Complement-fixing intercellular antibodies in a case of cicatricial pemphigoid.

A patient with ocular and oral lesions, as well as cicatricial pemphigoid, had intercellular antibodies to a titer of 80 by indirect immunofluorescence (IF) microscopy staining tests with her serum. These were identified as pemphigus-like antibodies, since direct IF microscopy indicated no intercellular IgG deposits. Complement IF microscopy tests of the serum gave a closely comparable titer (40) of intercellular antibodies. Since serum samples of patients with true pemphigus either fail to fix complement or have much lower complement fixation titers, the complement IF microscopy tests of patients' serum samples may be of value in differentiating pemphigus-like antibodies from pemphigus antibodies.

Microscopic pathology of liver of capuchin monkey (Cebus albifrons) infected with Athesmia foxi (dicrocoelidae : trematoda) : a pictorial illustration.

Histopathological changes in the livers of two white fronted capuchin monkeys, Cebus albifrons, associated with Athesmia foxi (Dicrocoelidae : Trematoda) infection are illustrated through photomicrographic plates. The lesions are confined in and around the hepatic biliary system. The bile ducts are greatly distended and thickened because of fibroblastic activity and collagen deposit in its walls. In one of the livers desquamative, necrotic and reorganizational changes are found to be well marked. At places the bile duct lumen contains infiltrating inflammatory cells and shed pieces of biliary epithelia mixed in a scanty necrotic tissue while elsewhere the necrotic changes are more pronounced and the bile duct contents are presented only by homogeneous amorphous debris. The overall picture is that of chronic cholangitis coupled with cholangiectasis.

Understanding of diseases and treatment-seeking pattern of childhood illnesses in rural Haryana, India.

Two hundred and sixty respondents were interviewed to know the causes of ten common childhood illnesses, in one community development block in rural Haryana. Chickenpox, measles and marasmus were attributed to supernatural causes by 27%, 28%, and 50% respondents respectively. Furthermore older age groups, illiterates, schedule caste and people living more than 5 km from a Government health post believe more in supernatural causation of disease. By and large, Registered Indigenous Medicine Practitioners (RIMP's) and traditional birth attendants were utilised for curative medicine and obstetric care.

Kwashiorkor in the United States.

Kwashiorkor, with typical edema and skin rash is occasionally seen in affluent countries as a result of severe protein restriction. Treatment is with a lactose-free formula after rehydration and sometimes after a period of parenteral alimentation. Complications of therapy included: diarrhea, congestive heart failure, infections, a bleeding tendency, and seizures. Two of the 14 children in this series died.

Hybrid resistance to EL-4 lymphoma cells. I. Characterization of natural killer cells that lyse EL-4 cells and their distinction from marrow-dependent natural killer cells.

Natural killer (NK) cells from nonimmunized mice capable of lysing EL-4 (C57BL/6 strain H-2b) tissue culture-adapted lymphoma cells have been analyzed and compared with NK cells which lyse YAC-1 (A-strain, H-2a) lymphoma cells. A correlation was seen in the ability of inbred and B6D2F1 mice to reject C57BL/6 (B6) bone-marrow grafts and the ability of their spleen cells to lyse EL-4 cells in vitro. This suggests that hybrid or hemopoietic histocompatibility antigens, (Hh-1b), relevant in the rejection of B6 stem cells may also be the relevant target structures for the anti-EL-4 NK cells. Certain features of these NK cells are similar to the NK cells reactive against YAC-1 cells. Both types of NK cells are present in athymic nude mice, are not affected by treatment with anti-immunoglobulin plus complement, and are not depleted by techniques that remove macrophages. NK activity against both targets is stimulated 3 d after injection of Corynebacterium parvum, and 24 h after challenge with polyinosinic:polycytidylic acid. Hydrocortisone acetate and cyclophosphamide lead to reduction of NK activity within 2-3 d after administration. However, the anti-YAC and anti-EL-4 NK reactivities differed in several important respects. Treatment of mice with 89Sr, the bone-seeking isotope, to deplete marrow-dependent cells, depleted the anti-YAC-1 but not anti-EL-4 cell functions. Anti-EL-4 NK cells were unaffected by silica particles in vivo or in vitro; the NK cells reactive to EL-4 cells matured functionally much earlier in life (5 d of age) and the function did not decline with age. Irradiated mice reconstituted with syngeneic marrow or spleen cells developed functional NK cells against EL-4 targets before they developed anti-YAC-1 NK cells in their spleen. Thus anti-EL-4 NK cells that express hybrid resistance in vitro appear to differ from anti-YAC-1 NK cells and do not require an intact marrow microenvironment for functional differentiation. Despite differences in the NK-cell types involved in the lysis of YAC-1 and EL-4 cells, these two tumor cells share certain common determinants. This was ascertained both by cold competition and by utilization of YAC-1 and EL-4 cell monolayers as immunoadsorbents. We conclude that Hh-1b is the common antigen present in EL-4 and YAC-1 cells, because B6D2F1 anti-B6 (anti-Hh-1b) cytotoxic T lymphocytes lysed both the tumor cells. Our data suggest that Hh-1b antigen is recognized by both types of NK cells, but that additional determinants must be present on YAC-1 cells. Two models of NK cell lysis compatible with the data are presented.

Antigenic components of Taenia saginata and their relevance to the diagnosis of bovine cysticercosis by immunoelectrophoresis.

22 antigenic components of Taenia saginata proglottides were detected by immunoelectrophoretic (IEP) study. Cross-absorption of the hyperimmune serum, raised in rabbits against T. saginata, with heterologous and host antigens showed that two thirds of these components were not specific for the cestode. Among the calves experimentally infected with T. saginata eggs, 3 precipitation patterns in IEP were identified depending on the evolutionary stage of the infection. The hydrosoluble extract of T. saginata was used as antigen. A short precipitating arc present near the antigen well and appearing 3 to 4 weeks post-infection of the calves and a long precipitating arc extending towards the anodic end of the slide and appearing in later stages of infection were found of high diagnostic value. No false-positive reaction or cross-reaction were found. Among the calves harbouring less than 50 cysticerci, false-negative reactions were seen.

Structural requirements for progestational activity. Synthesis and properties of rac-8alpha,9beta,10alpha,14beta-progesterone.

rac-8alpha,9beta,10alpha,14beta-Progesterone, 1, has been synthesized and subjected to X-ray crystallographic analysis which established that the ring conformations are A, 1beta-sofa; B, chair; C, chair; and D, intermediate between an envelope and a half-chair. This compound is 10% as active as progesterone in the Clauberg assay and has an affinity for the uterine cytosol (rabbit) receptor for progesterone 2% as great as that of progesterone.

Aminoglycoside antibiotics. 2. N,N-Dialkylkanamycins.

N6',N3''-Dialkyl derivatives of kanamycins A and B were prepared regiospecifically from the parent antibiotics. Although the dimethyl and diethyl derivatives of kanamycin A were inactive in standard antibacterial assays, the dimethyl derivative of kanamycin B showed moderate activity, especially against various strains of Pseudomonas aeruginosa. A method for the selective dimethylation of the 3''-amino group of kanamycin A also was developed.
