[Subacute and subchronic percutaneous tolerance testing (28-day and 90-day test) of epicutaneously applied dis-infectants as demonstrated by the example of peroxyethanoic acid (author's transl)].

As compared to control animals, guinea-pigs to the shaved backs of which a dose of 0.16 ml of a 0.12% solution of PES/100g. of body mass had been applied twice daily for periods of 28 and 90 d, respectively (5 d of application being followed by 2 d without treatment), showed erythema, loss of hair, slower increase in mass, increased heart rate, smaller body mass/kidney mass and body mass/spleen mass ratios, increases in leucocytes, ASAT, ALAT, LDH I and LDH III. In the 28-d test, slight inflammatory symptoms of the liver, kidneys and heart were observed in the experimental animals. These symptoms were more marked in the 90-d test, granulomata in the livers being particularly striking. A pneumonia of moderate to very marked degree is suggestive of the activation of a clinically latent PES infect whereby the inflammatory alterations in the kidneys might be explained by the formation of metastases.

[Photodynamic injuries of erythrocytes during illumination with blue light in patients with icterus neonatorum].

In a prospective study the influence of intermittent phototherapy (with and without riboflavin substitution) on red blood cells of newborns was examined in vivo. Hemoglobin concentration and hematocrit values did not differ significantly between treated patients and untreated controls. However, after illumination during 24 h (real exposure time 12 h) with a highly effective NC 400-04 Narva lamp, we found a completely reversible decrease of the mean erythrocyte diameters. The changes were statistically significant at the 8th day of life and disappeared up to the 16th day after birth. Riboflavin administration immediately before starting the phototherapy was able to prevent this phenomenon. The glutathione reductase activity (method by BEUTLER) was not affected, neither by phototherapy alone nor by riboflavin administered additively. As a conclusion of our results we would like to recommend a general riboflavin substitution in newborns after phototreatment.

[Tryptophan metabolism and oligophrenia (author's transl)].

After dealing with the biochemistry of tryptophan metabolism the most important results obtained in humans are presented. Special emphasis is given to the hereditary defects of tryptophan metabolism associated with mental retardation and convulsions due to lack of pyridoxine. The author's findings demonstrate the existence of a hereditary disturbance of the tryptophan metabolism via kynurenine in a certain part of oligophrenic patients. This metabolic defect can be controlled by high doses of vitamin B6. Furthermore investigations conducted with a view to interpreting these results are discussed, especially the determination of kynureninase activity, serotonin blood levels and pyridine nucleotide synthesis.

[Phenylalanine hydroxylase activity in the liver as a parameter for distinguishing various forms of hyperphenylalaninemias (author's transl)].

The activity of phenylalanine hydroxylase in the liver without the cofactor 2-amino-4-hydroxy-6,7-dimethyl-5,6,7,8-tetrahydropterin is similar in cases of classical PKU (14 cases), mild hyperphenylalaninemias (2 cases), heterozygotes (1 case) and normal controls (18 cases). The addition of cofactor increases the activity 30-40-fold in normal controls and 2-5 fold in mild hyperphenylalaninemias or heterozygotes. In classical PKU the addition of cofactor is without effect or causes even a diminution of activity. Therefore, for classification of the various forms of hyperphenylalaninemias the effect of cofactor on enzyme activity is important. For decisions from this findings to be made as to whether treatment should or should not be used further investigations are needed.

[Tryptophan metabolism research in oligophrenic children. 4.: Activity and possibility of activating hepatic kynureninase after vitamin B6 treatment].

In 8 selected test persons of the 3rd series of experiments who were showing signs of pathological kynureninase activity, hepatic kynureninase was again established after 3 week's treatment with large doses of vitamin B6. The enormous in-vivo activation of the kynureninasis established goes to prove that apoenzyme synthesis may be stimulated by pyridoxine. On the basis of these results and the relative frequency of disorders dependent on B6, the question arises whether children from endangered families should not be given prophylactic vitamin B6 treatment from birth.

[Determination of kynureninase activity in oligophrenic children with hereditary taint, before and after vitamin B6 treatment].

Kynureninase activity assay in oligophrenic children with hereditary taint, before and after treatment with vitamin B6. The hepatic kynureninase activity was assayed in 25 oligophrenic children with hereditary taint, and having a positive tryptophan load test. Eight cases (32%) showed an enzyme activity below the 2-s limit; in two cases, the pathologic enzyme activity could be almost normalized by addition of higher P-5-P concentrations. In a second set of studies, eight children with an abnormally low kynureninase activity were re-examined following a 3-week treatment with vitamin B6. The enormous in-vivo-activation of kynureninase reached in these studies lends support to the assumption of a vitamin-B6-dependent "inborn error of metabolism" in the tryptophan-kynurenin pathway in some of the investigated oligophrenic children with hereditary taint.
