[Serum factors in rabbits with acute pancreatitis as stimulators of regeneration of pancreatic islet B-cells in experimental diabetes].

The experiment reveals the stimulating effect of pancreatic rabbits' serum on the reparative regeneration of pancreatic islets' B-cells of rabbits with alloxan diabetes. Serum, got 12 hours after mechanical pancreatic gland trauma proves to be most effective. The maximum rise of mitotic activity is in 24 hours after 3 daily injections of serum. In the experiment with the decrease of the level of proliferation of B-cells compensation of insulinary function by acinoinsulary transformation prevails.

[Species specificity of serum factors in rabbits with acute pancreatitis in stimulated regeneration of pancreatic islet B-cells in experimental diabetes].

The experiment has shown that the serum of rabbits with acute pancreatitis produces proliferative effect on pancreatic islets' B-cells in rats with the acute form of experimental diabetes. The stimulation of miotic activity of B-cells, processes of acino-insular transformation, compensation of the insular function in rats with alloxan diabetes proved to be lower than in rabbits under the same experimental conditions. This testities to relative species specificity of factors in the serum of rabbits with pancreatic gland trauma.

[Morphofunctional characteristics of the cat spiral ganglion in neurosensory hypoacusis].

Morphofunctional disorders in spiral ganglion in animals of different groups of age with neurosensory hardness of hearing due to ototoxic antibiotics have been shown. Antibiotic intoxication in kittens in early postnatal period arouses essential deviation from normal process of development of neurocytes in spiral ganglion. In the first rate low level of their metabolic activity must be shown. Delay of differentiation of neurocytes makes essential influence on the development of spiral ganglion as the unit of transmission of audio-information. The main feature of morphofunctional disorders in neurocytes of ganglion of grown-up animals is inhibition of protein synthesis mechanism activity.

[Fast transport of proteins in the axons of spinal cord motor neurons in rats adapted and unadapted to physical loading].

The prolonged nonintensive swimming without load (12+ +2 h) provoked a decrease of the rate of fast axonal transport in the motor fibres of the sciatic nerve by 18% and of the overall amount of transported proteins 2-fold as compared with control. The relatively short-term but more intensive activity (swimming with the load 1/11 of body weight during 60+ +10 min.) results in an increase of the rate by 10% and the overall amount of fast transported proteins 2-fold. Daily swimming (3 h. without load and 10-15 min. with load) during 20 days returns the above parametres to the controls. The compensative-adaptive changes of fast anterograde transport are the fact that may be involved in trophic communication between neuron and target cell.

Adaptation, Physiological↗

[Proliferation and differentiation of nerve cells of the human NN. supraopticus et paraventricularis transplanted to the brain of adult rats].

Human fetal hypothalamic neurosecretory cells (NSC) were studied after transplantation into the third ventricle of the adult rat brain. 3H-thymidine uptake data have shown that the time of origin and proliferation of NSC of grafted tissue and of supraoptic and paraventricular nuclei in normal embryogenesis was similar. Specific staining revealed that the start of neurosecretory function in grafted tissue corresponded to the date when NSC began functioning in normal ontogenesis.

[Immunogenesis and axoplasmic transport in Wistar rats].

Immunization of Wistar rats with thymus dependent antigens (sheep red blood cells-SRBC) is accompanied by a reliable increase in the synthesis of RNA and proteins in thalamic cerebral cortex and spinal marrow (48 hrs after antigen injection) and also in an increase in the intensity of rapid axoplasmic transport (RAT) along motor fibers of sciatic nerve (5,48,72 hrs following the beginning of immunization). There was a consecutive augmentation in AFC number in mesenteric and partly in inguinal lymph nodes (96 hrs after SRBC injection). Thus, time dependence between immunogenesis and axoplasmic transport in experimental animals (Wistar rats) was determined for the first time. It identifies another, previously unstudied, channel in interactions of immune and nervous systems.

[The dynamics of the chromatin transcription activity in cerebellar Purkinje cells after systemic circulatory arrest].

Transcriptional activity of the dark and light Purkinje cells' nuclei in cerebellar cortex was studied in rats 1h, 24h, 96h and 7 days post a systemic blood circulation arrest. The study was performed with the help of the histoautoradiographic method of Moore. Some substantial changes in template activity of the Purkinje cells' chromatin were observed in the postresuscitation period. Those began to occur immediately after resuscitation and were of phasic nature. The shifts in transcriptional activity of nucleolar and extranucleolar chromatin appeared to be analogous and of similar dynamics. The changes were more significant in the light neurons than in the dark ones. It has been supposed that the changes revealed might be one of the causes of development of neurologic disorders induced by severe hypoxia.

[Changes in the structural and functional states of chromatin of the neurons of the cerebral hemisphere cortex in the early post-resuscitation period in rats].

Structuro-functional state of chromatin of the neurons' nuclei of the third, fourth and fifth layers of the sensorimotor cortex was investigated by Moore's method and with the help of ammonia silver colouring an hour and 24 hr post resuscitation in rats who had endured a systemic arrest of blood circulation. It was determined, that the level of labelling of nucleoli of light neurons increased in the early postresuscitation period, while that of dark neurons decreased as compared to check samples. The level of labelling of extranucleolar chromatin increased in both light and dark neurons in the cortex layers under investigation during the 1st hour post resuscitation. The transcriptional activity of extranucleolar chromatin of neurons of both types remained increased in the third and fourth cortex layers 24 hr post resuscitation, while in the fifth one it tended to normalize during the same period. Essential restructuring of the neurons's chromatin was determined with the help of ammonia silver in all cortex layers, which was indicative of a possible change in the spectrum of the synthesized proteins.

[Transcription of nucleoplasmatic and nucleolar sites of DNA sensitive to the protease inhibitor antipain].

Age-related changes in the transcriptional activities of Purkinje cells chromatin and sympathetic cranial cervical ganglion under the influence of morphine and some natural and synthetic peptide compounds were found. It was shown that at different stages of the ontogenesis, morphine was capable of increasing the template activity of nucleolar and/or extranucleolar chromatin, and that this process was inhibited by antipain. The findings suggest the presence of DNA sites in the eucaryotic genome, that are induced by the protease mechanism and are likely to be similar to the SOS-genes of procaryotes. The SOS-genes of procaryotes are activated under unfavourable conditions and promote the survival of the microbial population, while in eucaryotes these genes are believed to provide the formation of definite cell populations whose composition is the most adequate for the living conditions of an adult organism.

[Cosinor analysis of the circadian dynamics of transcription in populations of neurocytes from a number of sections of the rat nervous system].

Diurnal fluctuations of endogenous RNA polymerases activity were studied in the cell nucleus of the cerebral stem and spinal nervous system populations (neurocytes of the hypothalamic suprachiasmatic nuclei, superior cervical ganglia, spinal ganglia L5, motoneurones of the spinal cord and Purkinje cells) were revealed. The acrophases of the visual cortex neurones were observed just before light reception.

[H3-dopamine and H3-quinuclidinylbenzilate binding by autonomic ganglia tissue in rats of various age].

Binding of tritiated ligands of muscarinic and dopamine receptors was analysed in rats 1, 7, 14, 28, 60 days and 24-30 months old. The following ganglia were studied: the nodose ganglion, the lumbar ganglia of sympathetic chain, the main pelvic ganglion in male rats and the paracervical ganglion in female rats. The same level was found for binding of each of ligands for all investigated ganglia. Parameters of postnatal dynamics of development M-cholino- and dopamine reception systems prove to be quite similar, but not identical. Both of the systems reach matured level during first 2 postnatal weeks. In comparison with quinuclidinylbenzilate binding failure of dopamine binding in aged rats is to be more pronounced.

[Changes in the axonal protein transport under conditions of elevated functional activity of the neurons].

The prolonged nonintensive physical activity by swimming without load (12 +/- 2 h.) has no effect on the overall amount of fast and slow transported proteins of transport velocity in rat central and peripheral sensory fibres of the sciatic nerve. However, the rate of fast axonal transport in the motor fibres decreases by 18% and the amount of proteins by a factor of 2 as compared with control. The rate of slow axonal transport does not change, but the mean level of transported labeled proteins decreases by 1.9 times. The relatively short-term but more intensive activity (swimming with the load during 60 +/- 10 min.) provokes an increase of the rate by 10% and the overall amount of fast transported proteins by 2 times. The rest of the animals during 6 h. returns the above parameters to control values. A suggestion is made that the rate and the amount of transported proteins depend on the variations in functional state of the neurons and their axons.

[The development and functioning of embryonic pancreatic islet cells in the anterior chamber of rats].

In the present study the morphogenesis and differentiation processes in embryonic pancreatic gland implants into Wistar line rat anterior eye chamber have been investigated. The conditions therein were found to be favourable for the endocrine tissue functioning; a number of morphogenetic changes resulting in the formation of islet structure acting as a morphophysiological unit were noted as well. Endocrine cells possess some selective properties as compared to the exocrine tissue. Alloxan diabetic animals demonstrated the most optimum conditions for the endocrine cells development and functioning.

[Cytochemical study of the neurocytes of the rat locus coeruleus after guanethidine sympathectomy].

The histone localization, template activity of chromatin and epinephrine fluorescence in L. Coeruleus (LC) neurons of normal a. guanethidine sympathectomized (GS) rats (15 mg/kg during 3, 8 or 14 days after birth) were studied. The absence of chromatin rearrangements connected with genome reprogramming under sympathectomy was documented by the fact that maximal doses of guanethidine gives no changes in ammoniacal silver staining pattern. However GS caused the decrease in nuclear and extranuclear template activity and in epinephrine fluorescence proportionally with GS. It is obvious that LC cells tend to form discrete classes according to their transcription levels.
