[The direct effect of pancreatic kallikrein (andecalin) on blood coagulation and fibrinolysis indices].

A direct effect of the preparation of pancreatic kallikrein on plasma hemostasis state was shown in experiments in vitro. Incubation of plasma with kallikrein led to an insignificant activation of contact factors of coagulation and suppression of the final stages of the clot formation at the level of the active thrombin formation and the fibrin-monomer polymerization. The growth of the anticoagulant and fibrinolytic activity was found. The possibility of studying the preparation as a thrombolytic agent in thrombotic situations is discussed.

[Effect of the pH of the medium and chemical modification of protein on the activity of bradykinin angioreceptors].

Study of the effect of medium pH on the mechanism of bradykinin interaction with receptors allowed a suggestion about the presence of a protein component in the composition of bradykinin receptor. Quantitative determination of the pH-dependence of the kinetic parameters of the kininergic reaction indicates the involvement of sulfhydryl groups in the formation of the active center in bradykinin receptor. This finding is supported by experiments with receptor blockade by reagents used for chemical modification of protein. The data obtained also attest to a possible involvement of the imidazole group of histidine in the realization of the myotropic effect of bradykinin.

[Role of the blood kallikrein-kinin system in activating hemodynamic effects induced by L-thyroxine].

Intravenous injection of large L-thyroxine doses (200 micrograms/100 g) was shown to activate kinin formation and to reduce arterial pressure by 33% in experimental white rats. It was established that sensitivity of bradykinin receptors increases during experimental thyrotoxicosis. During kininogenesis inhibition with contrykal, a kallikrein inhibitor, as well as during the kinin system components deficiency, intravenous injection of the same thyroxine doses did not result in arterial pressure changes. The results obtained suggest that hypotensive effect of thyroxine is mediated via the kallikrein kinin system.

[Use of thermostable DNA polymerase from Thermus thermophilus KTP in a combined reverse transcription and amplification reaction of detecting interleukin 2alpha RNA and determining expression of the multidrug resistance gene (MDR-1)].

According to our data native Tth DNA polymerase displays higher reverse transcription activity than Taq DNA polymerase. This allows one to use Tth DNA polymerase in the complete reaction of reverse transcription and amplification (RT/PCR). We used this enzyme to synthesize the interleukine (IL-2 alpha) RNA template synthesized by the RT/PCR method in vitro. The conditions for RNA IL-2 alpha detection were optimized. The maximum yield of the specific product was obtained at pH 8.5-9.0. The influence of bivalent cations on the efficiency of RT reaction of coupled RT/PCR can be expressed as: Mn2+ > or = Cu2+ > Mg2+ > Cd2+ >> Co2+. The optimal ratio is 1.25-1.88 for Mn2+/dNTPs and 1.88-2.5 for Cu2+/dNTPs and Cd2+/dNTPs. The maximum yield of the RT/PCR product is found at Mg2+/dNTPs = 3.75. When Mn2+ is used instead of Mg2+ in the PCR reaction the efficiency of RT/PCR decreases. The RT/PCR method embracing thermostable Tth DNA-polymerase provides detection of 10(3) copies of RNA IL-2 alpha. An efficient method of the express-diagnostics of MDR-1 gene expression by coupled RT/PCR using Tth DNA polymerase is described.

[Use of thermostable DNA polymerase from Thermus thermophilus KTP in a combined reverse transcription and amplification reaction for detecting CD4 receptor mRNA].

The RT/PCR method was applied to study a possible use of Tth DNA-polymerase for coupled reaction of reverse transcription and polymerase chain reaction (RT/PCR) on the CD-4 receptor mRNA template in the total cellular RNA. The conditions for detecting the CD-4 receptor mRNA were optimized. The pH-optimum for RT reaction was 8.8. The influence of Mn2+, Cu2+, Co2+, and Cd2+ cations in RT and PCR reaction was investigated. The efficiency of the RT reaction was shown to be the highest in the presence of Mn2+ (optimal concentration 1 mM). At Mn2+ concentration > or = 3 mM complete inhibition of RT/PCR was observed. The Tth DNA polymerase in RT/PCR was shown to be more effective than Taq DNA polymerase. The Tth DNA polymerase allows observation of the specific product in the gel containing ethidium bromide using 20 ng of the total RNA. High sensitivity and specificity of RT/PCR performed with the Tth DNA polymerase allow its wide application in the detection, quantitative analysis and cloning of cellular and viral RNAs.

[The demonstration of the genes controlling enterotoxigenicity in Staphylococcus aureus strains by using the polymerase chain reaction].

The selection and subsequent synthesis, according to the nucleotide sequences of S. aureus genes responsible for the expression of enterotoxins A and B, of highly specific primers for polymerase chain reaction Pcr were carried out with the use of the program "Primer". The optimum temperature conditions of polymerase chain reaction for all pairs of primers were selected. The method for the rapid determination of the enterotoxigenic properties of S. aureus strains by means of Pcr was proposed. The enterotoxigenic properties of several S. aureus strains were determined, which revealed that 3 clinical isolates had the gene of enterotoxin A in their genome, while laboratory strain FRI-722(H) carried the genes of enterotoxins A and B.

[Cloning of the gene for thermostable Thermus aquaticus YT1 DNA polymerase and its expression in Escherichia coli].

Using the phasmid vector pSL5, the genomic DNA fragment of T. aquaticus YT1 which contained the thermostable DNA polymerase (Taq-polymerase) gene was cloned. The BglII fragment of this genome locus was subcloned in the BamHI site of the pUC19 plasmid. To optimize the Taq-polymerase gene expression in E. coli cells, the gene was cloned in the correct reading frame regarding the initiation ATG codon of the pPR-TGATG-1 expression vector. The gene expression in this vector was controlled by the phage lambda PR promoter and the temperature-sensitive phage lambda repressor. We used PCR to amplify the short 5'-end fragment of the Taq-polymerase gene coding for the part into which an artificial SacI site was introduced. This site has been used for cloning the PCR product into the pPR-TGATG-1 vector, and the missing gene part was cloned into the KpnI site of the PCR product from the natural cloned gene. The cells of the E. coli PVG-A1 strain, which was obtained in the end, expressed efficiently the Taq-polymerase gene at the nonpermissive temperature. The content of the recombinant Taq-polymerase in the cells was about 1-2% of total proteins. The purified nearly homogeneous Taq-polymerase amplified efficiently in the PCR DNA fragments up to 5.5 kb long and was useful in DNA sequencing the by Sanger method. The half-life of the purified Taq polymerase was about 60 min at 95 degrees C, it was active for at least 65 standard PCR circles. The specific activity of recombinant enzyme preparations was about 180-200,000 units per mg of protein. The E. coli PVG-A1 strain enables one to isolate up to 500,000 units of purified enzyme from 2 l of bacterial culture.

[The effect of different motor regimens modulating spontaneous activity on rat behavior].

A method was developed of non-stressful modulation of spontaneous motor activity of rats. Restraint of mobility was found to inhibit spontaneous activity. Physiological stimulation of muscle activity by means of complication of food-procuring behaviour was accompanied by increase of spontaneous activity. Physiological stimulation of motor activity was characterized by stability of orienting-exploratory behaviour, emotional reactivity, expression of "freedom response", the best learning and working abilities of the animals. Regimes with imposing or restriction of muscle activity favoured the inhibition of spontaneous activity and the decrease of efficiency of adaptive behaviour. Motor regimes accompanied by increase of spontaneous activity were found to be optimal for adaptive behaviour.

[A DNA analysis of Vibrio cholerae strains by the polymerase chain reaction].

The method for the analysis of cholera toxin gene in V. cholerae strains was developed on the basis of polymerase chain reaction (PCR). This specific and highly sensitive method using primers affecting the site of the DNA of the operon of cholera toxin gene made it possible to identify one copy of V. cholerae genome. For the first time the content of cholera toxin gene in 4 V. cholerae (eltor) strains, obtained from the clinical material of cholera patients in Tajikistan and Dagestan, was shown with the use of PCR.

[A comparative study of the information value of the immunofluorescence and immunochromatographic identification of Chlamydia trachomatis antigens in smear material from the cervical canal and of the cytological picture of a vaginal discharge in pregnant women].

The cytological picture of the vaginal discharge and scrape material, obtained from 30 pregnant women of the control group (not infected with C.trachomatis) and 61 pregnant women with chlamydiosis; of these, in 42 women the comparative identification of chlamydiae by the methods of direct immunofluorescence and immunochromatography was carried out. Direct immunofluorescence was carried out with the use of a set of reagents "MicroTrak" (USA) and immunochromatographic identification was made with the use of a set of reagents "Clearview Chlamydia" (Britain). The comparison of the results of immunochromatography and direct immunofluorescence revealed that the sensitivity of the set "Clearview Chlamydia" was 100.0% in comparison with the data obtained in the examination of women with the use of the set "MicroTrak". The negative results coincided in 90.1% of cases. The common features of the cytological picture of vaginal samples taken from pregnant women were established. The optimum system of the examination of pregnant women suspected for chlamydiosis, as well as for the evaluation of the effectiveness of its treatment, was proposed.
