Role of anticholinesterase mechanism in suppression of antibody formation during acute poisoning with organophosphorus compounds.

Experiments on Wistar rats showed that acute poisoning with organophosphorus compound dimethyl dichlorovinyl phosphate (0.5 LD(50)) was accompanied by two opposite effects: inhibition of acetylcholinesterase in T lymphocytes leading to suppression of thymus-dependent antibody formation (predominant effect) and acetylcholine-induced stimulation of antibody production. Acetylcholine activated acetylcholinesterase in intact T cells.

[Lectin-induced platelet aggregation inpatients with unstable angina pectoris treated with anti-aggregation agents].

Lectin-induced platelet aggregation was studied in patients with unstable angina pectoris treated with tiklid and aspirin. ADP, PHA-P, WGA, Con A served as the aggregation inductors. In unstable angina pectoris lectin-induced platelet aggregation was enhanced. Changes in this aggregation in response to treatment with antiaggregants with different mechanisms of action are demonstrated.

[Role of parathyroid hormone in regulating the functional activity of platelets].

We studied the influence of parathyroid hormone (PTH) on the functional activity of white rat and human platelets, and examined in particular possible mechanisms of PTH influence on the platelet aggregation activity. It has been stated that PTH renders a marked dose-dependent proaggregative effect on platelets. Possible mechanisms of proaggregative effect of parathyroid hormone were examined on platelets using substances with defined mechanisms of the effect. Examination of PTH effect on lectin-intermediated aggregation in a suspension of washed platelets shows that metabolic activation of platelets by PTH causes an increased expression on their plasmic membrane mainly of glycoprotein complex IIb-IIIa and in a lesser degree of glycoprotein complexes Ia-IIa and IV which take part in the formation of interplatelet contact.

[Comparative evaluation of the efficiency of the effect of very high frequency electromagnetic waves on platelet functional activity].

A comparative analysis was made of the effect of two kinds of EMI MMD-radiation: EMI MMD-waves, generated by a vehicle "Jav-1 M" (42.2 and 53.5 HHz), and EMI MMD-waves exerting influence with frequencies of molecular spectrum of radiation and nitric oxide absorption (150.176-150.644 HHz), obtained with a specially created generator, with respect to their influence on the functional ability of platelets of unstable angina pectoris patients. It was shown that in vitro EMI MMD-fluctuations with frequencies of molecular spectrum of radiation and nitric oxide absorption exert a stronger inhibiting influence on the functional activity of platelets of unstable angina pectoris patients. Features of the action of various kinds of EMI MMD-effect on the activative-high-speed characteristics of platelet aggregation are shown.

[The effect of electromagnetic waves of very high frequency of molecular spectra of radiation and absorption of nitric oxide on the functional activity of platelets].

A study was made of the effect of electromagnetic EMI MMD-fluctuation on the frequencies of molecular spectra of radiation, and nitric oxide absorption under in vitro conditions on the functional activity of platelets in patients with unstable angina pectoris, with the help of a specially created generator. At amplitude-modulated and continuous modes of EMI MMD-irradiation of platelet-rich plasma for 5, 15 and 30 min the platelet functional activity decreases, which was shown up in reduction of their activation and fall of aggregative ability. The degree, to which platelet functional activity was inhibited, depended on the mode of irradiation and on duration of EMI MMD effect. The most obvious changes in platelet activation and in their readiness to aggregative response were observed at a continuous mode of irradiation within a 15 min interval.

Lipid peroxidation during experimental cholera intoxication.

Cholera intoxication in albino mice was induced by intraperitoneal injection of endotoxin in doses of LD(16), LD(25), and LD(50) and combination of endo- and enterotoxin in doses equivalent to LD(25). Dose-dependent activation of superoxide dismutase, phasic changes in the contents of MDA and conjugated trienes and dienes, and modulatory influence of enterotoxin on catalase activity in the blood were observed during intoxication.

Mechanisms of immunotoxic effects of acrylonitrile.

Cholinesterase reactivator dipyroxime and hydrogen cyanide antidote anticyan partially restore cell and humoral immune response in mice after acute acrylonitrile poisoning (0.5 LD(50)). When used in combination these drugs virtually completely prevented the suppression of immune reactions. The main mechanism of immunotoxic effect of acrylonitrile is mediated through inhibition of T lymphocyte esterases and a(3) component of cytochrome c oxidase of immunocyte mitochondrial respiration enzymes, which is important for prevention and treatment of immune disturbances caused by this toxin.

Blood irradiation by He-Ne laser induces a decrease in platelet responses to physiological agonists and an increase in platelet cyclic GMP.

The effect of He-Ne laser irradiation on platelet adhesion, activation and aggregation was investigated. Citrated whole blood was irradiated in vitro by He-Ne laser (632.8 nm, 7 mW) and then subjected to shear stress (1300 s-1) on subendothelial extracellular matrix (ECM)-coated plates. Laser irradiation was followed by a decrease in platelet adhesion and aggregation on ECM under flow conditions in a time exposure-dependent manner (by 30-40%). The inhibiting effect of laser light on platelets was detectable up to 1 h after the termination of irradiation. Laser irradiation of either platelet-rich plasma, gel-filtered platelets, platelet-poor plasma, or packed blood cells followed by whole blood reconstitution revealed a marked decrease in platelet deposition on ECM only in the cases of platelet-rich plasma or gel filtered platelets. In conventional aggregometry, laser-treated platelet-rich plasma demonstrated a diminished platelet response to both thrombin receptor-activating peptide (TRAP), converting a two-wave aggregation curve to reversible, and to the protein kinase C activator PMA (by 45%). In flow cytometry analysis, irradiated platelets presented lower fibrinogen binding and P-selectin expression in response to TRAP. Laser irradiation had no additional inhibitory effect on dibutyryl cGMP- and dibutyryl cAMP-pretreated platelets. A 50% increase in cGMP level was observed in laser-treated gel filtered platelets, both in the presence and in absence of the phosphodiesterase inhibitor, isobuthylmethylxanthine. The results suggest that guanylate cyclase is one of the primary mediators of the laser effect on platelet function.

[Antiaggregation activity of the vascular wall and pancreatic ultrasound pattern in the development of chronic alcohol-induced pancreatitis].

There are ultrasonographic signs of pancreatic edematous changes in early chronic alcohol-induced pancreatitis and signs of fibrous changes in the advanced (over 10-year) disease. Progressive chronic alcohol-induced pancreatitis is followed by decreased vascular wall antiaggregatory activity more commonly detected in pancreatic tissue fibrous changes, which should be borne in mind in choosing the optimum treatment policy for the patient.

[Antithrombogenic activity of the vascular wall, hemostasis, and rheological characteristics of blood in patients with unstable angina pectoris and various types of hyperlipoproteinemia].

AIM: To examine antithrombogenic activity of the vascular wall, some hemostatic and rheological properties of blood in patients with unstable angina (UA) and various types of hyperlipoproteinemia (HLP). MATERIAL AND METHODS: 102 UA patients were divided into groups by HLP types according to D. S. Frideriksen. Antithrombogenic properties of blood vessel wall were examined with the cuff test, blood rheology--with AKP-2 analyser, hemostasis and lipid metabolism--with standard test. RESULTS: UA patients with HLP have low antithrombogenic activity of vascular wall. The most noticeable shifts to hypercoagulation were found in patients with HLP types 2a and 2b. Blood rheology was abnormal (high red cell aggregation in low deformability) in patients with HLP types 2a, 2b and 4. CONCLUSION: Patients with UA demonstrate relationships between antithrombogenic activity of vascular wall, intensity of lipid metabolism, hemostasis and blood rheology.

[Effect of certain lectins on platelet aggregation in healthy people].

A study was made of the ability of some plant lectins, which bind specifically to different carbohydrate determinants of glycoproteins, to induce the platelet aggregation in healthy humans. It has been shown that phytohemagglutinin (PHA) and wheat germ agglutinin (MGA) induce a more marked platelet aggregation than concanvalin A (Con A). Lens culinaris agglutinin (LCA) had a slight aggregate activity. It was pointed at different roles played by carbohydrate determinants of platelet glycoproteins in fulfilling their aggregation function.

[Laser spatial scanning in flow cytometry].

The main flow laser cytometry principles, based on the elastic light scattering, spheres of its applications, problems of its realization and utilization in the immunological investigations and diagnostics are analysed. The experimental model of a flow cytometer with laser probing beam space scanning, originally proposed by the authors, is described. The apparatus was tested by polystyrene latex spheres and biological objects. The experiments showed that the achieved sensitivity was enough to register red blood cells, their complexes and bacterial cells.

[The hemostatic system function of patients with severe forms of diphtheria].

Hemostasis of 100 patients with severe diphtheria infection was studied throughout the disease. The patients were found to have marked procoagulant, anticoagulant and fibrinolytic disorders. Antithrombogenic activity of the vascular wall was also abnormal. The above impairments correlated with the symptoms severity and are interpreted as DIC syndrome which ran subclinically or as hemorrhagic syndrome. The majority of the patients underwent a hyperhypocoagulant phase of DIC syndrome.
