[Diversity and unity of methods of replication of viral DNA].

A systematization of the modes of viral DNA genomes replication was attempted. The key problem of replication, the complete copying of the genome, has several fundamentally different ("strategic") solutions, which utilize different mechanisms of initiation of a round of replication. Noticeable diversity may exist also within a given strategy, since consecutive replication steps may be accomplished by several (but not many) distinct means. There appears to exist a significant similarity between the viral and cellular replication systems. The approach used permits an assessment of the actual diversity in DNA replication systems.

[Synthesis and membrane-dependent processing of a precursor of tick-borne encephalitis virus (flavivirus) structural proteins in cell-free systems].

Conditions that permitted cell-free synthesis of at least one of the non-structural, in addition to two-structural, polypeptides of tick-borne encephalitis virus have been found. The time course of accumulation of virus-specific polypeptides in extracts of Krebs-2 cells and reticulocyte lysates as well as the peptide maps of the products synthesised were studied. A model of generation of viral structural polypeptides has been proposed, according to which a common precursor of these proteins while in a nascent form, is processed in a membrane-dependent reaction into a C-terminal segment, corresponding to the polypeptide moiety of envelope glycoprotein E, and an N-terminal segment, doublet p36/33. Subsequently, an N-terminal segment, corresponding to the core polypeptide C, is cleaved off from p36/33. The remaining C-terminal segment of p36/33 is possibly a precursor of the membrane polypeptide M. The translational strategy of flaviviruses is compared to that of other positive-stranded RNA viruses.

[Intertypic antigenic determinants of intracellular proteins of different poliovirus serotypes].

Antibody against intracellular virus-specific proteins was produced by immunization of monkeys with extracts of cells infected with poliovirus of one of the three serotypes. Interactions of homotypic and heterotypic antibodies with labeled intracellular virus-specific proteins were studied. Immune complexes formed at this interaction were adsorbed on the surface of Staphylococcus aureus (strain Cowan I) cells, and the virus-specific proteins contained in these complexes were then analysed by polyacrylamide gel electrophoresis. The experimental results indicate the occurrence of identical (or close) antigenic determinants at least in the majority of similar intracellular proteins of polioviruses of the three serotypes.

[The critical significance of a conserved single-stranded interdomain linker in the 5'-untranslated region of poliovirus RNA].

Translation of polioviral RNA is initiated by interaction of a small ribosomal subunit with internal segments of the 5'-untranslated region (5'UTR). Several mutations were constructed within 5'UTR segment 425-449. All of them (including a single C444-->U replacement) inhibited in vitro translation, which decreased about 10-fold. Two mutant constructs, pPV12-05 (C444-->U) and pPV12K (containing also an AAUU insert between positions 441 and 442) produced plaques on monolayers of susceptible cells. All the viruses isolated from these plaques exhibited a reversion at position 444; the template activities of the revertant RNAs were restored completely or significantly. The results show the importance of the relevant 5'UTR segment for the initiation of polioviral RNA translation.
