[Morphogenetic aspect of the influence of purified birch tar on skin].

Changes of the histological structure of rat's skin were studied two weeks following the daily applications of native birch tar and of purified birch tar, devoid of the components that could be potentially dangerous for humans. The morphometric investigations were performed with the use of computer technologies on the apparatus-programme complex (DiaMorph, Russia). It has been established that the main morphological manifestations of specific dermatotropic activity of birch tar in the present experiment included: epidermal hypertrophy, hyperkeratosis, stimulation of the activity of all dermal cellular elements, intensification of blood supply, reduction of sebaceous glands. Analysis of morphological findings indicates the bioequivalence of the native and purified birch tar in terms of its specific dermatotropic activity.

[Morphological aspects of dermatotrophic action of methyluracil applied epicutaneously].

The changes of the histological structure of intact skin and of the skin within the area of burn trauma, caused by treatment with methyluracil-containing ointments, were studied. Morphometric investigations employing computer technologies have been performed using "DiaMorph" device-program complex (Russia). It was established that methyluracil possessed wound-healing properties by stimulating epidermal proliferation in the zone of injury and by modifying an inflammatory reaction enhancing its fibroblastic component. Daily applications of methyluracil to intact skin for a period of half a year resulted in some reduction of reserve capabilities of cambial keratinocytes, that was objectively manifested by a decrease of epidermal germinal layer thickness.

[Retinoic acid modification of cell culture used for reproduction of enteropathogenic viruses].

The 0.001-0.005% retinoic acid injection into the growth cultural medium of prime and continue cell cultures 12-24h before inoculation considerably raised the cell sensitivity to animal entero- and coronaviruses. The entero- and coronaviruses concentrations in cultural medium increased by 10(1.58) and 10(1.68)TCID 50/1.0 respectively. The optimized parameters of the cell culture processing for the enteropathogenic viruses reproduction improvement are proposed.

[DNA synthesis and content in the nuclei of epidermal cells of mice during their differentiation and specialization].

Synthesis and content of DNA in the nuclei of differentiating cells of mouse skin epidermis was studied by using cytomorphometric, autoradiographic and cytophotometric methods. It has been shown that the cells of the keratinoid series divide only in the basal layer and contain 2-4c DNA. Keratinocytes of the thorny layer are mostly tetraploid, 2c cells are lacking. H4c and 8c cells comprise 12% of the population. In the keratinocytes of the granular layer DNA content is somewhat lower due to nuclei break down and conversion of cells into anucleate scale. Part of the melanocytes of the basal layer also contain 4c DNA. Highly specialized element of the basal layer Merkel and Langerhans cells are polyploid. Conclusion is drawn that DNA hyper-replication by multiplication of the whole genome is part of the development program of the population.

[Changes in the functional activity and kinetics of macrophages and lymphocytes as affected by retinoic acid].

The influence of water-soluble compound of retinoic acid on the ability of human blood cell precursors to transform into macrophages, phagocytic macrophage activity, proliferation and marker properties of lymphocytes and the condition of erythrocytes was studied. It was found that retinoic acid caused erythrocyte damages, induced the ability of precursor cells to transforms into macrophages, did not increase significantly their functional activity and facilitated the formation of granulocyte-lymphocyte-macrophage aggregates. In the absence of macrophages, the action of retinoic acid was followed by the decrease in the expressivity of precursor cells to transform into macrophages, did by the suppression of their proliferative activity.

[Effect of retinoic acid on the aggregation of erythrocytes and leukocytes in the blood].

Human and animal blood smear staining with PAPh has revealed mononuclear leukocyte-erythrocyte aggregates. Administration of retinoic acid increased concentration and dimensions of these aggregates and was followed by preferential accumulation of PAPh-negative osmotically unstable erythrocytes. Similar changes were detected in the blood samples of women engaged in the production of retinoic acid. Aggregate concentration showed positive correlation with erythrocyte sedimentation rate and no correlation with prothrombin time.

[Ligament system of the large intestine].

Unconstant ligaments of the large intestine perform the function of its additional fixation. Their number increases as the mobility of the large intestine intensifies. During operative interventions the ligaments of the large intestine should be preserved. If their cutting is necessary, they should be subsequently restored.

[Effect of retinoic acid on indices of humoral immunity].

The influence of all-trans- and 13-cys-methylretinoate on antibacterial and antiviral immunity was studied in experiments on noninbred C57Bl/6 and (CBA X C57Bl/6) F1 mice. All-trans-methylretinoate was shown to stimulate the production of antibodies to E. coli antigens, with the effect being dose-dependent. At the same time both compounds inhibited the production of inhibitors, interferon and antibodies to influenza virus.

[Erythrocyte-damaging and immunomodulating effect of retinoids].

The influence of retinyl acetate, retinoic acid and its esterified cis- and trans-derivatives, retinoids C15 and C20 on erythrocytes, liver macrophages and white pulp of the spleen was studied in the experiments on mice of different strains. It was found that administration of these compounds leads to the damage of the blood cells, increase in the number of liver macrophages and the share of white pulp in the spleen and stimulation of the local response to tuberculin. The interdependence of changes in the blood and adjuvant is suggested.

[Effect of retinoids on rat spleen].

It has been shown in experiments on Wistar male rats with the use of light and electron microscopy and morphometry that a single intraperitoneal injection of high doses of all-trans-methylretinoate and methyl-7,8- dihydroretinoate leads to an increase in the spleen weight, the appearance in the spleen of red cells changed in the shape, activation of phagocytic intensity of macrophages, intercellular interaction, lymphocyte proliferation and formation of new lymph follicles.

[Effect of retinoids on the development and growth of experimental tumors].

The ability of six retinoids to inhibit cancer growth was tested on mice with Lewis lung carcinoma, PRG and RSM-5 tumors. It was shown that all tumors possess individual sensitivities to retinoids. The most effective substances were: methyl retinoate (all-trans and 13-cis isomers) and a retinoid with a small side chain and an oxidated beta-iononic ring (substance 3). Each of those three retinoids was tested for an ability to prevent methylcholanthrene-induced carcinogenesis in the skin. All-trans methylretinoate had the most pronounced effect; however, it was the most toxic, too.

[Antitumor and toxic properties of retinoid C15].

A study was made of antitumor and toxic properties of retinoid C15 which has some characteristic structural features of retinoic acid natural metabolites; such as ethyl-2E, 4E-3-methyl-5-/2,6-dimethyl-6-ethoxycarbonyl-/3-oxo-1-cyclohexene-1-yl/-2,4-pentadienoate. The substance inhibited the growth of transplantable tumors of the uterine cervix. Lewis carcinoma and rumen cancer. This ability was dependent on the tumor growth rate. The substance proved less toxic then retinoic acid or methyl retionate. Injection of retinoid C15 was accompanied by the widening of the paracortical zone of the mesenterial lymph node and by the increased number of lymphoid follicles in the spleen.

[Enhanced growth of transplanted tumor in appendectomized rats].

The effect of appendectomy on carcinoma RS-1 growth has been studied in young and old Wistar rats and in randombred ones. It has been found that the tumor grew more intensely in the appendectomized animals. This effect was evident in the young animals, both linear and randombred ones. It remained unchanged after administration of the retinoids (all-transmethylretinoate and retinoid C15) but did not develop in the old animals.
