[Transplantation of organs and tissues. The idea and practise in the course of time (author's transl)].

The idea of organ transplantation is old and can be traced back to mythology. Even the practice of skin grafting is already over two thousand years old. Nevertheless, today we are perhaps only at the beginning of the phase of broad clinical realization of transplantation surgery. It became possible by accurate scientific working out of the basic immunological problems in this century. At the present time, between the possible organ and tissue transplantations a wide range from a high probability of success (eg cornea) to extensive lack of success (eg lung, small intestine) is to be seen. This is due particularly to the still unsolved immunological problems. Among the organ transplantations in the strict sense only renal transplants can be said to be the fully recognized clinical method of treatment today.

Sugar transport by brush border membrane vesicles isolated from human small intestine.

Uptake of D- and L-glucose, and fructose by purified brush border membrane vesicles isolated from human small intestine was studied using a rapid filtration technique. The uptake of D-glucose by the vesicles was osmotically sensitive and represented transport into an intravesicular space and not binding to the membranes. Transport of both, D- and L-glucose was inhibited by phlorizin. Uptake of D-fructose into the brush border vesicles was not stimulated by sodium. In the presence of a sodium gradient D-glucose was taken up 5 times faster than L-glucose. The amount of D-glucose transported into the vesicles in the presence of a sodium gradient was transiently higher than the amount of D-glucose taken up at equilibrium (over-shoot). D-Glucose transport was stimulated only by a sodium gradient; other monovalent cations had no effect. In the presence of a sodium gradient D-glucose transport was increased by the nearly impermeable anion sulfate as compared with uptake of D-glucose in the presence of a sodium chloride gradient. This indicates an influence of the electrical membrane potential on the sodium coupled non-electrolyte transport.

[Catamneses of YY carriers in prison].

Among the 700 inmates of a big Austrian prison 5 cases of XYY caryotype were detected by means of the Quinacrinfluorescence method and subsequent lymphocyte culture. Keeping in mind that, if at all, only a small amount of criminal behavior may be explained by chromosomal factors three hypotheses about possible links between the XYY abnormality and criminal behavior are discussed in this paper. 1. the connection is a spurious one, as both criminal behavior and the XYY caryotype are more frequent in a working class population without being necessarily linked causally. 2. criminal behavior may be a consequence of the bodily abnormalities connected with the XYY abnormality. 3. the XYY type may contribute to criminal behaviour by acting upon neuro-endocrinological structures.

Phosphate transport into brush-border membrane vesicles isolated from rat small intestine.

Uptake of Pi into brush-border membrane vesicles isolated from rat small intestine was investigated by a rapid filtration technique. The following results were obtained. 1. At pH 7.4 in the presence of a NaCl gradient across the membrane (sodium concentration in the medium higher than sodium concentration in the vesicles), phosphate was taken up by a saturable transport system, which was competitively inhibited by arsenate. Phosphate entered the same osmotically reactive space as D-glucose, which indicates that transport into the vesicles rather than binding to the membranes was determined. 2. The amount of phosphate taken up initially was increased about fourfold by lowering the pH from 7.4 to 6.0.3. When Na+ was replaced by K+, Rb+ or Cs+, the initial rate of uptake decreased at pH 7.4 but was not altered at pH 6.0.4. Experiments with different anions (SCN-,Cl-, SO42-) and with ionophores (valinomycin, monactin) showed that at pH 7.4 phosphate transport in the presence of a Na+ gradient is almost independent of the electrical potential across the vesicle membrane, whereas at pH 6.0 phosphate transport involves the transfer of negative charge. It is concluded that intestinal brush-border membranes contain a Na+/phosphate co-transport system, which catalyses under physiological conditions an electroneutral entry of Pi and Na+ into the intestinal epithelial cell. In contrast with the kidney, probably univalent phosphate and one Na+ ion instead of bivalent phosphate and two Na+ ions are transported together.

Transport of p-aminohippuric acid by plasma membrane vesicles isolated from rat kidney cortex.

Basal-lateral plasma membrane vesicles and brush border membrane vesicles were isolated from rat kidney cortex and the uptake of p-aminohippuric acid (PAH) into these vesicles was studied by Millipore filtration techniques. Both membrane preparations take up PAH into an osmotically reactive intravesicular space. The transport across the brush border membrane seems to involve only simple diffusion whereas in the basal-lateral plasma membrane in addition a specific transport system exists which is inhibited competitively by probenecid. The apparent affinity of this transport system for PAH is 5.4 X 10(-4) M and for probenecid 5.4 X 10(-5) M. PAH uptake into basal-lateral plasma membrane vesicles is influenced by alteration of the membrane potential. Maneuvers which render the intravesicular space more positive--as for example replacement of chloride by sulfate in the presence of a sodium gradient directed into the vesicles and addition of valinomycin in the presence of a potassium gradient directed into the vesicles--stimulate the uptake of PAH. Replacement of a sodium chloride gradient by a sodium thiocyanate gradient reduces the uptake probably by reducing the inside positive membrane potential. In the absence of salt gradients anion replacement and replacement of sodium by potassium does not affect PAH transport by basal-lateral plasma membranes. These results suggest that in isolated basal-lateral membranes transfer of PAH across the membrane is accompanied by a transfer of negative charge. They furthermore provide no evidence for the existence of a sodium-PAH cotransport system in this membrane preparation.

Treatment of paraphilia with luteinizing hormone-releasing hormone agonists.

Up to now there have been no published results of therapy of paraphilia (for example, pedophilia or sadism) and sexual aggressive impulsiveness with luteinizing hormone-releasing hormone (LHRH) agonists in the German-speaking countries. After a short introduction about physiologic features and the present state of investigations in treatment of paraphilia with LHRH agonists we describe 11 patients who were treated with the LHRH agonist Leuprolide Acetate over a period of 12 months. The patients showed no tendency toward sexually aggressive behavior and reported an evident reduction of penile erection, ejaculation, masturbation, sexually deviant impulsiveness, and fantasies. One patient died from suicide. In combination with other treatments, LHRH Agonists seem to be a very promising alternative to cyproterone acetate and its possible carcinogene effects.
