Modes of binding of alpha (1-2) linked manno-oligosaccharides to concanavalin A.

Three-dimensional structures of the complexes of concanavalin A (ConA) with alpha(1-2) linked mannobiose, triose and tetraose have been generated with the X-ray crystal structure data on native ConA using the CCEM (contact criteria and energy minimization) method. All the constituting mannose residues of the oligosaccharide can reach the primary binding site of ConA (where methyl-alpha-D-mannopyranose binds). However, in all the energetically favoured complexes, either the non-reducing end or middle mannose residues of the oligosaccharide occupy the primary binding site. The middle mannose residues have marginally higher preference over the non-reducing end residue. The sugar binding site of ConA is extended and accommodates at least three alpha(1-2) linked mannose residues. Based on the present calculations two mechanisms have been proposed for the binding of alpha(1-2) linked mannotriose and tetraose to ConA.

Lingual dermoid.

Dermoid cysts are congenital lesions derived from ectodermal differentiation of multipotential cells. Only 7% of all dermoid cysts occur in the head and neck region and about a quarter of them are seen in the floor of mouth. Dermoids confined to the tongue are rare entities. Only 13 such cases, mostly in infants, have been reported in literature. One such case of an infant with intralingual dermoid and feeding difficulty, relieved following cyst excision, is reported.

In vitro segregation of marker genes in anther cultures of rice.

Segregation of genes controlling expression of anthocyanin pigmentation in rice (Oryza sativa L. subsp. indica) leaf blade and leaf sheath was examined in the microspore-derived plants. The segregation pattern of marker genes was found to fit closely the expected gametic segregation ratios among microspore-derived green as well as albino plants. Microspore-derived in vitro regenerated plants expressed genetic traits similar to seedlings. The results indicate that the germ cell culture technique can be of significance while monitoring gene action, i.e. anthocyanin synthesis at monoploid phase of plant development.

Antibody mediated targeting of radioisotopes, drugs and toxins in diagnosis and treatment.

The recent resurgence of interest in site specific delivery of radioisotopes, chemotherapeutic drugs and toxins for the diagnosis and treatment of cancer, and for the selective manipulation of the immune system, can be directly related to the need for improved diagnosis and the fact that for many cancers, for example lung, colon and gastric, the conventional treatments of surgery, radiotherapy and chemotherapy have reached a plateau in terms of the number of patients cured. To date, because of their specificity, the major emphasis has been on the use of antibodies as carriers and extensive in vitro, in vivo preclinical and clinical evaluation is underway. The aim of this article is to review recent progress, highlight avenues being explored to overcome limitations and to indicate new approaches that are evolving in antibody mediated targeting.

Use of nesiritide (human B-type natriuretic peptide) in infants following cardiac surgery.

We report the use of nesiritide in two infants following cardiac surgery. Both infants had increased intracardiac filling pressures postoperatively, despite traditional afterload reduction and diuretics. Both infants demonstrated clinical improvement with nesiritide therapy. There is currently limited data available describing the use of nesiritide in infants.

Perceptions on effects of environmental pollutants in Hyderabad City.

Five hundred twenty adults were interviewed on the perceptions of effects of the environmental pollutants in Hyderabad city of Andhra Pradesh. The information was gathered on polluted water, polluted air, solid wastes, food contamination and readiness to participate in corrective actions. Both initial response and response after probing were recorded. Analysis was done basing on the level of literacy. The correct initial responses on effects of pollutants ranged from 28.3 to 35.4% while responses after probing ranged from 79.2 to 88.1%. 45.8% were ready to participate manually in the corrective actions while 24.6% were eager to contribute financially in remedial actions. The study reveals the need for the initiation of aggressive environmental education.

Production of hybrids secreting bispecific antibodies recognising CEA and doxorubicin.

A monoclonal anti-CEA secreting hybridoma (11-285-14) was made hypoxanthine, aminopterin and thymidine (HAT) sensitive by back selecting it in increasing concentrations of 8-azaguanine. Eight 8-azaguanine resistant fusion partners were selected based on growth characteristics and continued anti-CEA production. Since doxorubicin (Dox) is a hapten, it was conjugated to the carrier proteins keyhole limpet hemocyanin (KLH) or bovine serum albumin (BSA) using 1-ethyl-3- (dimethyl-aminopropyl) carbodiimide. Dox-KLH and Dox-BSA conjugates were used to immunize mice and spleen cells from these mice were used for fusions with the HAT sensitive anti-CEA partner using standard hybridoma procedures. Enzyme linked immunosorbent assays (ELISAs) were developed to test the hybrids obtained for anti-CEA, anti-DOX, anti-BSA and bispecific monoclonal antibody (BsMab) activity. Sixteen fusions with spleen cells from DOX-KLH immunized mice yielded 621 hybrids of which 47 showed low level BsMab activity by ELISA. Eight fusions with spleen cells from DOX-BSA immunized mice yielded 297 hybrids. Fifty of these hybrids showing dual reactivity have been cloned and subcloned to yield 7 subclones with stable BsMab activity for CEA and doxorubicin.
