[The effect of the one-time and long-term administration of preparations acting on biogenic amine metabolism on the blood level of pressor peptide hormones].

Blood contents of pressor peptide hormones vasopressin and angiotonin II were studied in patients with neuro-endocrine syndrome before and after single intake and prolonged treatment with anti-serotonin drug peritol and cholinergic agent parlodel which affect biogenic amine metabolism and, consequently, influence blood pressure. Single doses of the drugs were established to cause different blood dynamics of vasopressin and angiotonin II which classified as marked and paradoxic reactions on peritol and parlodel used separately and associatively. Fall of blood vasopressin content induced by single dose of parlodel was accompanied by blood pressure decrease. Tree-week treatment with peritol and parlodel exerted hypotensive effect and significantly reduced vasopressin blood content.

[Effect of pathogenetic treatment on vasopressin secretion in patients with hypothalamic syndrome].

Blood vasopressin concentration, hypothalamic response to stress resultant from insulin hypoglycemia and to acute furosemide load were measured in 72 patients with neuro-endocrine-metabolic form of hypothalamic syndrome. Pathogenetic treatment was decided upon by sensitivity to dopaminergic drug parlodel and antiserotonin drug peritol. According to the sensitivity tests the patients received either parlodel (5 mg/day) or peritol (12 mg/day) for 3-6 months. There were also patients on symptomatic treatment aimed at reduction of body weight. Peritol treatment promoted a decline in basal blood level of vasopressin and better response to insulin hypoglycemia and furosemide test. Parlodel treatment normalized vasopressin blood concentration and hypothalamic response to stimulators. Routine symptomatic therapy did not induce differences in vasopressin level compared to active stage of the disease.

[Radioimmunological analysis in assessing the function of the hypothalamo-neurohypophyseal system in patients with hypothalamic syndromes].

Assessment of vasopressin by radioimmunoassay has shown an increase in its blood concentration and a disturbed reaction of the vasopressinergic structures of the hypothalamus to metoclopramide, furosemide, insulin hypoglycemia, and exercise. Functional tests with the dopaminergic drug bromocriptine and antiserotoninergic drug cyproheptadine help to make an individual choice of the most effective drug for therapy of the hypothalamic syndrome of neuroendocrine-metabolic type. The patients can be divided into sensitive to either the first or the second drug of both which is important for adequate pathogenetic therapy.

[Changes in the secretion of the hormones of the hypothalamo-hypophyseo-adrenal system in Itsenko-Cushing disease under the influence of insulin-induced hypoglycemia and thyroliberin].

The concentration of corticotropin and cortisol in the blood was determined 120 patients with Itsenko-Cushing radioimmunologically after insulin hypoglycemia, after administration of insulin and thyroliberin. It was established that healthy persons and patients with Itsenko-Cushing disease showed no differences in the effect of insulin and thyroliberin as compared with insulin hypoglycemia on the state of the pituitary-adrenal system.

[The renin-angiotensin-aldosterone system in hypertension of hypothalamic origin].

The results of the study of angiotensin-II and aldosterone++ concentrations, renin activity in blood plasma, the response of adrenal cortex glomeruli and juxtaglomerular system to insulin hypoglycemia, metoclopramide++, furosemide and exercise were considered for 119 patients with neuroendocrine form of hypothalamus syndrome. It was found that hypothalamic regulation of renin-angiotensin-aldosterone++ system underwent changes. Hypersecretion+ of aldosterone++ is an important factor in pathogenesis of arterial hypertension in hypothalamic syndrome.

[Prostaglandins in Itsenko-Cushing syndrome].

The results of the study of depressor (PGE+A) and pressor (PGF2 alpha) prostaglandins in the blood plasma and of PGE2 and PGF2 alpha excretion in the urine of 52 patients with an acute stage of Itsenko-Cushing disease are presented. It has been established that the different components of the PG system have dissimilar changes: the absence of differences in the content of depressor and significant increase in pressor PG, which may be the cause of hypertension in Itsenko-Cushing disease. Stress due to insulin hypoglycemia and furosemide load brought about disturbances in the reaction of the PG system, which were more pronounced in the system pressor component. Disturbances of the PG system which were more pronounced in the presence of stable hypertension were established in all patients.

[Effect of beta-endorphin on catecholamine levels in the rat hypothalamus and cerebral cortex].

The present paper deals with the effect of beta-endorphin on catecholamine content in the hypothalamus and cerebral cortex of male rats. beta-endorphin was found to decrease catecholamine content in the rat brain, with the degree of reduction depending on the brain topography and the time following the peptide administration. 5 min later no changes in catecholamine content were observed either in the hypothalamus or in the cerebral cortex. 20 min later beta-endorphin induced a statistically significant fall of catecholamine concentration in the hypothalamus. A tendency towards its decrease was also observed in the cerebral cortex. 60 min later beta-endorphin produced an insignificant decrease in catecholamine level in both brain areas under study. It may be therefore suggested that beta-endorphin-induced decrease of catecholamine content in the hypothalamus and cerebral cortex represents one of the mechanisms underlying beta-endorphin stimulating action on a number of trophic functions of the hypophysis.

Effect of the antiserotonin drug, cyproheptadine, on plasma beta-endorphin and beta-lipotropin in patients with Itsenko-Cushing's disease.

In the present paper evidences are provided for the changes occurring in blood opioid neuropeptides in patients with Itsenko-Cushing's disease), who were treated with an antiserotonin drug, cyproheptadine (peritol). An increase in the plasma contents of beta-endorphin and beta-lipotrophin in the above patients has been found to be one of the factors conditioning an enhanced activity of the hypothalamic-pituitary-adrenal system. Treatment of the patients suffering from Itsenko-Cushing's disease with cyproheptadine (peritol) resulted in a decrease of the plasma beta-endorphin and beta-lipotropin, as well as a display in 60% of cases of clinical and laboratory remission. Absence of the clinical remission in some patients treated with peritol indicated a need for intervention in the metabolism of other monoamines involved in the modulation of the action of opioid neuropeptides on the activity of the hypothalamic-pituitary system.
