Trisomy of 1q in preleukaemia with progression to acute leukaemia.

Marrow cells from an untreated man with preleukaemia were found to have trisomy for the long arm of chromosome 1 which was translocated to the end of the long arm of number 11, i.e., 46,XY, -11,+t (1:11) (q11 or q12; q25). The same abnormality was found in metaphases from 8 individual granulocytic colonies. With development of acute myelomonocytic leukaemia, in addition to the basic chromosome abnormality, trisomy of chromosome 8 and an extra chromosome number 19 with partial deletion were found. Fibroblasts grown from marrow biopsy material showed a normal diploid complement, 46,XY.

Isochromosome 17 in a case of chronic erythroleukaemia.

In a patient with chronic erythroleukaemia in isochromosome 17q was identified in all bone marrow metaphases analyzed. In addition, these cells were characterized by hypodiploidy and the presence of 3 different marker chromosomes. Approximately 50% of dividing cells in the marrow were bizarre-looking polyploids. The identification by Giemsa banding of i(17q) in this case of erythroleukaemia supports the concept that there are common cytogenetic characteristics among malignant myeloproliferative disorders.

Agar culture and chromosome analysis of eosinophilic leukaemia.

The blood and bone marrow of a 75-year-old woman with eosinophilia were studied in an attempt to characterize further the entity eosinophilic leukaemia (EL). Haemopoietic cell culture in agar gave patterns of colony size and number and cell morphology indistinguishable from those seen in classical chronic granulocytic leukaemia (CGL). Analysis of myeloid cells showed an extra C group chromosome that proved on Giemsa banding to be number 10. These results suggest that some cases of EL represent a chronic myeloproliferative disease that resembles classical CGL but can be distinguished from it by chromosomal analysis.
