[Inhibition of alpha-interferon and cinnamic acid on proliferation of human lung cancer cell].

BACKGROUND & OBJECTIVE: Recently, the research showed that cinnamic acid (CINN) could inhibit some cancer cells. This study was designed to investigate the effects of alpha-interferon and CINN on proliferation of human lung cancer cells. METHODS: Human lung adenocarcinoma cell A549 were treated with alpha-interferon and CINN. The growth curve, soft-agar colony-forming rate, and methyl greenpyronine staining were used to analyze the inhibition to cell proliferation. The cell cycle was detected by flow cytometry(FCM). RESULTS: alpha-interferon and CINN could markedly inhibit proliferation of A549 (P < 0.05), promote the differentiation (P < 0.05), and reduce the colony-forming rate in the soft-agar drastically(P < 0.05), FCM showed that the cell cycle was arrested in the G1/G0 phase. CONCLUSION: alpha-interferon and CINN can inhibit the proliferation of lung adenocarcinoma cell. Combination of the alpha-interferon and CINN show the stronger effect that the alpha-interferon or CINN alone.

[A study on the use of mycobacterium vaccine in the treatment of tuberculous pleurisy].

OBJECTIVE: To study the effects of immunotherapy with mycobacterium vaccine in patients with tuberculous pleurisy. METHODS: The time for effusion resolution and lymphocyte counts in the effusions were observed in patients with tuberculous pleurisy who received anti-tuberculous therapy and thoracentesis (group A) or anti-tuberculous therapy and thoracentesis plus intramuscularly administered polysaccharide nucleic acid fraction of Bacillus Calmette Guerin (PSN-BCG,) (group S). In situ end-labeling technique of fragment DNA was used to detect the apoptosis of lymphocytes in the effusions. RESULTS: The average resolution times in group A and group S were 21.2 days and 28.7 days respectively (P < 0.01). The lymphocyte counts in the two groups after 1 week, 2 week, and 3 week therapy were 1.6 x 10(9)/L (A) and 2.1 x 10(9)/L (S) (P < 0.01), 0.83 x 10(9)/L and 1.52 x 10(9)/L (P < 0.01), 0.55 x 10(9)/L and 1.16 x 10(9)/L (P < 0.01) respectively. The average apoptotic half-time was 94 h (A) and 124 h (S) (P < 0.01), 84 h and 123 h (P < 0.01), 79 h and 120 h (P < 0.01) respectively. CONCLUSIONS: Mycobacterium vaccine was found to prolong lymphocyte activation, inhibit lymphocyte apoptosis, and delay the resolution of pleural effusions. It was found not helpful as an adjunct therapy for tuberculous pleurisy.

A pilot study on the combined therapy of granulocyte-macrophage colony-stimulating factor and hepatitis B vaccine on chronic hepatitis B virus carrier children.

OBJECTIVE: To observe the efficacy of treating intrauterine infected chronic hepatitis B virus (HBV) carrier children with a combination of granulocyte-macrophage colony-stimulating factor (GM-CSF) or hepatitis B immunoglobulin (HBIG) plus recombinant hepatitis B vaccine (rHBvac). METHODS: A total of 27 chronic HBV infected children, who were born to HBV carrier mothers and received hepatitis B immunoprophylaxis at birth, were randomized into 2 groups: one receiving a combined therapy of 50 micro g of GM-CSF plus 10 micro g of rHBvac injected intramuscularly at the same location (GM-CSF group, 14 children) or 200 IU HBIG and 10 micro g rHBvac in different muscles (HBIG group, 13 children) on a monthly four-dose schedule. HBV-DNA quantification and other HBV serological markers were tested before and after the four-dose therapy. RESULTS: Twelve children in each group completed the study. Of them, 3 children in the GM-CSF group and 4 in the HBIG group had elevated serum alanine transaminase (ALT) before the trial, and then 2 in each group became ALT normal after the treatment. Before the therapy, hepatitis B e antigen (HBeAg) positivity was found in nine children in the GM-CSF group and 10 in the HBIG group. One from each group had an HBeAg/anti-HBe seroconversion after the treatment. The quantity of HBV-DNA was significantly lower after the treatment (P = 0.023) in GM-CSF group, but was not significantly reduced in HBIG group. No subjects were found to be negative for hepatitis B surface antigen (HBsAg) after the treatment, and no serious adverse events occurred in either group. CONCLUSION: Combined GM-CSF and rHBvac therapy inhibit HBV replication in carrier children who were not protected after treatment with immunoprophylaxis.

[Anti-emetic effect of ethanol extract from "Wuzhuyu broth"].

OBJECTIVE: To investigate the anti-emetic effect of ethanol extract from "WuZhuYu broth" and its mechanism. METHOD: Three experiments were carried out test which extract has anti-emetic activity, such as CuSO4-induced pigeon's emetic response, gastric emptying in mice and ACh-induced or 5-HT-induced contraction in vitro gastric muscle in rats. Meanwhile, effect of anti-emetic extract on concentration-response curve to ACh, 5-HT, histamine was investigated. RESULT: 50% ethanol extract and 70% ethanol extract were identified as having significantly stronger anti-emetic activities with little side effect, which showed the significant effect on concentration-response curve to ACh, 5-HT, histamine. CONCLUSION: 50% ethanol extract and 70% ethanol extract contain more anti-emetic fractions, more anti-emetic fractions can be gained at the concentrations of 50% and 70% ethanol; the mechanism of anti-emetic effect is related to its antagonism to the receptors of ACh, 5-HT, histamine.

[Two mechanisms of the color-tunable organic electroluminescence by the voltage applied].

Two devices structures of ITO/PVK: Rubrene/Al and ITO/Alq3:MN-PPV/Al were developed. Their characters of photoluminescence and electroluminscence were studied. The result showed that there were great difference between the photoluminescence and electroluminscence at a doping ratio of 2 wt% in ITO/PVK: Rubrene/Al. It is concluded that energy transfer in these dye-doped devices occurred by Förster energy transfer processes. And the trap effect of electrons of Rubrene on holes of PVK resulted in different combination probability of PVK excitation in photoluminescence and electroluminescence. At the same time, the two EL devices were color-tunable when the bias voltage was increased. The mechanisms which cause color variation are different. In the first device, it indicates that the energy transfer process from PVK to Rubrene is not complete. The combination probability of PVK excitation which does not contribute to the emission of Rubrene is increased when the bias voltage increases. But in the second device, there are many micro-interfaces due to the phase-separation just like the interface in a double layer device. The tunneling interface barriers of carriers is the main cause of color variation in the second device.

Drug abuse, innate immunity and hepatitis C virus.

Since its discovery in 1989, hepatitis C virus (HCV) has become a major public health problem. HCV chronically infects an estimated 170 million people worldwide. The seroprevalence of anti-HCV antibody in the United States has been estimated at 1.8%, which corresponds to approximately 4 million people. HCV is the most common chronic blood borne infection in the United States, and the leading cause of liver transplantation in developed countries. Injection drug use is the dominant mode of HCV transmission and accounts for up to 90% of current infections. Opiates and other drug abuse, such as alcohol, have been implicated as cofactors in the pathogenesis of HCV disease. Injection drug use has been the most common risk factor identified in alcoholics with HCV infection. Both opiates and alcohol contribute significantly to morbidity and mortality from HCV disease. These drugs most likely act synergistically to promote the development and progression of HCV disease. However, there is limited information available concerning the interaction of the drug abuse with the host cell innate immunity against HCV infection, which is a major barrier to fundamental understanding of the immunopathogenesis of HCV disease. Therefore, defining the role of the drug abuse in the development of chronic HCV infection is of crucial importance and should provide practical guidance toward the reduction of risk factors that interfere with therapeutic approaches for HCV infection and disease. This review paper focuses on the interplay between drug abuse (opiates and alcohol), innate immunity and HCV in the context of the development of HCV disease.
