A simplified method of specimen preparation for X-ray microanalysis of muscle and blood cells.

The effect of various steps in the preparation of muscle and blood cells for electron microscopic X-ray microanalysis, was studied with reference to calcium, zinc and copper levels. It was found that these elements are readily detectable in material fixed in glutaraldehyde and embedded directly in Epon 812. Alcohol dehydration, buffer rinses and post-osmication caused marked depletion of all elements examined.

Growth of diseased human muscle in combined cultures with normal mouse embryonic spinal cord.

Normal and diseased human muscle cells have been grown in combined cultures with 12-14 day embryonic mouse spinal cord explants. Nerve endings on myotubes were found by light and scanning electron microscopy, and motor end-plates were identified by a histochemical reaction for acetylcholinesterase. Contracting myotubes, never observed in cultures of human muscle alone, were found in a culture from normal muscle. Histochemical studies demonstrated the presence of strongly and weakly reacting myotubes for both ATPase pH 9.4 and NADH-TR, but could not be related to the development of fibre types. No differences in morphology or histochemical reactions were found between normal and diseased muscle cells in these combined cultures.

Neurological fragility in the newborn: Influence of medication in labour.

New anaesthetic and analgesic drugs continue to be introduced in labour, with little apparent concern about possible effects on the foetus or neonate beyond the more gross changes reflected by the Apgar score. A review is presented of some drugs currently used and some of the newer methods available for the assessment of the neonate. It is important that attention should be given to the subtler effects of drugs on the nervous system in addition to the gross effects on the respiratory and other systems as judged by the Apgar score. It is also essential to correlate neurobehavioural status of the newborn infant with an accurate assessment of gestational age, in order to produce a scale of reference against which effects of drugs administered in labour can be judged.
