The granulocytopoiesis in the spleen of newborn rats.

The granulocytopoiesis in the spleen of newborn, 1h-old rats was analysed. It was found that the transit times of mycleocytes and metamyelo cytes were remarkably shorter than in adult animals, and that the mitotic frequency of myelocytes was only nearly half as high. The average generation time of immature myelopoietic cells was estimated to be 8.4h, the S-phase duration 4.0h. The findings may be attributed to an elevated input from stem cells in contrast to the undisturbed granulocytopoiesis of adult rats, which is obviously maintained by a higher divisional rate.

Diagnosis of hairy-cell leukaemia by tartrate-resistant acid phosphatase activity in paraffin-embedded tissue sections.

Tartrate-resistant acid phosphatase activity may be demonstrated in paraffin-embedded liver and spleen specimens as well as in decalcified bone marrow biopsies after fixation in a mixed glutaraldehyde-formaldehyde-calcium acetate solution. This technique may routinely be applied to haematologically relevant material and aids in the differential diagnosis of hairy-cell leukaemia.

[Actinomycosis of the liver (author's transl)].

A case report is given of a 61 year old female patient, who came to the hospital with chronic pain in the right upper abdominal quadrant, fever, chills, dyspnea, and general weakness. 4 days later she died in hemorrhagic shock after having had melena. At postmortem an actinomycotic abscess of the size of a childs head was found in the left lobe of the liver; in addition there was a highly differentiated leiomyosarcoma of the duodenum. Isolated visceral actinomycosis is rare; it should however be thought of when the causes of liver abscesses are being scrutinized. Diagnosis can be established only bacterioiogically or histologically.

[Pathology of liver lesions in malignant non-Hodgkin lymphomas (author's transl)].

The Kiel classification of lymphomas based on cytological criteria can be applied to tumor cell types found in liver infiltrates as well. Lymphomas with low degree of malignancy are delineated distinctly against the normal parenchyma; the borderline is slightly blurred only in the centroblastic-centrocytic subtype. Generally, the portal fields in these cases are round-shaped, expanded, and filled with lymphoid cells. Lymphomas with a high degree of malignancy are characterized on the contrary by portal fields with very irregular borderlines; in some cases there is a certain similarity to piece meal necrosis. There is an increased incidence of mitoses in the malignant tissue. Bile ducts are often disrupted. Hair cell leukemia can be differentiated in paraffin sections too, if tartrate-resistent acid phosphotase is present.

Morphologic and cell kinetic investigations of the spleen after repeated in situ freezing of liver and kidney.

In order to evaluate a possible primary or secondary immunologic response of the spleen after single or repeated in situ freezing of parenchymal organs such as liver and kidney within a four week period, light microscopic and cell kinetic investigations with tritiated thymidine were performed on spleens of non-germfree rats. Sham operations served as controls. The sham operations did not induce any significant morphological or cell kinetic changes in the splenic white pulp. After cryolesions were produced in the liver and kidney, the percentages of activated germinal centers, labeled germinal center cells, and cells in the perifollicular area of the lymphatic mantle and marginal zones increased, with maxima during the first 3 days. The investigations show that the cellular reaction of the spleen starts earlier and is more prominent after repeated in situ freezing than after a single cryolesion. These findings point to an immunologic response of the anamnestic type, and correspond to results after repeated freezing of normal and malignant tissue of the urogenital tract. These cell kinetic results are important in the evaluation of further immunologic studies involving the cryotherapy of malignant tissues.

[Animal experiments with 99mTc-diethyl-HIDA in acute complete bile duct occlusion (author's transl)].

In order to establish whether a complete obstructive jaundice can abolish the accumulation of diethyl-HIDA (EHIDA) in the liver parenchyma, the common bile duct was ligated in 14 mongrel dogs. Before as well as at regular intervals after ligature of the common bile duct, a sequence scintigraphy was performed with 2 mCi 99mTc-EHIDA. For evaluation, time-activity curves (Tmax, T1/2), and analogue scintigrams as well as laboratory parameters were used for assessment. Up to seven weeks after ligation of the common bile duct, there was a marked accumulation of EHIDA in the liver parenchyma. The relative liver uptake (liver/background ratio) fell from 8.9 to 2.7, whereas conversely the cholestasis indicators aP and bilirubine rose markedly. Tmax did not show any significant alterations, whereas T1/2 was prolonged from about one week after ligation. Because of the duct ligation, there was no excretion of activity into the intestines. Immediately after ligation of the common bile duct, the gallbladder was shown up as a "hot" area in which the majority of the applied activity appeared from about one hour p.i. Begining with the fifth to the seventh day after ligation, the gallbladder was seen as a "cold" area in the liver paraenchyma. Bilirubine and aP were raised by about 50 times the initial value. With longer lasting cholestasis, the scintigram no longer altered whereas bilirubine and aP rose further. Histological examination after ligation for more than five weeks showed slight alterations as a whole. Gamma-GT and in particular GPT were likewise slightly raised compared to bilirubine and aP. The conclusion was drawn from this that the good accumulation of EHIDA in the liver parenchyma which is to be observed without exception even in cholestasis lasting for several weeks could be explained by a relatively slight hepatocellular damage. Only when there is a consecutive parenchymal damage in extrahepatic jaundice, accumulation of EHIDA in the liver can be abolished.

DNA-synthesis in the rat thymus after focal thermolesions on abdominal organs.

The changes in the DNA-synthesis of lymphocytes in the different zones of the rat thymus were autoradiographically studied with 3H-thymidine after focal thermonecroses (740 degrees C/4 sec) on abdominal organs and sham operation (laparotomy). A single thermolesion was performed in separate groups of rats on the liver and spleen, whereas a third group received multiple heat lesions (liver, kidney, stomach wall). A slight decrease of the labeling index occured during the first 36 h after the sham operation and was attributed to an unspecific stress effect leading to an increased release of corticosterones. The most remarkable and contrasting result was the observation that after thermonecroses a decrease of the labeling index of thymus-lymphocytes did not happen during the first 48 h but rather a marked increased with a maximum on the fifth day. It was concluded that the stimulated DNA-synthesis in the thymus might be related to a cell mediated immune response against thermocoagulated tissue components or to an immediate T-cell dysfunction, which is known to develop after burns. In both cases the thymus might react with an increased division and maturation of lymphocytes regulated through a feed-back mechanism.

The wound healing of the liver after thermonecrosis. Autoradiographical investigations with 3H-thymidine on rats.

Histological and autoradiographical studies were performed on the liver of rats after local thermonecrosis. The results were compared with those after local cryonecrosis. After thermocoagulation (740 degrees C/4 sec) each animal received an i.p. injection of 3H-TdR (2.5 micronC/1 g body weight). The survival times ranged from 12 hours to 30 days. In the HE stained histological slides the morphological changes and the percentages of the cells in the granulation tissue and in the stripping film autoradiograms the percentages of radioactively labeled fibroblasts and hepatocytes were determined. After 12--24 hours the thermonecrosis is complete with leucocytic demarcations. Five days later widespread granulation tissue develops in the marginal zone of the necrosis. 20 to 30 days after thermocoagulation small necrotic parts are still seen in the center sorrounded by a fibrous scar. Autoradiographically the highest percentages of labeled epithelial and mesenchymal cells are observed between the second and the third postoperative day. Four weeks after thermonecrosis there are still increased cellular proliferative activities. Therefore, it can be stated that the inflammatory resorptive woundhealing processes following thermonecrosis in the liver are maintained much longer than after other physical injuries especially after cryonecrosis.

[Pulmonary rhabdomyosarcoma (author's transl)].

A 55-years old patient is reported who became ill with recurrent attacks of coughing and haemoptysis some 3 weeks before death. An extensive mediastinal tumor was demonstrated radiologically. Pathoanatomical examination showed a pleomorphic rhabdomyosarcoma of the lower left pulmonary lobe with metastases in the right lung and regional lymph nodes.

Histological and autoradiographical findings in the immunologically stimulated spleen.

In order to estimate the role of a possible immunological coeffect on the splenic cellular proliferation during wound healing after mechanical or thermal lesions of internal organs, light microscopical and autoradiographical investigations with tritiated thymidine were performed on spleens of non-germfree rats, immunized by a single injection of sheep erythrocytes. Two days after an initial dissociation of preexisting germinal centers in the spleen a steep rise of the percentages of labeled cells in newly formed germinal centers occurs with a maximum on the fifth day. In the marginal zone an increased cell proliferation (mostly large lymphoid blasts) starts at 12 hours. In the lymphatic mantle zone a marked increase of labeled basophilic blasts (immunoblasts) can be observed with a maximum on the second day. The values are higher in the perifollicular B cell region than in the periarteriolar T cell region. In the red pulp of the spleen the highest percentages of labeled cells occur five days after the antigen injection. These findings in the spleen after a strong antigenic stimulation are characteristic for an immunological reaction of anamnestic type. In comparison with investigations of the spleen after cryolesions in internal organs such studies may be particularly helpful in judging the proportion of an unknown immunological reaction in the spleen after such an operation, which by tissue necroses and cellular destruction may have caused denaturation of self components.
