Rapid physical mapping of the Mycoplasma mobile genome by two-dimensional field inversion gel electrophoresis techniques.

A macrorestriction map of the 780 kbp Mycoplasma mobile (ATCC 43663) genome was constructed. Linking fragments were identified on two-dimensional pulsed-field electrophoresis gels. Either complete double restriction digests or partial and complete single digests were separated in the first and second dimension, respectively. 19 restriction sites of four enzymes could be assigned to the map. These rapid methods do not require DNA probes and are applicable to the long-range restriction mapping of all genomes that yield resolvable patterns on two-dimensional gels.

The nasal polyps as a tool for basic research in cystic fibrosis.

Total RNA and mRNA were prepared from cystic fibrosis (CF) and control nasal polyps and nasal epithelial cells. Genomic clones from the chromosomal region of the CF locus were screened by northern blots. A representative cDNA library from nasal polyps was cloned in the vector lambda gt10. For the construction of a physical genomic map around the CF locus single gene markers were isolated from metaphase 1:7q2qter chromosomes by laser micro-dissection and subsequent microcloning. A linkage study with the polymorphic markers met-H, met-D, and pJ3.11 was performed in 53 German CF families with at least 2 children. No significant correlation of any haplotype on the CF chromosomes with the clinical severity of the course of the disease could be observed, which provides evidence that cystic fibrosis is genetically homogeneous.

NotI macrorestriction analysis suggests a clonal relationship of Salmonella enterica, ser. enteritidis phage lysotype 4 strains.

30 different strains of Salmonella enterica ser. enteritidis (S. enteritidis) isolated at the Institute for Medical Microbiology, Hannover, were subjected to a NotI macrorestriction analysis. The results were compared with the respective phage lysis patterns. 25 of 30 isolates belonged to phage lysotype 4; all these strains displayed an identical genomic NotI restriction fragment pattern. In contrast, five isolates of other phage lysotypes revealed more or less prominent differences in their NotI restriction fragment pattern. These results provide genetic evidence for a clonal expansion of a single phage lysotype 4 strain underlying the recent surge in salmonella infections and support phage lysotyping as a reliable epidemiological typing method for S. enteritidis.

Successful treatment of a Trichosporon beigelii septicemia in a granulocytopenic patient with amphotericin B and granulocyte colony-stimulating factor.

Invasive fungal infections have become an increasing problem in severely immunocompromised hosts. We here report a case of septicemia, caused by Trichosporon beigelii, an unusual pathogen of systemic infections. This infection was acquired during a period of severe neutropenia after chemotherapy for relapsed acute myelogenous leukemia following allogeneic bone marrow transplantation. The patient recovered from a life-threatening T. beigelii septicemia due to early intensified treatment with amphotericin B and a rapid neutrophil recovery, enhanced by granulocyte colony-stimulating factor (G-CSF). According to the current literature, amphotericin B is the treatment of choice for systemic T. beigelii infections. In patients with severe granulocytopenia, the rapid recovery of neutrophils remains the most important factor for the outcome of this infection.
