinoma cell-lines HEC-1-A (endometrial carcinoma) and OvCar-3 (ovarian carcinoma) to photodynamic therapy in vitro was examined. The porphyrin compound Photosan III (Ph III) was used for photosensitization of the cells after incubation times of 24 h (HEC-1-A) and 48 h (HEC-1-A and OvCar-3). The Ph III doses varied from 0-10 micrograms/ml medium. Irradiation was performed with laser light at 630 nm. Irradiation doses up to 20 J/cm2 were applied at an irradiance of 40-100 mW/cm2. Cell vitality of the untreated control groups and of the therapy group was determined 48 h after irradiation, using the trypan blue exclusion test. The experimental results show that treatment of OvCar-3 cells with 10 J/cm2 resulted in a decrease in vitality dependent on photosensitizer dose (0-5 micrograms/ml, 48 h incubation time) but independent of the irradiance (40-100 mW/cm2). Complete cell death was observed after application of irradiation doses in the range of 5-20 J/cm2 combined with drug concentrations of 10-2.5 micrograms/ml, at a fixed incubation time of 48 h. HEC-1-A cells did not survive photodynamic therapy with 10 J/cm2 after incubation with 5 micrograms/ml for 48 h. After a shorter incubation time of 24 h, 10 micrograms/ml Ph III was necessary for the same effect. There was a maximum decrease in cell vitality when measured 48 h after irradiation. This was not improved at 72 h.

[The use of homologous spongiosa chips in the treatment of juvenile bone cysts].

Juvenile bone cysts are characterized by trials of hypothetic interpretations in case of aetiology and general disagreement in relation to indicated therapy. The therapeutic spectrum is ranging from primary conservative treatment till subtotal e.g. total cyst resections. Because in juveniles a quantitative limitation for gathering of autogenous material exists the use of allogenous cancellous bone chips has a definitive place in treatment strategy.

[Photodynamic therapy of bronchial cancer].

Photodynamic therapy (PDT) with porphyrin derivatives (haematoporphyrin and dihaematoporphyrin äther/ester) and laser light of the wavelength 640 nm, is a new procedure for the treatment of carcinomas. This form of therapy is selective, can be applied through the bronchoscope, and is aimed at accomplishing curative elimination of central tumours of the lung. To date, we have successfully applied PDT in four selected patients. Our first patient was treated in April, 1987. The irradiation modalities and the clinical course are described.

[Experiences with the use of cerium nitrate silver sulfadiazine in the local treatment of thermal injuries in childhood].

Ointment containing cerium nitrate and silver sulphadiazine (Flammacerium) was applied to 47 children with second-degree to third-degree thermal injuries, with something between five and 60 per cent of the body surface being affected, as had been reported in a one-year prospective study. This ointment proved to be superior to several alternative approaches with regard to local properties, antimicrobial effectiveness, and applicability to several degree of burns. The process of treatment can be optimised due to unproblematic applicability in open and closed variants under both hospitalised and outpatient conditions. This assessment has been underlined by good to very good therapeutic results.

Molecular cloning of an endogenous cat retroviral element (ECE 1)--a recombinant between RD-114 and FeLV-related sequences. Brief report.

A lambda L 47.1 clone library was constructed from BamHI digests of cellular cat DNA and was screened with cDNA probes specific for the 3'-terminus of the replication competent RD-114 virus. The first analysis of one clone (lambda-ECE 1) shows that it consists at least of a 3'-LTR and a 5' adjacent env gene. The LTR nucleotide sequences of the RD-114 cDNA clones and ECE 1 are identical within the R region and unrelated to FeLV. Otherwise, the region upstream from the 3'-LTR of ECE 1 shows homology to the transmembrane protein encoding gene of FeLV, but not to RD-114. These results imply that ECE 1 is a recombinant between an RD-114-like 3'-LTR and a FeLV-related env portion.

[Benign tumors of the spine].

The most common benign tumors and tumor-like lesions are presented from the aspects of localization in the spinal column and the resulting implications for diagnosis and therapy. For some of these tumors (aneurysmal bone cysts, osteoblastomas, giant cell tumors), von Enneking's suggested staging classification (latent, active, aggressive) is valuable for planning the operation (curettage, tumor removal with healthy tissue margins, wide en bloc resection). If the tumor stage is evaluated preoperatively, problems can be avoided such as overtreatment leading to loss of stability that could have been avoided or the danger of recurrence because the operation was not sufficiently radical. Embolization of the tumor vessels, as an adjuvant measure, not only makes the excision of highly vascular tumors easier, but also can be a means of curbing tumor growth in inoperable cases.

[Tendon sutures with a new monofilament synthetic absorbable suture material (PDS-suture of 6-0 strength). Results of animal experiments].

The absorbable synthetic suture (PDS suture 6-0 with spatula needle) was examined by experiments on the tendon of the M. flexor digitorum pedis communis of the hare. A total of 34 tendon sutures was prepared histologically and evaluated. The new suture material is distinguished by greatly extended absorption time, low foreign body reaction and an improved tensile strength. With this suture, the conditions in which absorbable suture material can be used are increased. This is particularly true of so-called bradytrophic tissue.

[Determination of copper in liver puncture samples in the diagnosis of Wilson's disease].

Wilson's disease, a hereditary disorder of copper metabolism, is characterized by excessive storage of this metal in several organs. Storage of copper in liver tissue is of great importance in diagnosis and therapy. By means of the combination of two methods - low-temperature ashing and flameless atomic absorption - the copper determination can be made even in smallest liver samples (needle-biopsy samples). It is therefore possible to perform a histological examination, as well as a quantitative copper determination on one puncture sample.

[A photometric determination of copper in urine (author's transl)].

A photometric determination for urinary copper excretion in 24-hour urine is described. 6-methylpicolonic acid-thioamide as a 3.3 mmol/l solution in methanol is used as the reagent. It is highly specific and sensitive for copper-I ions, and stable. The method can be used for qualitative screening for Morbus Wilson, and for monitoring the urinary copper excretion fo Morbus Wilson patients under penicillamine therapy.

[Human urinary fluoride excretion at various ages (author's transl)].

3 groups, each consisting of 5 subjects in the following age ranges (A 4 to 6 years, B 25 to 45 years, C 60 to 70 years) were given fluoride in the form of a single dose of 6 mg. Fluoride excretion in the urine was subsequently investigated over a period of 24 hours. In a second experiment a daily dose of 6 mg of F- was given to the 3 groups over a period of 10 days. The urinary excretion was determined in 24-hour urine samples. The results are presented in two figures. 1. A time drift in urinary fluoride excretion in the direction of delayed fluoride metabolism was seen in group C subjects. 2. A periodic increase in the urinary fluoride values was also seen in these elderly subjects, indicative of an altered regulatory mechanism.

[Surgery in Diagnosis and therapy of infant rheumatoid arthritis (author's transl)].

In our hospital, diagnostic surgery is applied in as follows: confirmatory incisions of joints, excision of rheumatic nodulis, tissue excision of skin and muscles (dermatomyositis?), excision of mucosa of the rectum (amyloid?) and open biopsy of the kidney (amyloid?). Therapeutic measures are: total synovectomy of the knee joints or other joints, teno-synovectomy at the hands; treatment of contractions of the hip by myotenotomies and of the knee-joint by elongation of tendons and treatment of knock-knee by incision of the tractus ilio-tibialis and elongation of the tendon of the bicipital muscle.
