N=82 shell quenching of the classical r-process "waiting-point" nucleus 130Cd.

First beta- and gamma-spectroscopic decay studies of the N=82 r-process "waiting-point" nuclide 130Cd have been performed at CERN/ISOLDE using the highest achievable isotopic selectivity. Several nuclear-physics surprises have been discovered. The first one is the unanticipatedly high energy of 2.12 MeV for the [pi g(9/2) multiply sign in circle nu g(7/2)] 1(+) level in 130In, which is fed by the main Gamow-Teller transition. The second surprise is the rather high Q(beta) value of 8.34 MeV, which is in agreement only with recent mass models that include the phenomenon of N=82 shell quenching. Possible implications of these new results on the formation of the A approximately 130 r-process abundance peak are presented.

Precision measurement of the 7Be(p,gamma)8B cross section with an implanted 7Be target.

The 7Be(p,gamma)8B reaction plays a central role in the evaluation of solar neutrino fluxes. We report on a new precision measurement of the cross section of this reaction, following our previous experiment with an implanted 7Be target, a raster-scanned beam, and the elimination of the backscattering loss. The new measurement incorporates a more abundant 7Be target and a number of improvements in design and procedure. The point at E(lab)=991 keV was measured several times under varying experimental conditions, yielding a value of S17(E(c.m.)=850 keV)=24.0+/-0.5 eV b. Measurements were carried out at lower energies as well. Because of the precise knowledge of the implanted 7Be density profile, it was possible to reconstitute both the off- and on-resonance parts of the cross section and to obtain from the entire set of measurements an extrapolated value of S17(0)=21.2+/-0.7 eV b.

Efficacy of Echinacea purpurea in patients with a common cold. A placebo-controlled, randomised, double-blind clinical trial.

Common colds are one of the most frequent acute illnesses with major economical impact. Echinaceae purpureae herba (Echinacin, EC31J0) has shown promising results in the relief of common cold symptoms and the time taken to improvement compared to placebo. This study was aimed to confirm these findings by performing a randomised, double-blind, placebo-controlled clinical trial. A total of 80 adult male or female patients with first signs of a cold were recruited. The number of days of illness with a complete picture of the common cold (defined by the modified Jackson score of at least 5 points and experience of rhinorrhea and/or a subjective sensation of having a cold) was the primary end-point. In the verum group the median time of illness was 6.0 days compared to 9.0 days in the placebo group, assigning zero time for patients without a complete picture (one-sided p = 0.0112). EC31J0 was well tolerated and clinically effective in alleviating symptoms more rapidly than placebo in patients with a common cold.

Psychophysiological stress responses in amputees with and without phantom limb pain.

The present experiment tested the hypothesis that phantom limb pain amputees show a different pattern of psychophysiological reactivity to stress compared with painfree amputees. Six phantom limb pain (PLP) and five painfree upper-extremity amputees were administered two groups of tasks: stressful and relaxing. The measured dependent variables were skin temperature recorded at both stump and intact site, heart rate, blood pressure, subjective pain and stress ratings. Phantom limb pain patients were characterized by higher stump temperature compared with phantom limb painfree patients. This effect was observed during the whole recording. Consistently with the between-subjects effect, when PLP patients exhibited higher skin temperature (during relaxation, compared with during stressful tasks), they perceived more pain. The experiment showed higher cardiovascular reactivity in PLP patients, specifically to the stressful free-speech task, which focused on recollection of the amputation event. As compared with the painfree patients, during the personal stressor, PLP patients' heart rate and systolic blood pressure increased, indicating a greater sympathetic response. Results indicate that peripheral factors, such as stump temperature reliably differentiate PLP from painfree patients. Moreover, the clear cardiovascular hyperreactivity observed in PLP patients during their report of amputation suggests that PLP is associated with a long-term emotional memory for the painful experience of that event. These results are consistent with most reports in the literature relating phantom pain development to the experience of preamputation pain.

Biotransformation of the aerosol propellant 1,1,1,2,3,3,3-heptafluoropropane (HFA-227): lack of protein binding of the metabolite hexafluoroacetone.

The biotransformation of the aerosol propellant 1,1,1,2,3,3,3-heptafluoropropane (HFA-227) was investigated in rats in vivo and in rat and human liver microsomes. In the urine of rats exposed to 5000 ppm HFA-227 for 6 hr, very small amounts of hexafluoroacetone trihydrate were identified as an HFA-227 metabolite by 19F-NMR. Fluoride concentrations in the urine samples (0-48 hr after the end of the exposure) from exposed animals were not significantly different from those found in samples from nonexposed rats. In rat and human liver microsomes, fluoride and hexafluoroacetone trihydrate formation from HFA-227 was detected in very low levels only in liver microsomes from pyridine-treated rats and in two of eight human liver microsome samples, which exhibited the highest cytochrome P4502E1 activities. Because some aldehydes may covalently bind to proteins and the formation of fluorinated protein adducts has been implicated in immune-mediated hepatitis induced by halothane, the binding of hexafluoroacetone trihydrate to proteins was also investigated. Hexafluoroacetone trihydrate also gave only a very small resonance in fluorine NMR experiments when binding to human serum albumin was studied in comparison with the acylating agent S-ethyltrifluoroacetate. Moreover, no fluorine-containing products were formed by the reaction of hexafluoroacetone trihydrate with N alpha-acetyl-L-lysine, and hexafluoroacetone trihydrate was not metabolized to fluorine-containing metabolites or inorganic fluoride in rats. Comparative studies in human liver microsomes demonstrated that a halothane metabolite may covalently bind to proteins; in contrast, metabolism and covalent binding of HFA-227 could not be demonstrated. In summary, these data indicate that HFA-227 is biotransformed at very low rates to hexafluoroacetone trihydrate but irreversible binding of hexafluoroacetone trihydrate cannot be demonstrated, even with the application of very sensitive methods, and is considered unlikely, based on the combination of the results obtained.

Role of P4502E1 in the metabolism of 1,1,2,2-tetrafluoro-1-(2,2,2-trifluoroethoxy)-ethane.

1. The metabolism of 1,1,2,2-tetrafluoro-1-(2,2,2-trifluoroethoxy)-ethane (HFE), a prospective chlorofluorocarbon alternative, was studied in rat and human liver microsomes and in rat in vivo. 2. HFE was metabolized to inorganic fluoride, trifluoroacetaldehyde hydrate, trifluoroacetic acid and difluoroacetic acid, which were identified by 19F-nmr in microsomal incubation. After i.p. dosing with 200 mg/kg HFE to rat, trifluoroacetic acid and trifluoroacetaldehyde hydrate were identified as urinary metabolites. 3. The formation of inorganic fluoride from HFE was used to quantify oxidative metabolism. In liver microsomes from untreated rat, formation of inorganic fluoride could not be detected. However, microsomes from rats treated with P4502E1 (2E1) inducers ethanol and pyridine catalysed the formation of fluoride at different rates. The extent of fluoride formation in microsomes correlated with the amount of 2E1 protein as determined by immunoblots with a polyclonal antibody and with the extent of oxidation of p-nitrophenol and chlorzoxazone, two specific substrates for 2E1. 4. In different samples of human liver microsomes, the formation of inorganic fluoride correlated well with the ability of the microsomes to oxidize chlorzoxazone and p-nitrophenol and the amount of 2E1 protein as determined by immunoblots. 5. The obtained results suggest that 2E1 plays a major role in the metabolism of HFE in rat and man.

Enhanced expression of the protooncogenes c-myc and c-fos in normal and malignant renal growth.

The protooncogenes c-myc and c-fos play an important role in growth and differentiation of renal tissue. They are highly expressed during embryogenesis in the mitotically active tubule epithelium, while in terminally differentiated tubule cells of the kidney the expression is completely shut off. Furthermore, induction of cell proliferation in cultured renal cells by addition of growth factors is preceded by enhanced expression of c-myc and c-fos. Increased expression of these protooncogenes is also obtained by treatment of kidney cells in culture with the potent nephrocarcinogen N-dimethylnitrosamine and also with the nephrotoxin and possibly nephrocarcinogen S-(1,2-dichlorovinyl)-L-cysteine. Finally, the expression of c-myc and c-fos is induced after unilateral nephrectomy during compensatory renal growth in the remaining kidney and also during regenerative cell proliferation after in vivo application of the strong nephrotoxins folic acid and mercury chloride.

[Treatment of frostbite of the extremities by combined axillary plexus and peridural anesthesia: a case report including blood levels of bupivacaine].

To enhance peripheral perfusion after local frostbite, temporary or definitive interruption of the sympathetic activity in the region of injury is useful as an adjunct to systemic treatment. The common catheter techniques of axillary or epidural block provide the advantage of continued sympathicolysis without the risks of serial punctures or irreversible surgical alterations of the autonomic nervous system. bupivacaine into the three catheters (Fig. 2). A very satisfactory clinical result was observed. There was no loss of fingers

Isobutene formation during halothane anaesthesia in man.

The concentrations of volatile hydrocarbons were studied in breath samples from patients before and after anaesthesia. In patients to whom halothane was administered, there was a significant increase in the concentration of isobutene (from 0.43 +/- 0.09 to 4.66 +/- 2.22 nmol litre-1). There was no change in the concentration of isobutene in six patients anaesthetized with ketamine and diazepam. The concentrations of ethane and pentane were not altered significantly in either group.

Comparative metabolism of the amphetamine drugs of dependence in man and monkeys.

The use of animal models to study drug dependence and tolerance requires that the species used metabolizes the drugs like man, a condition frequently not fulfilled by non-primate species. The metabolic fate of several amphetamine drugs, namely amphetamine, norephedrine, chlorphentermine and phenmetrazine, in the rhesus monkey and the tamarin and two non-primate species has been investigated and compared to that found for man. The findings show that the two primate species metabolize these drugs in a manner similar to that in man.

The nephrotoxin dichlorovinylcysteine induces expression of the protooncogenes c-fos and c-myc in LLC-PK1 cells--a comparative investigation with growth factors and 12-O-tetradecanoylphorbolacetate.

Previous studies in kidney cells showed that S-(1,2-dichlorovinyl)-L-cysteine (DCVC) induces both direct DNA damage and DNA double-strand breaks by activation of Ca(2+)-dependent endonucleases. The objective of this study was to investigate the effects of DCVC on the expression of the protooncogenes c-fos and c-myc in cultured kidney cells (LLC-PK1). Supplementation of the incubation medium with 10% FCS after 24 hr incubation in 0.2% FCS resulted in a clear, but comparatively weak induction of the expression of c-fos and c-myc in LLC-PK1 cells. Addition of 500 microns DCVC to the high serum incubation medium induced a further three-fold increase of the transcript levels. A similar increase in the absolute amount of c-fos mRNA was induced by a mixture of growth factors (epidermal growth factor/insulin/transferrin) and of c-myc mRNA with 12-O-tetradecanoylphorbolacetate. However, the kinetics of gene expression were different. In the presence of DCVC the expression of c-fos and c-myc increased continuously in a time-dependent manner during the entire incubation period. In contrast, with growth factors and 12-O-tetradecanoyl-phorbolacetate the maximum transcript levels were detected after 0.5 hr (c-fos) and 1 hr (c-myc), respectively; thereafter, a slight decrease was observed up to the end of the incubation time.
