Mechanisms of genetic resistance to Friend virus leukemia. III. Susceptibility of mitogen-responsive lymphocytes mediated by T cells.

Friend leukemia virus (FV) suppressed the proliferative responses of spleen, lymph node, marrow, and thymus cell populations to various T- and B-cell mitogens. Cells taken from mice, e.g. BALB/c genetically susceptible to leukemogenesis in vivo were much more susceptible to suppression of mitogenesis in vitro than similar cells from genetically resistant mice, e.g., C57BL/6. Nylon wool-purified splenic T cells from BALB/c and C3H mice lost susceptibility to FV-induced suppression of mitogenesis but became suppressible by addition of 10% unfiltered spleen cell. Thus, FV mediates in vitro suppression of lymphocyte proliferation indirectly by "activating" a suppressor cell. The suppressor cell adhered to nylon wool but not to glass wool or rayon wool columns. Pretreatment of spleen cells with carbonyl iron and a magnet did not abrogate the suppressor cell function. Suppressor cells were not eliminated by treatment with rabbit antimouse immunoglobulin (7S) and complement (C). However, high concentrations of anti-Thy-1 plus C destroyed suppressor cells of the spleen; thymic suppressor cells were much more susceptible to anti-Thy-1 serum. Nude athymic mice were devoid of suppressor cells and their B-cell proliferation was relatively resistant to FV-induced suppression in vitro. The suppressor cells in the thymus (but not in the spleen) were eliminated by treatment of mice with cortisol. Thus, FV appears to mediate its suppressive effect on mitogen-responsive lymphocytes by affecting "T-suppressor cells." Spleen cells from C57BL/6 mice treated with 89Sr to destroy marrow-dependent (M) cells were much more suppressible by FV in virto than normal C57BL/6 spleen cells. However, nylon-filtered spleen cells of 89Sr-treated C57BL/6 mice were resistant to FV-induced suppression in vitro, indicating that the susceptibility of spleen cells from 89Sr-treated B6 mice is also mediated by suppressor cells. Normal B6 splenic T cells were rendered susceptible to FV-induced suppression of mitogenesis by addition of 10% spleen cells from 89Sr-treated B6 mice. Thus, M cells appear to regulate the numbers and/or functions of T-suppressor cells which in turn mediate the immunosuppressive effects of FV in vitro. Neither mitogen-responsive lymphocytes nor T-suppressor cells are genetically resistant or susceptible to FV. The genetic resistance to FV is apparently a function of M cells, both in vitro as well as in vivo.

Immune response of guinea-pigs to chemically abbreviated prepatent Metastrongylus apri infection.

The effect of chemical abbreviation of the primary infection dose (PID) of 160 infective larvae of Metastrongylus apri on the immune status of the guinea-pig host was studied. The criteria used for assessing the status of immunity consisted of clinical manifestations following administration of a challenge infection dose (CID) of 800 infective larvae of M. apri, the rate of worm recovery 15 days post-CID and the rate of mortality following administration of CID. Among the guinea-pigs of the main experimental group, where 15-day-old PID was abbreviated by two parenteral doses of levamisole, a strong immunity to CID given 35 days post-PID was built-up. Against this, all the guinea-pigs of a control group, which did not receive PID, died between 16 and 22 days post-CID. The increase in serum gamma-globulin level of the guinea-pigs, where the PID was abbreviated chemically, suggested that the rise of this globulin fraction in the serum could be in some way related to the resistant state of guinea-pigs.

Combined lactose-D-xylose tolerance test in infancy.

A modified combined oral lactose-D-xylose tolerance test done on ten normal infants showed peak levels of blood glucose and D-xylose levels comparable to the peaks obtained when these tests were done separately in the same subjects. The combined test is recommended to evaluate digestive absorptive function in preference to two separate tolerance tests.

Histochemical localization of glucose-6-phosphatase and 5-nucleotidase in the digestive system of Ophiocephalus Channa punctatus.

The histochemical localization of G6Pase and 5-Nase in the digestive system of Ophiocephalus (Channa) punctatus was studied. The highest activities of these enzymes were found in the liver. Appreciable activity was also found in the anterior intestine (duodenum) and pyloric caeca. The activity faded toward the middle and posterior intestine and rectum. In the stomach the activity was moderate. The activity of 5-Nase was weaker than that of G6Pase. In the stomach the enzymes were localized in the mucosa and gastric glands. The absorptive columnar epithelial cells were the major sites of localization in the intestine. The goblet cells were negative. The G6Pase activity was associated with the cytoplasm, while the 5-Nase activity was found in the cell membranes and the nuclei.

Antifibrin action of phenformin.

Effects of phenformin on blood sugar, serum triglyceride, thrombin time, euglobulin clot lysis time and cardiovascular complications were studied in maturity onset diabetes and in atherosclerotic patients with or without diabetes, for a period of 14-18 months. Phenformin has shown the characteristic properties of an antifibrinopathic agent in that it prolongs thrombin time and enhances fibrinolysis. The hypoglycaemic effect of phenformin was found to be directly related to its antifibrinopathic action. Plasma lipids fell in all cases. Absence of fresh cardiovascular complications and improvement in anginal symptoms were observed. The metabolic, haematological and clinical benefits of phenformin and its limitations in maturity onset diabetes and atherosclerosis may be explained by the effects of the drug upon the thrombin-fibrinogen reaction. These results lend support to the hypothesis of a primary fibrinopathic pathogenesis in maturity onset diabetes mellitus and atherosclerosis.

Nasitrema attenuata (Trematoda) infection of Tursiops truncatus and its potentialities as an aetiological agent of chronic pulmonary lesions.

Two cases of Nasitrema sp (Trematoda) infection of the bottlenosed dolphin, Tursiops truncatus, are reported and N. attenuata is briefly redescribed. The adults of the trematode occurring in the nasal sinuses and posterior nasal passage of the dolphins are considered as practically harmless for the host but thier eggs, aspirated deep into the bronchial tree, may initiate a foreign-body of inflammatory reaction in the lungs and continuous aspiration of such eggs may provoke a chronic pneumonia condition.

Carbohydrate intolerance in infants with acute diarrhoea and its complications.

Two hundred and seventy-one infants with acute diarrhoea were studied for the presence of carbohydrate malabsorption and 110 infants (40.6%) were found to have carbohydrate intolerance. Malnutrition and severe diarrhoea were found to increase the predisposition to carbohydrate intolerance. The incidence of major complications, protracted diarrhoea and mortality were significantly higher in the carbohydrate intolerant infants as compared to those with carbohydrate tolerance.
