Detection of human papillomavirus gene sequences in cell lines derived from laryngeal tumors.

The role of Human Papillomaviruses (HPV) in laryngeal carcinomas has been studied with conflicting results. To evaluate the etiologic relationship between HPV infection and epithelial malignancy of the larynx we studied five laryngeal carcinoma cell lines obtained from patients undergoing surgery for laryngeal tumors. The paraffin embedded biopsy samples of the original tumor and different passages of the new established cell lines were investigated by PCR with consensus primers specific for HPV DNA. The findings indicate that HPV infection is associated with some larynx carcinomas. The positive association has been enhanced when a method of enrichment of epithelial cells from fresh tumor samples was used. All tumor cells enriched smears were positive for HPV DNA not only by PCR but also by in situ hybridization (ISH). Investigated by PCR, different passages of larynx tumor cell lines maintained expression of HPV DNA. At subsequent passages ISH gives constantly no signals suggesting a minimal amount of viral harbored sequences. In one cell line propagated more than 60 population doublings, the chromosomal frequency distribution shifted from modal number 46 at the 5(th) passage to 63 at the 60(th) passage. The mechanisms by which persistent HPV infection maintains continuous cell proliferation were discussed.

An outbreak of aseptic meningitis due to ECHO 30 virus in Romania during the 1999 summer.

Between April-September 1999 an unusually high number of acute aseptic meningitis was reported in six North Eastern counties of Romania. At the end of 1999 summer the epidemic area extended in South Romania including the Capital City Bucharest. The total number of cases surpassed 5500. The majority of confirmed cases were young children under 15 years of age. More than 1800 cases were registered in the same interval in the neighboring Republic of Moldavia. We report the isolation of ECHO 30 virus from 12 CSF samples of 52 tested on RD and human diploid cells. A small case control study identified high seroneutralization titers confirming a recent ECHO 30 infection in 74% of patient's samples and in 54% of samples from control children tested. The time spent outdoors and poor hygiene practices were prominent risk factors for transmission.

[Epidemiological and virological research on influenza and viral respiratory infections in the southeastern region of Romania in the cold season of 1991-1992].

Investigations pointed out the high level of respiratory diseases and pneumonias in children, especially in the youngest ones (0-1 year-old) during all the surveyed period. An influenza outbreak during the second half of February was registered in the infantile population--school and low-age children--which gave most of the cases. Serology and isolations incriminated an influenza virus related to the A/Beijing 353/89 (H3N2) strain. Isolations worked out after the influenza epidemics pointed out circulation of parainfluenza and adenoviruses. The necessity is emphasized of the anti-influenza vaccination of low-age children.

Antiviral activity of some copper complexes of symmetrical triazines.

Copper complexes of hydrazones derived from symmetrical triazines inhibit the multiplication of three different types of viruses (Sendai, HSV and VSV) showing as well a scavenger activity on O2- radicals. The results emphasize the antiviral activity of these complexes which have at the same time a O2- radical scavenger activity.

[The incidence of viral and microbial antigens and the serum interferon titer in certain forms of rheumatism].

The presence of some viral and inframicrobial antigens in peripheral leukocytes was investigated by the indirect immunofluorescence technique in 120 patients with different forms of rheumatism and 50 clinically healthy controls. Mycoplasma pneumoniae and type 3 para-influenza virus were detected most frequently. The determination of serum interferon titer revealed a rise of this product in rheumatic patients.

[Dynamics of interferon in the rhinopharyngeal secretions of adults vaccinated with the NIVGRIP inactivated influenza vaccine by the nasal route].

The study of the kinetics of interferon in the nasopharyngeal secretions (NPS) of adults vaccinated by intranasal route with the NIVGRIP inactivated influenza vaccine and of unvaccinated controls led to the following observations: the large variety of the individual reactions to the IFN inducing action of the NIVGRIPR vaccine: the presence of low titers of a "physiological" IFN in the NPS of 26% of the patients before vaccination; a positive conversion to IFN in 60% of the patients after the first and in 33% after the second administration of the vaccine. No changes were noted in the percentage of IFN positive controls during the observation period. A "physiological" IFN was detected in the NPS of 10% of the controls.

[The antiproliferative effects associated with vincristine and human interferons in experimental in vitro systems].

Cytotoxic potential of suboptimal doses of vincristine associated with human interferon was studied in two cell lines of tumoral origin, as compared to the action of or gamma type interferon preparations. Results show that the vincristine cytotoxic effect may be synergistically augmented in both culture types by simultaneous interferon administration.

[Anti-proliferative and antiviral effects of human alpha-interferon on tumor cells].

Antiproliferative and antiviral activities of a type alpha human leukocytic interferon on several heteroploidic cell lines: HeLa, HEp-2, and T-10, a cell line of malignant origin (glioblastoma) were investigated, as compared to subcultures of human embryo fibroblasts. The tumor cell multiplication rate decreased proportionally to the amount of interferon in the culture medium. The highest interferon concentration used in our experiments (1,000 mu/ml) induced a decrease of the normal cell multiplication rate (human embryo fibroblasts). The same amount of interferon had a cytotoxic effect against the T-10 cells, but this phenomenon is reversible if the interferon is excluded after 24 h from the culture medium. There was no quantitative relation between the magnitude of the antiviral and of the cytotoxic effects of the type alpha human interferon on the tested cellular substrates.

[The humoral and cell-mediated immune response induced by the NIVGRIP inactivated influenza vaccine].

A comparative study was conducted in patients vaccinated with the NIVGRIP trivalent inactivated influenza vaccine and in placebo receiving controls on the kinetics of the serum hemagglutination inhibiting (HAI) antibodies and the neutralizing secretory antibodies in the nasopharyngeal secretions (NPS), of the blastic transformation of lymphocytes index, of the rosette formation index and of the serum immunoglobulins. A significant rise of the H.A.I. and the neutralizing secretory antibodies as well as of the blastic transformation of lymphocytes index was recorded after stimulation with the influenza vaccine. There were no significant changes in controls. No significant variations of the blastic transformation of lymphocytes index after stimulation with P.P.D. and of the rosette formation index were recorded in both investigated groups. Serum immunoglobulin titres showed significant variations in vaccinated as well as in control groups. The results point out the stimulating effect of the NIVGRIP inactivated influenza vaccine on both humoral and cell mediated immune responses.

The course of some viral respiratory infections in the south-east area of Romania during the cold season 1993-1994. Epidemiological serological and viral data.

Since 1991 we studied yearly the course of some respiratory infections in nine districts of the south-east area of Romania, by collecting epidemiological and sero-viral data during the cold seasons (1) (2). The results pointed out the most frequent circulating viruses, as well as the influenza strains which caused epidemic outbreaks. The same line of investigation was followed during the cold season November 1993-- March 1994 in the mentioned geographic area. In the present study are shown the results obtained in this period.

Investigations on the influence of the experimental infection with the herpes simplex virus type 1 on the electrophoretic mobility of some cultured cells.

Two types of cell cultures, BSC and HE-p2, have been studied from the microelectrophoretic point of view. Determinations have been made for infected cells (collected at 48 or 72 hours after infection) and for control cells, immediately after their separation from the culture, or after preserving them for 24 hours or more in migration buffer. The electrophoretical mobility increases with an average of 10% in the case of infected cells, as compared to the controls. This parameter offers a distribution of values varying from the gaussian curve to the bimodal one. From the microelectrophoretic point of view, the HEp-2 cells are more stable and respond more uniformly than BSC cells.

[The action of amphiphilic drugs on human fibroblasts in vitro. Their possible use in antiviral and antitumor chemotherapy].

The LDH cytotoxicity test is able to emphasize the stabilisation of human embryo fibroblast membranes, in vitro, by two amphiphilic drugs: the metomidate and the thymolol. The membrane stabilisation is emphasized by the reduced LDH externalization as compared with the untreated cells. The use of the LDH test for the selection of some natural complexes or synthetic drugs with membrane stabilising and potential antiviral activity is proposed. The inhibition of LDH by metomidate was recorded, as decreasing of the enzyme activity with increasing metomidate concentration. This fact explains the known lactic acid accumulation, under metomidate treatment of human subjects. The use of metomidate in classical cancer treatment potentiation is proposed instead of lactate externalization inhibitors, already used for this aim.

Studies regarding a fibrosarcoma induced in Sprague-Dawley rats by a murine cytomegalovirus. Note I. Aspects related to the character and reproduction of the tumor.

Through passages on Sprague-Dawley rats (performed during seven years) the serial transmissibility of a fibrosarcoma type tumor, with a tendency to local recurrence and locoregional metastasis formation, is demonstrated. The tumor was initially obtained by in vitro transformation of primary total rat embryonic cultures under the influence of a murine cytomegalovirus (M-CMV). In the course of passages, susceptible (S) and resistant (R) animal lines to the respective tumor could be obtained. The R and S crossings demonstrated the dominant transmission of S.

Epidemiological, viral and serological studies concerning the evolution of some acute respiratory diseases between November 1992 and March 1993 in nine districts of the south-east area of Romania.

Viral and serological studies led to the conclusion that parainfluenza type 3 and influenza type A(H3N2) viruses were the most implicated in the etiology of acute respiratory diseases (ARD), viral pneumonia and clinical influenza in nine districts of the south-east area of Romania during the November 1992--March 1993 period. Epidemiological survey pointed on the 0-1 year group of age as the most affected by the above mentioned respiratory diseases.

[Membrane microviscosity in cell-virus systems].

The use of fluorescent probes for the appraisal of microviscosity properties of cell membranes, both inside the lipid double layer and/or near the double layer surface, are today an usual tool in cell biology. In the present paper an attempt was made to establish if there are significant changes in the mechanical properties of the host cell membranes.

Experimental infections with some pneumotropic viruses in the mouse. Note I. Virological and pathomorphological aspects of the associated infections with influenza virus, adenovirus and respiratory syncytial virus.

In white mice, the associated infections with the influenza virus type A(H3N2), the adenovirus type 3 and the long strain of the respiratory syncytial virus, detected by the appearance of homologous serum antibodies and by positive I.F. reactions in pulmonary tissue, induce histological, histochemical and histo-enzymatic lesions, the severity of which exceeds that of the lesions in animals infected with only one of the mentioned viruses. In comparison with the morphological picture in animals inoculated with a single virus, expressed by inflammatory, infiltrative, lymphohistiocytic pulmonary (infiltrative, interstitial, diffuse, sometimes "cuffing", peribronchial and perivascular broncho-pneumonias), hepatic and renal processes, in the case of associate infections pathomorphological changes appear on wider surfaces and with a high frequency, manifested also by hyalinosis of the vascular media in the lung and the spleen, vacuolization of the cytoplasm of hepatocytes, hypertrophy of the lymphoid follicles in the spleen and the lymph nodes (in associated infection due to the influenza virus+adenovirus and vice versa) or, on the contrary, hypotrophy of the lymphoid follicles in the spleen and lymph nodes (in the associated infection due to the influenza virus+respiratory syncytial virus and vice versa).
