[The action of trypsin and zymosan on the respiration of cultivated endothelial cells].

Trypsin and Zymosan induce an increase in respiration activity of cultivated endothelial cells. These effects are not due to the production of oxygen radicals. The effect of trypsin will be explained by interfering with the cellular homeostasis of calcium. The stimulated respiration after incubation with zymosan may be connected with the increased phagocytosis.

[Parathyroid sonography in long-term dialysis].

In a prospective study the prevalence of sonographically enlarged parathyroid glands in patients on maintenance hemodialysis was investigated and correlated with clinical and laboratory findings of secondary hyperparathyroidism. We examined 97 unselected patients with a 5 MHz probe. In 34% (33/97) a total of 69 parathyroid glands were detected with a statistically significant correlation between the period of dialysis and the incidence of parathyroid enlargement; 47.8% of the glands were hypoechoic, 34.8% moderately echogenic and 17.3% gave echoes similar to the normal thyroid parenchyma. A correlation was found between the finding of enlarged parathyroid glands and blood levels of calcium and alkaline phosphatase. Parathyroid enlargement was found sonographically before, or in the absence of clinical and laboratory findings of secondary hyperparathyroidism. Sonography is therefore a useful technique for early diagnosis of hyperparathyroidism in patients on maintenance dialysis.

Cerium as amplifying agent--an improved cerium-perhydroxide-DAB-nickel (Ce/Ce-H2O2-DAB-Ni) method for the visualization of cerium phosphate in resin sections.

A new visualization (Ce/Ce-H2O2-DAB-Ni) procedure for cerium (Ce III) phosphate in semithin and ultrathin plastic sections (Epon 812, Lowicryl K4M, glycol methacrylate) of rat kidney tissues that had been incubated before embedding for the demonstration of phosphatases (alkaline and acid phosphatase, 5(1)-nucleotidase, Mg-dependent ATPase) is described. For this purpose the hydrophobic Epon resin was removed in NaOH-ethanol solution, whereas the hydrophilic Lowicryl and methacrylate sections did not required any etching. The primary reaction product Ce III-phosphate was amplified in a Ce III-citrate solution, subsequently oxidized with H2O2 and then visualized in a H2O2 containing DAB-nickel medium (Ce IV-perhydroxy induced DAB polymerization principle). The method yielded a very clear localization of enzyme activity. The final reaction product (DAB-nickel polymers) in 0.5 - 2.0 microns semithin sections is blue-black; the background staining is completely prevented. An increase of the staining contrast was obtained by posttreatment with OsO4 (osmium black formation). Furthermore, the enzyme reaction product could be demonstrated in 40 nm thick ultrathin sections by silver intensification, which utilized the high argyrophilia of the polymerized DAB-nickel complexes. This procedure replaces the earlier published technique.

[Fetal movement and acceleration behavior in hypertension in pregnancy].

Under standardized conditions, 26 pregnant women with pregnancy-related hypertension of different degrees were submitted to cardiogram-synchronous recording of fetal body and respiratory movements. The gestational age was between the completed 34th and 38th weeks. Of the 26 fetuses of the risk group, 20 were normotrophic and 6 hypotrophic. 40 normotrophic unimpaired fetuses of the same gestational age served as a control group. The average duration of the examination period was 70 minutes. The fetal movement and acceleration behaviours did not differ between the normotrophic fetuses of the risk group and those of the control group. The hypotrophic fetuses from the risk group, however, had significantly lower fetal body and respiratory activities than the normotrophic fetuses from this group. The same holds true for the degrees of heart rate activity. It can be concluded that pregnancy-related hypertension as the only pregnancy complication does not have a measurable influence on fetal movement and acceleration behaviours. This is true for both treated and untreated pregnancy-related hypertensions. The severity of pregnancy-related hypertension is not primarily reflected in changes of fetal movement and acceleration behaviours, either. This means that the parameters presented are not suited to prognosticate the effects of pregnancy-related hypertension on the fetus a priori. The situation becomes quite different if in addition to pregnancy-related hypertension some intrauterine fetal growth retardation develops. In this case, the fetus indicates its impairment by increasingly impaired movement and heart rate activities as a consequence of chronic oxygen deficiency.

[Synchronous cardiotocographic registration of fetal body and respiratory movements in placenta insufficiency].

The extent of fetal movements and their reflections in the cardiogramme were studied in 44 normotrophic and 39 hypotrophic fetuses under standardized conditions using real time ultrasound examination of fetal body and respiratory movements and synchronous registration of fetal cardiogrammes from a uterus without labour. Altogether, hypotrophic fetuses exhibited fewer movements than normotrophic fetuses did. Numbers and parameters of accelerations in fetal heart rate occurring in connection with fetal body movements were less marked, too. The acceleration amplitude was particularly significant. Further comparisons between fetuses born with pHa greater than or equal to 7.20 and less than 7.20 or 1- and 5-minute Apgar scores greater than or equal to 8 points and less than or equal to 7 points revealed statistically significant (alpha = 0.05) differences in the frequency of movements and movement-associated accelerations in fetal heart rate only in fetuses with a body weight less than or equal to the 5th weight percentile according to Kyank (n = 21). Fetuses with a body weight between the 6th and the 10th weight percentiles (n = 18) and normotrophic fetuses did not, or did only faintly, reveal these differences. A ratio calculated from the acceleration amplitudes and the durations of associated body movements differed significantly between impaired and unimpaired born fetuses during defined intervals of the duration of fetal body movements.

[The behavior of cytoplasmic progesterone receptors in the cycling endometrium and the effect of hormonal contraceptives].

Concentration of cytosolic progesterone receptor (PR) was measured by means of an exchange procedure in the endometria of 80 women. Investigations carried out in 35 normal menstruating females showed a gradual increase of PR during the preovulatory and immediate postovulatory phases, decreasing to the lowest level at the end of the cycle. Measurements in endometrial samples obtained from women on combined hormonal contraceptives demonstrated a significant decline of PR concentration in the periovulatory phase resulting in a flattening of the sinusoid PR concentration curve. The stronger down regulation effect of the preparation containing 1.5 mg of dienogest is due to the higher progestogenic efficacy of dienogest in the respective dosage. We consider the down regulation phenomenon a useful means among others to characterize the progestogenic potency of a steroid.

Identification of [1-14C]pantothenic-acid-mediated modified mitochondrial proteins.

The in vivo administration of [1-14C]pantothenic acid, which is the precursor of coenzyme A, resulted in the radioactive labelling of several mitochondrial proteins in rat liver. The incorporated radioactivity could be released by glutathione or 2-mercaptoethanol. Two mitochondrial matrix proteins acetyl-CoA acetyltransferase (liver and heart), an enzyme involved in the biosynthesis or degradation of ketone bodies, and 3-oxoacyl-CoA thiolase (liver), a protein participating in fatty acid oxidation were identified as modified proteins. The radioactivity was localized exclusively in forms A1 and A2 indicating that these forms represent the modified states of the acetyl-CoA acetyltransferase protein. Kinetics of incorporation of radioactivity revealed an accumulation of the modified forms. The ratio of specific radioactivities of A2 compared to A1 was 2.41 +/- 0.15 (n = 10). After in vivo labelling with [14C]leucine, the specific radioactivity of acetyl-CoA acetyltransferase depended on the state of the enzyme protein. The unmodified enzyme exhibited a lower specific radioactivity than its modified forms suggesting different turnover rates of these proteins.

Light-microscopic histochemistry of non-specific alkaline phosphatase using lanthanide-citrate complexes.

New lanthanide methods for the histochemical detection of non-specific alkaline phosphatase in the light microscope are described and compared with already existing techniques for the light microscopical demonstration of this enzyme. To avoid formation of insoluble lanthanide hydroxide at alkaline pH citrate complexes with the capture ions cerium, lanthanum and didymium were used. A molar ratio of 11 mM citrate/14 mM capture reagent is proposed. For preincubated sections, pretreatment in chloroform-acetone and fixation in glutaraldehyde, for non-preincubated sections fixation in glutaraldehyde yielded the best results. 4-Methylumbelliferyl and 5-Br-4-Cl-3-indoxyl phosphate were found to be the most suitable substrates. For routine purposes 4-nitrophenyl, 1-naphthyl, 2-naphthyl and 2-glycerophosphate were also sufficient; naphthol AS phosphates were inferior but still suitable. After incubation for 5-60 min at 37 degrees C lanthanide phosphate was converted into lead phosphate which was visualized as lead sulfide. At pH 9.2-9.5 enzyme activity was demonstrated at many sites such as intestinal, uterine, placental, renal and epididymal microvillous zones, plasma membranes of arterial, sinus and capillary endothelial cells, vaginal and urethral epithelium, smooth muscle cells, myoepithelial cells as well as excretory duct cells of salivary and lacrimal glands and in secretory granules of laryngeal glands. In comparison with Gomori's calcium, Mayahara's lead, Burstone's and Pearse's azo-coupling, McGadey's tetrazolium salt and Gossrau's azoindoxyl coupling technique the lanthanide methods detected alkaline phosphatase activities at identical or additional sites depending on the respective procedure.(ABSTRACT TRUNCATED AT 250 WORDS)

The cerium perhydroxide-diaminobenzidine (Ce-H2O2-DAB) procedure. New methods for light microscopic phosphatase histochemistry and immunohistochemistry.

New light microscopic visualization methods were developed for the histochemical detection of non-specific alkaline and acid phosphatase, Mg-, Ca- and Na, K-dependent adenosine triphosphatase, myosin adenosine triphosphatase, glucose-6-phosphatase, 5'-nucleotidase and thiamine pyrophosphatase with cerium ions as trapping agents in cryostat and plastic sections. The techniques are based on the conversion of cerium phosphate into cerium perhydroxide by H2O2 which decomposes at 55 degrees-60 degrees C into cerium hydroxide and oxygen radicals. These radicals are able to oxidize diaminobenzidine (DAB) to DAB brown. Addition of nickel ions to the DAB-H2O2 mixture generates bluish-black stained nickel-DAB complexes. Compared with the classical metal precipitation, azo, azoindoxyl and tetrazolium procedures the H2O2-DAB and especially the H2O2-DAB-nickel methods provided identical or superior results in catalytic phosphatase histochemistry and immunohistochemistry when using non-specific alkaline phosphatase as the enzyme label.

[Critical evaluation of various methods of determining markers of fetal maturity in amniotic fluid].

11 methods for investigation of fetal lung maturity, fetal skin maturity and common maturity signs were described. All investigations have been carried out on the base of 61 amniotic fluid samples. A certain declaration of fetal lung maturity is possible by: 1. The absorbance of native amniotic fluid at 650 nm 2. Shake-test (Clements) 3. Average quantities of the keratinocytes of the amniotic fluid cell population. Good results, a small amniotic fluid quantity and a simple procedure characterise these methods. They are usefull in clinical practice.

[Determination of immunoglobulins IgG, IgM and IgA in maternal serum in patients with intrauterine growth retardation].

Immunoglobulin IgG, IgM, and IgA were analysed in maternal serum of 33 patients with intrauterine growth retardation and 30 patients without hypotrophic newborns. The estimations of the immunoglobulins were carried out by means of single radial immunodiffusion according to Mancini and coworkers. There were no significant differences between both groups of patients. Therefore IgG, IgM, and IgA are no additional parameters for estimation of the placental function of the 3rd trimester of pregnancy.

[Fetal tachycardia--a cause of nonimmunologic hydrops].

A case of nonimmune hydrops fetalis is demonstrated. The cause of this disorder was a severe fetal tachycardia. Prenatal findings, obstetric management, and neonatal outcome are described. Our data have been discussed with reference to the literature.

[Twin pregnancy--a special problem of modern obstetric medicine].

Basing on an exemplary organized health protection incorporating up-to-date technique, progressive management of delivery and optimal neonatal care, we were able to reduce the perinatal mortality of all pairs of twins at the UFK Jena in the periods between 1977/79 (n = 204) and 1980/82 (n = 174) from 8,33% to 3,30%. The difference was statistically significant (p less than 0.05). The late morbidity of these children also showed a statistically significant decline (p less than 0.05).
