Activity of isoenzyme A and N-acetyl-beta-glucosaminidase in fibrotic liver tissue.

Isoenzymes of N-acetyl-beta-glucosaminidase were separated in the supernatant of 72 homogenized normal and fibrotic human liver. Patterns of isoenzymes were obtained by electrofocusing in thin layers of polyacrylamide gel. The activity of mesenchymal reaction in the liver tissue was determined by histological examination. The distribution of N-acetyl-beta-glucosaminidase isoenzymes was compared with the mesenchymal activity in the liver tissue. The results show a good correlation of isoenzyme A activity with mesenchymal activity.

Activity of procollagen-prolyl-hydroxylase and N-acetyl-beta-glucosaminidase in liver biopsies from patients with chronic liver diseases.

In 5 mg of liver tissue obtained by needle biopsy from patients with chronic liver diseases, activity of both Procollagen-prolyl-hydroxylase and N-Acetyl-beta-glucosaminidase was measured. The activity of both enzymes increased due to the activity of fibrosis, but prolylhydroxylase increased more. The relation of these two enzyme activities in the development of liver fibrosis is discussed.

Influence of somatostatin on carbohydrate absorption in human small intestine.

Oral carbohydrate tolerance tests with xylose, galactose and lactose were performed. After an interval of at least 24 h the same tests were repeated following an i.v. bolus and during infusion of somatostatin. Somatostatin does not influence xylose absorption. However, absorption of galactose and lactose is significantly reduced (p less than 0.01/0.008) during somatostatin infusion. On the other hand, serum levels of galactose remain unchanged despite administration of somatostatin, when galactose is given parenterally. The results support the assumption that the absorption process in small intestine is affected by somatostatin. Possible effects of somatostatin on hormones regulating the intestinal absorption and on energy-depending carrier mechanisms are discussed.

[On the influence of clanobutine on the formation of restraint ulcer in rats (author's transl)].

Prophylactic effect of 4-[(4-chlorobenzoyl)(4-methoxyphenyl)-amino]-butyric acid (clanobutine, Bykahepar) on restraint stress ulcer formation was studied in male albino rats. Number and size of ulcers were counted, pH value of gastric juice and plasma levels of corticoids, glucagon and blood sugar were measured. Rats treated with clanobutine had only the one-third of ulcers compared with the untreated animals. This effect was found to be dose-dependent. Clanobutine did not change plasma levels of corticoids, glucagon or blood sugar. Rise in pH value of gastric juice during stress exposition was found to be lower in clanobutine treated animals than in untreated controls. Blood flow in gastrointestinal tract was seen to be much better under the influence of clanobutine. The mechanism of action of clanobutine is discussed.

[Isoenzyme P of N-acetyl-beta-glucosaminidase in sera from patients with liver diseases (author's transl)].

The isoenzyme P of N-Acetyl-beta-Glucosaminidase (beta-NAG) is demonstrated in sera from healthy persons and patients with acute and chronic liver diseases by electrofocusing in polyacrylamide gel. Elevation of isoenzyme P is observed only together with elevation of Glutamate-Pyruvate-Transaminase (GPT) in patients with acute and chronic liver diseases of different etiology and activity. The correlation between isoenzyme P and GPT is significant. On the contrary whole beta-NAG is elevated due the activity of the mesenchymal reaction in the injured liver tissue, not related to enzymes of the cytosol like GPT.

Isoenzymes of N-acetyl-beta-glucosaminidase from human liver and serum: separation by electrofocusing in thin layers of polyacrylamide gel.

Isoenzymes of N-acetyl-beta-glucosaminidase can be separated by electrofocusing in thin layers of polyacrylamide gel. Patterns of isoenzymes from human liver tissue and serum show changes in liver diseases. Preliminary results indicate, that N-acetyl-beta-glucosaminidase isoenzyme A is increased in liver tissue from patients with chronic liver diseases in relation to the activity of mesenchyme reaction. The described method will be discussed in comparison with known separation techniques.

[Consideration of the mesenchyme-reaction in patients with cronical liver-diseases (author's transl)].

The lysosomal glycosidase N-Acetyl-beta-glucosaminidase (beta-NAG) is involved in the breakdown of connective tissue. The activity of this enzyme was determined in sera and biopsies of liver tissue from patients with chronic diseases of the liver of different aetiology. Elevation of enzyme activity was found to be related to an increase of the mesenchymeraction in the injured liver tissue. No correlation was found between activity of beta-NAG and of GPT, gammaGT and CHE.

[Serum lysozyme concentrations in patients with morbus Crohn before and after bowel resection (author's transl)].

Serum lysozyme (muramidase) concentrations were determined in five patients with Morbus Crohn before and after resection of inflammated bowel areas. The serum lysozyme activity which was elevated before surgical treatment in all patients fell to normal values after bowel resection within a few hours. Our findings suggest, that the elevated serum lysozyme reflects an increased lysozyme production in the inflammated bowel tissue.

Prophylactic effect of somatostatin on stress ulcer formation in rats.

The aim of the present study was to investigate the effect of somatostatin (growth-hormone release-inhibiting hormone) on ulcer formation during immobilisation stress. This was done in male Albino-rats to study the effect of somatostatin on number and size of ulcers, to calculate ulcer index, to measure pH-value of gastric juice as well as plasma levels of gastrointestinal hormones. Rats treated with somatostatin before and during stress exposition had only the third part of the ulcers compared with the untreated animals. Total ulceration area was less than the tenth of the untreated rats. Normal corticoid plasma levels during stress exposition were found in the lower range of normal values in somatostatin treated rats. Decrease of plasma gastrin during stress exposition exceeded the gastrin decrease of somatostatin treated rats. Rise of plasma glucagon was completely inhibited during somatostatin application. Results of serum glucose paralleled those seen in glucagon.

[Combined ultrastructural morphometric and biochemical investigations on the rat liver after ligation of the bile duct (author's transl)].

In the present study an ultrastructural morphometric analysis of the rat liver 1, 3 and 7 days after bile duct ligation was performed. Corresponding biochemical investigations included the determination of the cytochrom P 450 contents in the microsomal fraction. In the serum of the rats the activities of GOT and GPT were determined. The quantitative results have shown that the decreased P 450 contents of the microsomal fraction coincided with a considerable increasement of the surface of the smooth endoplasmic reticulum. The smooth endoplasmic reticulum was significantly increased from 1.83 m2/cm3 hepatocytes to 3.49 m2/cm3 hepatocytes after 3 days and remained on this level after 7 days. These results may be considered as quantitative evidence for the term hyperplastic, hypoactive endoplasmic reticulum. - As a second important result our quantitative investigations have shown that the increasement of GLDH activity in the serum especially 1 day after ligation of the bile duct coincided with a signficantly increased mitochondrial volume at this time.

[Mitochondrial morphology and metabolism in extrahepatic obstruction (author's transl)].

Corresponding to the increased activity of mitochondrial Glutamate-Dehydrogenase (GLDH) in serum in extrahepatic cholestasis a swelling of hepatic mitochondria could be shown on ultrastructural morphometric analysis. The morphometric alterations parallel the serochemical findings. A significant correlation could be calculated between the swelling of mitochondria and the activity of GLDH in serum.

Inhibition of stress ulcer formation with somatostatin in rats.

Prophylactitc effect of somatostatin (Growth hormone inhibiting hormone) on restraint stress ulcer formation was studied in rats. Rats treated with somatostatin before and during stress had only the fifth part of the ulcers of the untreated animals after 9 hours of immobilisation. Pathophysiologic mechanism for stress ulcer production is as well discussed as the prohibiting effect of somatostatin on ulcer formation. Prophylactic clinical use has to be considered.

[On thyroid phosphatase in human serum (author's transl)].

Patients with hyperthyreoidism often present an increased activity of alkaline phosphatase in serum. Determination of isoenzymes may help to assign to different organs an elevated enzyme-activity in serum. This report deals with a patient suffering from hyperthyreoidism. In his serum alkaline phosphatase of the thyroid gland could be identified by comparison with organ-extracts. Raised activity of this isoenzyme can contribute besides bone, liver and small bowel to the increased serum-activity of total alkaline phosphatase in patients with hyperthyreoidism.
