The Drosophila gene Serrate encodes an EGF-like transmembrane protein with a complex expression pattern in embryos and wing discs.

We describe the molecular characterization of the Drosophila gene Serrate (Ser), which encodes an integral membrane protein. The extracellular domain contains two cysteine-rich regions, one of which is organized in a tandem array of 14 EGF-like repeats. Antibodies directed against part of the extracellular region confirm the localization of the protein in the membrane. In the wing imaginal discs, the protein is detected in those regions that are affected in the wings of two dominant mutations, SerD and SerBd. Both mutations as well as three out of eight newly induced revertants of SerD could be mapped molecularly to the transcribed region, confirming the identity between the gene Ser and the transcription unit characterized. During embryonic development, RNA and protein exhibit a complex expression pattern, which is, however, not correlated with an appropriate embryonic phenotype. Phenotypic interactions of Ser alleles with the neurogenic genes Notch and Delta coupled with the structural similarity of the proteins encoded by these three genes suggest close interactions at the protein level.

Subcellular distribution of peroxidized lipids in myocardial reperfusion injury.

Previous studies in our laboratory have demonstrated the peroxidation of myocardial phospholipid in a canine model of reversible global normothermic ischemia and reperfusion while on cardiopulmonary bypass. The present study examines the distribution of phospholipid peroxidation products in three major cellular organelle fractions of myocardium prepared by established centrifugal fractionation procedures (sarcolemma, sarcoplasmic reticulum, and mitochondria). These organelles were isolated from control (nonischemic) and ischemic-reperfused myocardium harvested during early reperfusion (5 min), when previous studies indicated maximal peroxidative injury in whole myocardial biopsies. Utilizing a more rapid analytic procedure for measuring phospholipid containing the conjugated diene chromophore in the polyunsaturated fatty acyl substituents, we were able to establish the fidelity of this procedure by comparing the results obtained with it to the previous more laborious analytic procedure (involving phospholipid hydrolysis with phospholipase A2 and subsequent derivatization for high-pressure liquid chromatography followed by gas chromatographic-mass spectrometric analysis). Analysis of phospholipid extracts from organelle fractions for evidence of peroxidative conjugated diene formation revealed that sarcolemmal membranes had the highest content of oxidized phospholipid containing the conjugated diene chromophore (mean 2.2 +/- 1.2 nmol phospholipid-conjugated diene/mumol phospholipid phosphorus, P less than 0.02 compared with control). Both sarcoplasmic reticulum and mitochondrial membranes were also peroxidized but to a much smaller extent (mean 0.4 +/- 0.2 and 0.3 +/- 0.25 nmol phospholipid conjugated diene/mumol phospholipid phosphorus).

[Antibodies against staphylococci in patients of a department for orthopedic surgery (author's transl)].

Sera from different groups of patients in a department of traumatic surgery were examined for staphylococcal alpha-antitoxin-titres. In comparison to a group of normal controls the following changes were found: 1. Patients, had significantly raised titres, if they had undergone surgery because of closed fractures, pseudarthrosis and luxations and even if the postoperative course was uneventful. This can be explained through the wide-spread occurrence of staphylococci in hospitals, which can invade the organisms through an incision, the skin or mucous membranes. These findings should be kept in mind while estimating staphylococcal antitoxin-titres in patients hospitalized over a long period. 2. Patients suffering from surgical infections and also paraplegics showed still more significantly raised antitoxin-titres in comparison to the normal controls or to patients, who had undergone aseptic surgery. Increased titres were found even in the sera of patients, whose wounds did not drain staphylococcus aureus. Patients suffering from chronic staphylococcal osteomyelitis showed the highest mean values of antitoxin-titres; these patients, however, had also the greatest variations with high and low levels. The question arouse whether a high or a low level corresponds to the severity of the disease. It was found that it is necessary to titrate different antibodies against staphylococcal toxins. In a second paper the results of the simultaneous determination of alpha-antitoxin and anti-PV-leucocidin in patients with posttraumatic bone infections will be presented.

[Studies on the influence of the short-time narcosis during bronchoscopy on the cellular immunity (author's transl)].

The investigations were carried out on 30 patients with different pulmological diseases before a bronchoscopy with Brevinarkon and one, four and seven days after the bronchoscopy. There were evaluated the absolute lymphocytes in the blood smear and the transformation rate in the phytohemagglutinin-induced lymphocyte transformation test. On the first day after narcosis the results showed a decrease of lymphocytes and on the following days there was to be found a suppression in the lymphocyte function.

[The evidence capacity of the lymphocyte-transformation-test and the quantitative immunoglobulin-determination in sarcoidosis (author's transl)].

In 50 patients suffering from a cytologically or histologically confirmed sarcoidosis the unspecific transformation rate by PHA in the lymphocyte-transformation-test (LTT) and the immunoglobulins IgA, IgG, IgM and IgD have been determined by the help of a simple radial immunodiffusion. The evaluation of the LTT took place morphologically. 600 cells were counted each set-up (3 different smears each PHA-set-up). For the purpose of a better morphological presentation of lymphoblasts in the LTT a smear of alkalineous hydrolysis was fixed and followed by a colouring according to PAPPENHEIM. There was a decreased PHA-transformation-rate in sarcoidosis as a symptom for a partial defect within the cellular immune area. The levels of immunoglobulins showed no deviations as against to standard values.
