[Experimental research on immunological rejection in neural stem cells allograft].

AIM: To investigate the occurrence of immunological rejection in brain transplantation of neural stem cells (NSCs) in the rat of Parkinson's disease model. METHODS: NSCs derived from the brain of E14.5d SD rat were cultured in vitro. Single cell suspensions were grafted into the striatum of the rat model of Parkinson's disease. Surviving animals were sacrificed at 10, 21, 35 and 60 d after transplantation, and the brain tissue was stained with HE and immunocytochemically to detect the expression of tyrosine hydroxylase (TH), CD4, CD8 and major histocompatibility complex class II antigens (MHC-II). RESULTS: The expression of elicited CD4, CD8 and MHC-II was detected within and around the allografts in the graft groups at 10 and 21 d after grafting and was subsiding at 35 d. At 60 d only very occasional immunopositive cells were present. The number of TH positive neurons was low at 10, 21 d, but increased at 35 d and 60 d. There was no significant difference between negative and positive control groups at different time points. The rotation behavior of PD was decreased 35 d after transplantation. CONCLUSION: Intracerebral transplantation of NSCs did not cause remarkable immunological rejection.

Effects of 5-hydroxytamine and its antagonists on hepatic stellate cells.

BACKGROUND: Hepatic stellate cell (HSC) plays a key role in hepatic fibrosis. This study was undertaken to investigate the expression of 5-hydroxytamine receptors in HSC and the effect of 5-hydroxytamine on biological characteristics of HSC. METHODS: Liver ex vivo perfusion of collagenase and density gradient centrifugation were used to isolate HSCs. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of 5-hydroxytamine receptor subtypes 1A, 2A, 2B and 3. Western blot hybridization was used to elucidate the effect of 5-hydroxytamine and its 2A receptor antagonist ketanserin and 3 receptor antagonist ondanosetron on the expression of transforming growth factor-beta1 (TGF-beta1) and Smad4 in HSC. RESULTS: HSC expressed 5-hydroxytamine receptor subtypes 1A, 2A and 2B. 5-hydroxytamine significantly increased the expression of TGF-beta1 and Smad4 in HSC (P<0.05). This action can be antagonized by ketanserin, not by ondanosetron. CONCLUSIONS: HSC expresses 5-hydroxytamine receptors. 5-Hydroxytamine could effect the biological characteristics of HSC through its receptor mediation, and may play a role in the pathogenesis of liver cirrhosis and portal hypertension.

[Design of a hollow-fiber bioreactor and perfusion study in vitro].

This article presents the design of a bioreactor using hollow fiber membrane and, isolated hepatocytes of suckling pigs, and the experimental study of its efficacy in vitro. Liver cells were harvested from suckling pigs with collagenase perfusion in situ, and parenchymal and non-parenchymal hepatocytes were cocultured in a hollow fiber module which was rotated sporadically. Bioartificial liver(BAL) was developed using this bioreactor, and the BAL was perfused with ascites of patients suffering from liver cirrhosis. The yield of viable hepatocytes was (6.29 +/- 0.37) x 10(8) cells, and cell viability was greater than 84%. Hepatocytes aggregated to multi-cells spheroids after being rotated every thirty minutes for three hours. The hepatocytes in the bioreactor could synthesize urea. Total billirubin was decreased, and AST was significantly increased in the group of bioreactor, as compared with that in the control group. Glucose decreased in the group of bioreactor, whereas there was no significant descent in the control group; and the difference between the two groups was significant. The above results demonstrate that this bioreactor is effective for decreasing total bilirubin and glucose.

[Effects of deltamethrin on neurobehavioral development of offspring of intoxicated rats].

OBJECTIVE: To investigate the effects of deltamethrin on the filial brain nitric oxide synthase (NOS) activity and neurobehavioral development of the exposed lactational rats. METHODS: Pregnant rats were randomizedly divided into the treated group and the control group. The treated group was administered orally with 3.35, 6.70 mg/kg deltamethrin every other day from postnatal day (PND) 1 to PND 19 while the control group was administered with the corn oil of same amount in the same period. The activity of NOS of filial brain and neurobehavioral functions of the filial rats were observed. RESULTS: The lactational survival rate (81.80%:78.60%) in both treated groups was decreased significantly (P < 0.01) compared with that in the control group. The body weight of filial rats on PND 10, 21 in 6.70 mg/kg DM treated group [(16.62 +/- 2.2 8), (31.34 +/- 6.94) g] was less than those in the control group (P < 0.05). The delayed time in the filial rats in 6.70 mg/kg group was (3.05 +/- 1.20) s and the positive rates of passive escaping response in 3.35 and 6.70 mg/kg DM treated group were 22.5% and 21.5% respectively. There was the trend of the developmental increase of the activity of filial brain NOS between PND 5 and PND 21 and the NOS activity of rat brain on PND 5 in 6.70 mg/kg group [(0.60 +/- 0.07) U.mg pro(-1).h(-1)] was lower than that in the control group (P < 0.05). CONCLUSION: Exposure to high dose of deltamethrin in lactational female rats will decrease the activity of NOS of brain and retard the neurobehavioral development of their filial rats.

[Purification and viral inactivation of hemoglobin from human placenta blood].

This study was intended to establish a method for preparation of purified and viral-inactivated placenta hemoglobin. The optimum preparative condition resulted in the up-grading of purity, recovery and so on. A quality control was also established for the purification of hemoglobin. Compared to present purification methods, this method is easy to operate, needs low investment and running cost, and has the advantage of simultaneous operation for purification and viral-inactivation. The resulted hemoglobin had high purity and recovery, and the physicochemical property measured up to that in international reports. So this method is suitable for preparing purified and viral-inactivated hemoglobin on an adequate scale, and is useful for further development of blood substitutes.

[Improving the ways to isolate, cultivate and identify rat Kupffer cells].

OBJECTIVE: Kupffer cells (KCs) are resident macrophages in the liver. Because the densities and sizes of KCs show a significant overlap with other sinusoidal cells, it is difficult to separate and purify them. We aim to find an improved procedure that could optimize the method for isolation of highly purified rat Kupffer cells. METHODS: In ex vivo rat liver perfusion with pronase E and collagenase IV, density gradient centrifugation by Histodenz and selective attachment of Kupffer cells were used to isolate them. Cell proliferation and morphological characterization were studied under light phase-contrast microscopes; KCs were also studied with transmission electron microscopy and scanning electron microscopy using standard techniques. Immunocytochemistry was used to detect the expression of ED2 CD163 and lysosome associated membrane protein 2 (LAMP2). Phagocytosis of latex-beads by KCs was also studied. RESULTS: The yield rate of KCs was 5 x 10(7) and the KCs viability exceeded 98%. The purity of KCs identified by ED2 was higher than 98%, and over 99% of the collected KCs were LAMP2 positive. CONCLUSION: In the future this simple, stable and effective method of collecting highly purified Kupffer cells is expected to help in further studies.

[Role of arterial myo-endothelial gap junction on the vascular contractile reactivity following hemorrhagic shock in rats].

OBJECTIVE: To observe the role of arterial myo-endothelial gap junction (MEGJ) on endothelium-dependent and independent vascular contractile reactivity following hemorrhagic shock in rats. METHODS: Fifty-six SD rats were randomly divided into noradrenaline (NE) group and myricetin group. NE group was further divided into 5 subgroups: normal control group, 0, 0.5, 1 and 2 hours after hemorrhagic shock groups. Myricetin group was further divided into 7 subgroups: normal control group, 0, 0.5, 1, 2, 3 and 4 hours after hemorrhagic shock groups. Superior messenteric artery (SMA) was used to compare the changes in vascular responsing to NE or myricetin with or without 18alpha-glycyrrhetinic acid (18alpha-GA). RESULTS: Compared with normal control group, the reactivity of SMA rings to NE was significantly increased at 0 and 0.5 hour after shock and then decreased at 1 hour and 2 hours after shock. 18alpha-GA had no effect on SMA constriction induced by NE. The reactivity of SMA rings to myricetin was increased at 0, 0.5, 1 hour and 2 hours after shock, and it reached the peak at 2 hours, then it significantly decreased at 3 hours and 4 hours after shock. 18alpha-GA antagonized SMA constriction induced by myricetin. The antagonizing effect on myricetin in shock 1, 2 and 3 hours groups was more obvious than that in the other groups. CONCLUSION: At the early stage after hemorrhagic shock, the endothelium-dependent and independent vascular contractile response show a compensatory increase. The endothelium-independent contractile response is significantly decreased at 1 hour and 2 hours after shock, and the endothelium-dependent contractile response is significantly decreased at 3 hours and 4 hours after shock. MEGJ plays an important role in endothelium-dependent vascular contractile reactivity following hemorrhagic shock.

[Effects of expiratory triggering sensitivity on patient-ventilator expiratory synchrony and work of breathing in patients with chronic obstructive pulmonary disease during pressure support ventilation].

OBJECTIVE: To study the effects of expiratory triggering sensitivity (ETS) on patient-ventilator expiratory synchrony and work of breathing in chronic obstructive pulmonary disease (COPD) patients during pressure support ventilation (PSV). METHODS: A total of 31 COPD patients were ventilated in PSV mode, and measured by a pulmonary monitor. Meanwhile, the electromyogram of the diaphragm (EMG(diaph)) was obtained with electromyography. Five levels of ETS, 1%, 15%, 25%, 35%, and 50% of peak inspiratory flow (PIF), were studied in random order. Each ETS level lasted 30 minutes and all the data were recorded simultaneously for 3 minutes at the end of each period. The effects of ETS on patient-ventilator expiratory synchrony were analyzed by measuring the phase angle of expiration between the EMG(diaph) and the flow wave curve, and the effects of ETS on work of breathing by calculating total work of breathing (Wtot), work of inspiration by patients (Wi, P) and expiratory work of breathing (Wex). RESULTS: Ten patients were excluded from the study. At the 25% PIF level of ETS, patient-ventilator expiratory synchrony was the best, theta = (8 +/- 3) degrees, 16 patients - 15 degrees < or = theta < or = 15 degrees, and the amount of Wtot, Wi, p, Wex was the smallest among all the 5 levels of ETS, which was (1.86 +/- 0.53) J/L, (0.54 +/- 0.13) J/L, and (0.16 +/- 0.08) J/L respectively. When the level of ETS decreased, the occurrence of delayed termination of inspiration and the amount of Wex increased. At the level of 1% PIF, 18 patients theta > 15 degrees, and Wex was (0.48 +/- 0.10) J/L; at this level of ETS, Wi, p also increased significantly to (0.65 +/- 0.16 ) J/L. But when the level of ETS increased, the occurrence of premature termination of inspiration and the amount of Wi, p increased: at 50% PIF level of ETS, theta < - 15 degrees and Wi, p was (1.33 +/- 0.14) J/L in 19 patients. CONCLUSION: The proper adjustment of ETS during PSV improves patient-ventilator synchrony and decreases work of breathing in COPD patients.

[The advancement of AFLP technology].

AFLP technology has been widely used in molecular biology due to its integration of several advantages of high throughput, high efficiency and requiring no sequence information, etc. Great changes have been achieved in recent years in AFLP-related technologies and platforms. There are several AFLP-expanded technologies available. These improved technologies are capable of distinguishing the heterozygote from the homozygote and of converting any AFLP band of interest, without much effort, into locus-specific markers, which can be deployed for massive locus detection and for gene isolation. This review focuses on these favorable changes from conventional AFLP technology into more effective and more practicable AFLP-related ones. Understanding these advancements and AFLP-expanded technologies will facilitate the achievement of our research goals.

[Phase I study of postoperative concurrent chemoradiation with capecitabine as adjuvant treatment for stage II/III operable rectal cancer].

OBJECTIVE: This phase I study is to determine the maximal tolerated dose and the dose-limiting toxicity of capecitabine combined with standard radiotherapy (RT) as postoperative adjuvant treatment for rectal cancer patients. METHODS: Stage II/III rectal cancer patients 18 - 75 years of age had undergone curative surgery with Karnofsky score > or = 70% were eligible to be included in this study. Total dose of RT DT 50 Gy was delivered to the pelvic area in fraction of 2.0 Gy per day for 5 weeks. Capecitabine was orally administered concurrently with radiotherapy for a total of 2 cycles in escalating doses: twice daily at 12 hour interval for consecutive 14 days as one cycle, separated by a seven day rest, then followed by another cycle. From March 2004 to May 2005, 24 patients were included and treated at the following dose levels: daily 1000 mg/m(2) (3 patients), 1200 mg/m(2) (3 patients), 1400 mg/m(2) (3 patients), 1500 mg/m(2) (3 patients), 1600 mg/m(2) (6 patients), and 1700 mg/m(2) (6 patients). Dose-limiting toxicities (DLT) including grade 3 or grade 4 hematologic and nonhematologic toxicity were observed. RESULTS: Dose-limiting toxicity was observed in one patient treated at dose of 1600 mg/m(2) with grade 3 diarrhea, and in 2 patients at dose of 1700 mg/m(2) with one grade 3 and one grade 4 diarrhea. CONCLUSION: Diarrhea is the most common dose-limiting toxicity. In our study, the maximal tolerated dose (MTD) of capecitabine given concurrently with radiotherapy was daily 1600 mg/m(2), from D1 to D14 separated by 7-day rest for 2 cycles. Capecitabine given concurrently with standard radiotherapy is safe and tolerable for operated stage II/III rectal cancer patients.

[The serodiagnostic value of antigens secreted from Mycobacterium tuberculosis].

OBJECTIVE: To study the serodiagnostic value of 3 antigens secreted from Mycobacterium tuberculosis: the 81,000 antigen, the 38,000 antigen and the CF-10 antigen. METHODS: The 38,000 antigen was purified from supernatant of exponential growth of Mycobacterium tuberculosis. The 81,000 antigen and CF-10 antigen were expressed by fusion with 6 histidine and purified by nickel chelate chromatographic column. Sera were obtained from 149 patients with tuberculosis (108 smear positive and 41 smear negative), 24 patients with non-tuberculosis lung diseases and 170 healthy human controls in Anhui province from March 2002 to April 2005. Antigenicity and specificity of the 3 antigens were detected by enzyme-linked immunosorbent assay. RESULTS: Purified antigens had a high purity (> 95%) and a high specificity (> 98%). The positive rates of the 81,000 antigen, the 38,000 antigen and the CF-10 antigen in tuberculosis patients were 64.4% (96/149), 57.0% (85/149) and 37.6% (56/149) respectively. The negative rates in nontuberculosis patient were all above 98%. The positive rate of the 3 antigens combined was 81.2% (121/149) and the negative rate was 98%. CONCLUSIONS: The specificity of the 3 antigens was high. Sensitivity and specificity of the 3 antigens combined was better than each antigen alone. It is clinically practical that detection of multiple antigens may be used to improve the serodiagnosis of Mycobacterium tuberculosis.

[Design and implementation of DB sequence optimization software].

TIR (Translation Initiation Region) efficiency is very important in prokaryotic expression. The TIR's efficiency is highly dependent on SD (Shine-Dalgarno) sequence, distance between SD sequence and start codon, DB (Downstream Box) sequence, TIR's second structure, codon adaptation and so on. In this paper, we designed and implemented the software to optimize DB sequence and 5' rare codons. It generated some optimization sequences by analyzing the target sequence and comparing it with 16S RNA. And the optimization sequences is sorting by number of base pairing, location of base pairing and codon adaptation. We drew up the algorithm and the core of code in this paper.
