Establishment and evaluation of the system of extracorporeal liver perfusion in pigs.

BACKGROUND: In recent years, extracorporeal liver perfusion (ECLP) has been regarded as a treatment of acute liver failure (ALF); but the system of ECLP has many problems. The purpose of this experiment was to detect the factors affecting the system of ECLP and to establish a stable and effective system of ECLP. METHODS: Livers were harvested from health pigs, according to the different styles of perfusion and oxygenation, which were randomly divided into 3 groups. The livers in group A (n=4) were subjected to single portal vein perfusion, oxygenating perfusion blood; the livers in group B (n=4) to dual (portal vein and hepatic artery) vessel perfusion, oxygenating blood, together; and the livers in group C (n=4) to dual (portal vein and hepatic artery) vessel perfusion, but oxygenating blood, separately. The perfusion time, the data of bile production, and hemodynamic parameters of extracorporeal livers in each group were tested. The histological examination of liver tissues from each group was performed at the end of perfusion. RESULTS: The perfusion time of the liver in group A is significantly shorter than in groups B and C (P<0.05). At 1, 3, 6 hours after perfusion, the data of bile production and hemodynamic parameters of livers in group A were statistically different from those of livers in groups B and C (P<0.05). At 1, 3, 6 hours after perfusion, the data of group B were not statistically different from those of livers in group C (P>0.05). But at 12 hours after perfusion, the data of group B were statistically different from those of livers in group C (P<0.01). CONCLUSION: The system of ECLP, which is performed by dual (portal vein and hepatic artery) vessel perfusion and oxygenating blood separately, is more stable and effective to keep the function of extracorporeal liver.

[Transurethral prostatectomy with the bipolar plasma kinetic technique for benign prostate hyperplasia: a report of 297 cases].

OBJECTIVE: To evaluate the effect and safety of transurethral prostatectomy with the bipolar plasma kinetic technique (PKRP) in the treatment of benign prostate hyperplasia(BPH). METHODS: Two hundred and ninty-seven BPH patients underwent transurethral prostatectomy with the bipolar plasma kinetic technique. The preoperative estimated weight of the prostate ranged from 35 g to 102 g, averaging 52 g. RESULTS: The operation lasted 40 approximately 65 min, averaging 51 min. The resected tissues weighed 40 approximately 80 g, averaging 46 g. During the operation no transurethral resection (TUR) syndrome occurred. The catheter was removed 4 approximately 5 days after the operation, all with fluent urination. The patients were followed up for 2 approximately 33 months. IPSS decreased from average 31.5 preoperatively to average 6.8 postoperatively (P < 0.001). Average maximum flow-rate (Q(max)) decreased from 6.3 ml/s preoperatively to 18.6 ml/ s postoperatively (P < 0.001). Preoperative average residual urine was 97 ml and reduced to average 9 ml after the operation. Temporary incontinence occurred in 4 cases, perioperative hemorrhage in 2, and urethral stricture in 1. CONCLUSION: Transurethral prostatectomy with the bipolar plasma kinetic technique is a safe and effective means for the treatment of BPH.

[Biomechanical evaluation of the valgus stability of elbow after reconstruction].

OBJECTIVE: To evaluate of the valgus stability of the elbow after excision of the radial head, release of the medial collateral ligament (MCL), radial head replacement, and medial collateral ligament reconstruction. METHODS: Twelve fresh human cadaveric elbows were dissected to establish 7 kinds of specimens with elbow joint and ligaments as follow: (1) intact (n=12); (2) release of the medial collateral ligament (n=6); (3) excision of the radial head (n=6); (4) excision of the radial head together with release of the medial collateral ligament (n=12); (5) radial head replacement (n=6); (6) medial collateral ligament reconstruction (n=6); (7) radial head replacement together with medial collateral ligament reconstruction (n=12). Under two-newton-meter valgus torque, and at 0, 30, 60, 90 and 120 degrees of flexion with the forearm in supination, the valgus elbow laxity was quantified: All analysis was performed with SPSS 10.0 software. RESULTS: The least valgus laxity was seen in the intact state and its stability was the best. The laxity increased after resection of the radial head. The laxity was more after release of the medial collateral ligament than after resection of the radial head (P<0.01). The greatest laxity was observed after release of the medial collateral ligament together with resection of the radial head, so its stability was the worst. The laxity of the following implant of the radial head decreased. The laxity of the medial collateral ligament reconstruction was as much as that of the intact ligament (P>0.05). The laxity of the radial head replacement together with medial collateral ligament reconstruction became less. CONCLUSION: The results of this study show that the medial collateral ligament is the primary valgus stabilizer of the elbow and the radial head was a secondary constraint to resist valgus laxity. Both the medial collateral ligament reconstruction and the radial head replacement can restore the stability of elbow. If the radial head replacement can not be carried out, the reconstruction of the medial collateral ligament is acceptable.

[Capsid-targeted viral inactivation for dengue virus infection].

To explore the feasibility of capsid-targeted viral inactivation for dengue virus infection, a newly-discovered antiviral strategy, a mammalian cell line stably expressing staphylococcal nuclease fused to the capsid protein of dengue 2 virus was established and the effects on the production of infectious virus particles were examined. The results presented evidence that the enzymatically active staphylococcal nuclease fused to capsid protein could be incorporated into the nascent virions during wild virus assembly, resulting in degradation of viral genomic RNA and decrease in infectivity. Comparing the effects of incorporated SN and SN*, an enzymatically inactive missense mutant form of SN, on the infectivity of progeny virions, nucleolytic activity of incorporated SN was responsible for the major antiviral effects. These results paved the road of developing capsid-targeted viral inactivation as a new antiviral strategy against dengue.

[Design, production and clinical application of custom total hip prosthesis].

The key to the research of the hip prosthesis is to prolong the effective using time of the hip prosthesis, but bone loss and aseptic loosening are the influential factors in this aspect. Since each person has his own characteristic, it is difficult for the standard hip prosthesis to fit with the patient's femoral canal, so the stability can not the ensured. Moreover, some patients have congenital abnormality or a serious defect in bone or arthrosis, which makes an obvious deviation from the normal, therefore they can not use the standard hip prosthesis. CAD/CAM is useful for to individual three-dimensional femoral reconstruction, which can improve the stability of the prosthesis and effectively prevent the loosening of prosthesis. This article introduces the advantage, design, production and the problems of custom CAD/CAM hip prosthesis.

[Effects of aldose reductase on the expression of fibronectin and collagen IV in cultured rat renal mesangial cells].

OBJECTIVE: To study the effect of aldose reductase (AR) on expression of fibronectin and collagen IV in cultured rat renal mesangial cells (MsC). METHODS: AR expression plasmid vector (pCDNA3-AR) was constructed by restriction endonuclease digestion and ligation procedures. Stable expression of AR in MsC was established by Lipofectin transfection. Western blot and immunofluorescence analyses were performed to verify the transfection efficiency. Expression of fibronectin and collagen IV proteins were analyzed using Western blot. RESULTS: Expression of fibronectin and collagen IV in naive MsC treated with TGF-beta1 was upregulated in comparison to that of the untreated naive MsC (P < 0.01). MsC transfected with pCDNA3-AR showed a remarkable increase of expression of fibronectin and collagen IV (P < 0.01). Aldose reductase inhibitors (Sorbinil and Zopolrestat) significantly inhibited the expression of fibronectin and collagen IV in naive MsC (P < 0.05). CONCLUSIONS: Overexpression or inhibition of AR activity significantly alters the expression of fibronectin and collagen IV proteins in cultured rat MsC, suggesting that AR plays a significant role in the pathogenesis of glomerulosclersis.

[Expression and clinical significance of KISS-1 and GPR54 mRNA in endometrial carcinoma].

OBJECTIVE: To investigate the expression and clinical significance of KISS-1 mRNA and GPR54 mRNA in endometrial carcinoma. METHODS: The expression of KISS-1 mRNA and GPR54 mRNA in 32 patients with endometrial carcinoma, 10 patients with endometrial intraepithelial neoplasia (EIN) and 12 patients with normal endometrium was detected by reverse transcriptase polymerase chain reaction (RT-PCR). RESULTS: The positive rate of KISS-1 mRNA in endometrial carcinoma, EIN and normal endometrium was 37.5%, 80.0% and 83.3% respectively (endometrial carcinoma vs EIN or normal endometrium, P < 0.05). The expression of KISS-1mRNA in patients with endometrial carcinoma was correlated with its clinical stage, myometrial invasion and lymph node metastasis (P < 0.05). In endometrial carcinoma, the more advanced clinical stage, the lower expression of KISS-1 mRNA was detected. The positive rate of GPR54 mRNA in endometrial carcinoma, EIN and normal endometrium was 78.1%, 70.0% and 66.7% respectively, with no significant statistical difference (P > 0.05). It was not correlated with the clinical stage, histology grade, myometrial invasion or lymph node metastasis (P > 0.05). CONCLUSION: The interaction of KISS-1 and GPR54 may play an important role in inhibiting the invasion and metastasis of endometrial carcinoma.

[Association between insulin resistance and cholesteryl ester transfer protein gene polymorphism in type 2 diabetes mellitus].

OBJECTIVE: To study the association between cholesteryl ester transfer protein (CETP) gene polymorphism and insulin resistance in type 2 diabetes. METHODS: CETP-TaqIB gene was genotyped with polymerase chain reaction-restriction enzyme fragment polymorphism analysis in 108 patients with type 2 diabetes and in 60 normal controls. Plasma lipids, fasting insulin, insulin sensitivity index and insulin resistance index were determined in 108 patients with type 2 diabetes. RESULTS: The distribution of CETP-TaqIB genotypes and B1B2 allele frequency in the patients with type 2 diabetes were similar to that in general population. High density lipoprotein cholesterol (HDL-C), apolipoprotein A1(apoA1) and insulin sensitivity index (ISI) levels were significantly higher in B2B2 genotype than in B1B1 genotype. Fasting insulin (FINS) and Homeostasis model assessment-insulin resistance (HOMA-IR) levels were significantly lower in B2B2 genotype than in B1B1 genotype. No significant differences in triglyceride (TG), total cholesterol(TC),low density lipoprotein cholesterol (LDL-C), apolipoprotein B (apoB) levels were observed among different CETP-TaqIB genotype groups. By multivariate analysis, the determinants of ISI and HOMA-IR were body mass index (BMI), TC, HDL-C and CETP-TaqIB genotype. CONCLUSION: The CETP-TaqIB genotype is independently associated with insulin resistance and lipid metabolism. It may be an important risk factor of insulin resistance in type 2 diabetes.

[Expression and nuclease activity analysis of staphylococcus nuclease in E.coli].

AIM: To express staphylococcus nuclease (SN) in E.coli and prepare rabbit antisera against SN. METHODS: The SN gene was amplified by high-fidelity PCR from plasmid pPLC-SN and then subcloned into expression vector pLEX to obtain the recombinant plasmid pLEX-SN. The expression of recombinant protein was induced by tryptophan. The expressed SN was used to immunize a rabbit to prepare specific antibody. RESULTS: SDS-PAGE analysis showed that the relative molecular mass (M(r)) of the expressed SN was about 17,000 and the expressed SN accounted for about 37% of total bacterial proteins. The prepared antisera were specific to react with recombinant SN. CONCLUSION: Expression vector of SN has been successfully constructed and rabbit antibody against SN was prepared. These results lay the foundation for developing SN as antiviral protein.

[P53 expression and its clinical significance in prostatic carcinoma].

OBJECTIVE: To investigate the expression of P53 protein and its clinical significance in prostatic carcinoma. METHODS: Formalin-fixed, paraffin-embedded tissue sections from 45 cases of prostatic carcinoma (PCa) and 10 cases of benign prostate hyperplasia (BPH) were analyzed retrospectively with immunohistochemical Elivision staining method. The relationship of P53 expression with prostate cancer stage, grade, PSA, endocrine therapeutic effect and prognosis was evaluated. RESULTS: The positive staining rates of p53 protein expression were 51.1% and 10.0% respectively in patients with PCa and BPH (P < 0.05); 70.0% and 25.0% in PCa patients at pathological stage D and stages A approximately C respectively (P < 0.05); 14.3% and 56.7% in those with Gleason score < or = 7 and > 7 (P < 0.05); 20.0% and 60.0% in those with PSA < or = 10 microg/L and PSA > 10 micro/L (P > 0.05 ); 25.0% and 72.3% in those who responded to endocrine therapy and those who failed to respectively (P < 0.05). Log Rank analyses showed that the survival time of the PCa patients with negative P53 expression was obviously longer than those with the positive (P < 0.05 ). CONCLUSION: There were correlations between P53 expression and tumor grade, tumor stage and survival time, so the expression of P53 could be regarded as a prognostic molecular marker and a predictor of endocrine therapeutic effect for prostate cancer.

[Synthesis and spectral analysis of metal-N-glycoside complexes and studies of catalyzed ester hydrolytic kinetics].

Four new complexes of Ni(II), Cu(II), Zn(II) and Co(III) with HL (HL= N,N'-di-beta-D-glucosylethylenediamine) have been synthesized in methanol solutions. The compositions of the complexes determined by elementary analysis are [Ni(HL)(H2O)2]2+ Cl2.CH3OH.2H2O, [Cu(HL)]2+ Cl2.CH3CH2OH.3H2O, [Zn(HL)]2+ Cl2.H2O and [Co(HL)(H2O)(OH)]2+ Cl2.CH3OH.2H2O respectively. They were also characterized by IR, UV and NMR spectra which show that complexes of Ni(II) and Co(III) have octahedral configuration, while complexes of Cu(II) and Zn(II) have tetralhedral configuration. The hydrolytic kinetics of p-Nitrophenyl picolinate(PNPP) catalyzed by these complexes in buffer solution was also studied spectrophotometrically.

[Effects of aldose reductase transfection on the proliferation of rat mesangial cells in vitro].

OBJECTIVE: To study the effects of aldose reductase (AR) on the proliferation of rat mesangial cells (MsC) in vitro and to investigate its mechanism. METHODS: Cell proliferation was assessed by MTT colorimetric assay. Cell cycle and apoptosis were analyzed by flow cytometry. The growth of normal MsC and AR transfected MsC was compared. The proliferation of PDGF-BB and cellular growth stimulation by 10% NBS were investigated using AR inhibitors (ARI) Sorbinil and Zopolrestat. The effects of PDGF-BB on the expression of AR, p65 and c-Jun were assessed by Western blot. Activation of AP-1 was measured by EMSA. RESULTS: AR expression of transfected MsC was distinctly higher than that of the control. Transfected MsC grew quicker than normal cells. ARI partially inhibited the proliferation of transfected MsC under the stimulation of PDGF-BB and 10% NBS, whereas 10% NBS had no effect on normal MsC. PDGF-BB upregulated the expression of AR and c-Jun, but had no effect on p65. The upregulation of c-Jun and the activation of AP-1 could be attenuated by ARI. CONCLUSION: AR may participate in the pathological proliferation of MsC through the pathway related to the activation of AP-1.

[Expression and significance of KiSS-1 and its receptor GPR54 mRNA in epithelial ovarian cancer].

OBJECTIVE: To study the expression and significance of metastasis suppressor gene KiSS-1 and its receptor GPR54 in epithelial ovarian cancer. METHOD: The expression and their clinical significance of KiSS-1 and GPR54 were evaluated by RT-PCR in 37 patients with epithelial ovarian cancer, 15 patients with borderline epithelial ovarian tumors, 15 patients with epithelial benign tumors and 11 patients with normal ovarian tissues. RESULTS: The positivity and relative contents of KiSS-1 mRNA in patients with epithelial ovarian cancer (68%, 0.82 +/- 0.09) and borderline epithelial ovarian tumors (60%, 0.80 +/- 0.10) were all higher than in patients with epithelial benign tumor (20%, 0.65 +/- 0.10) and normal ovarian tissues (18%, 0.66 +/- 0.06; P < 0.05). The positivity and relative contents of KiSS-1 mRNA in patients with epithelial ovarian cancer correlated with their clinical stage and lymph node metastasis (P < 0.05). There was no difference in the positivity and relative contents of GPR54 mRNA between patients with epithelial ovarian cancer (70%, 0.79 +/- 0.07), borderline epithelial ovarian tumor (67%, 0.76 +/- 0.10), benign epithelial ovarian tumor (60%, 0.73 +/- 0.07) and normal ovarian tissues (45%, 0.78 +/- 0.08), respectively (all P > 0.05). The positivity and relative contents of GPR54 mRNA in patients with epithelial ovarian cancer did not correlate with their clinical stage, histology grade, lymph node metastasis and production of ascites (P > 0.05). CONCLUSION: KiSS-1 and GPR54 may play an important role in inhibiting the invasion and metastasis of early epithelial ovarian cancer.

[Effect of seawater soaking on corneal epidermis injury].

PURPOSE: Use slit lamp microscope and optical microscope to observe the changes in corneal tissue after corneal epidermis injury and the effect of seawater soaking on the healing of cornea after explosive injury. METHOD: Corneal epidermis were scraped on 20 eyes of 10 adult grey rabbits. For each rabbit, the right eye was in experiment and the left one was for comparison. After the injury, the experimental eyes were soaked in seawater for 30 minutes and the comparable eyes were soaked in 0.9% sodium chloride injection for the same time. All eyes were underwent slit lamp examinations, fluorescein dyeing, and ultrasonic cornea thickness metering on the corneas before the injury and 1, 3, 5, 7, 10 days after the injury respectively. At the same time, 2 rabbits were killed and the eyes were examined with pathology. The cornea thickness numbers were compared using self-comparison t test and grouping t test. Result after scraping the whole corneal epidermis, slit lamp microscope examination found that the corneas were edema and turbidity, the experimental eyes were stricter than the comparable eyes. The corneas of the experimental eye were vascularized 10 days after the injury. Fluorescein dyeing showed that more apparent pigmentation was found on the experimental eyes than that on the comparable ones. Comparative examination under the optical microscope showed that the corneal tissue of the experiment eyes changed distinctly, its inflammation is patency and healing was delayed and the vascularization degree became high after the injury. Cornea thickness of the experimental eyes became apparently thicker than that of the comparable eyes. The difference was statistically significant. CONCLUSION: Seawater soaking will have bad effects on the healing of the corneal epidermis injury.

Tumor imaging by means of proteolytic activation of cell-penetrating peptides.

We have devised and tested a new strategy for selectively delivering molecules to tumor cells. Cellular association of polyarginine-based, cell-penetrating peptides (CPPs) is effectively blocked when they are fused to an inhibitory domain made up of negatively charged residues. We call these fusions activatable CPPs (ACPPs) because cleavage of the linker between the polycationic and polyanionic domains, typically by a protease, releases the CPP portion and its attached cargo to bind to and enter cells. Association with cultured cells typically increases 10-fold or more upon linker cleavage. In mice xenografted with human tumor cells secreting matrix metalloproteinases 2 and 9, ACPPs bearing a far-red-fluorescent cargo show in vivo contrast ratios of 2-3 and a 3.1-fold increase in standard uptake value for tumors relative to contralateral normal tissue or control peptides with scrambled linkers. Ex vivo slices of freshly resected human squamous cell carcinomas give similar or better contrast ratios. Because CPPs are known to import a wide variety of nonoptical contrast and therapeutic agents, ACPPs offer a general strategy toward imaging and treating disease processes associated with linker-cleaving activities such as extracellular proteases.

High-performance affinity monolith chromatography: development and evaluation of human serum albumin columns.

Several immobilization methods were explored for the preparation of high-performance affinity monolithic columns containing human serum albumin (HSA). These monoliths were based on a copolymer of glycidyl methacrylate and ethylene dimethacrylate. In one method, the epoxy groups of this copolymer were used directly for the immobilization of HSA through its amine residues (i.e., the epoxy method); in other approaches, these epoxy groups were converted to diols for later use in the carbonyldiimidazole, disuccinimidyl carbonate, and Schiff base methods. Each HSA monolith was evaluated in terms of its total protein content and its retention of several model compounds, including (R/S)-warfarin and D/L-tryptophan. The greatest amount of immobilized HSA was obtained by the Schiff base method, whereas the epoxy method gave the lowest protein content. The Schiff base method also gave the best resolution in chiral separations of (R/S)-warfarin and D/L-tryptophan. All of the immobilization methods gave similar relative activities for HSA in its binding to (R)- and (S)-warfarin, but some differences were noted in the activity of the immobilized HSA for D- and L-tryptophan. The efficiency of these monoliths was found to be greater than that of silica-based HSA columns for (R/S)-warfarin (i.e., analytes with high retention), but little or no difference was seen for D- and L-tryptophan (analytes with weak retention).

Enhancement of the solubilization capacity of water in Triton X-100/cyclohexane/water system by compressed gases.

The effect of compressed CO2 and ethylene on the properties of Triton X-100/cyclohexane/water systems was studied at different temperatures and pressures. Surprisingly, it was discovered that the compressed gases had the functions of co-surfactants. At suitable pressures, the water-to-surfactant molar ratio (W0) was enhanced significantly by the dissolution of the gas in the solution. The microenvironment in the reverse micelles was investigated by UV-visible spectroscopy by using methyl orange (MO) as a probe. The influence of n-hexane, Na2CO3, NaHCO3, H2C2O4, and CaCl2 at various concentrations on the solubilization of water in the absence of compressed gases was also investigated in order to obtain some information about the mechanism of the interesting phenomenon. This new route to stabilize reverse micelles may have potential applications to other similar systems. Moreover, the results of this work provide some useful information to get insight into the mechanism of co-surfactants, because a conventional co-surfactant usually contains both polar group and hydrocarbon chain, and it is very difficult to clarify the functions these two groups, while the gases used in this work are small nonpolar molecules, which solely have the function of the hydrocarbon chain in a co-surfactant.

[Expression and significance of caspase-3 gene in apoptotic muscle cells 103Pd radioactive stent bile duct in dogs].

OBJECTIVE: To discuss the expression and significance of caspase-3 gene in the apoptotic muscle cells in gamma-radiation-induced muscle cell lines. METHODS: The caspase-3 mRNA in the control and gamma-radiation induced apoptotic muscle cells was analysed by RT-PCR. RESULTS: The expression of caspase-3 gene transcript was higher in 103Pd radioactive stent dog bile duct than in general stent dog bile duct, and apoptotic muscle cells were higher in 103Pd radioactive stent dog bile duct than in general stent dog bile duct. CONCLUSIONS: The high level expression of caspase-3 gene may help to understand the muscle cells sensitivity to gamma-radiation apoptosis. 103Pd radioactive stent may increase the expression of caspase-3 gene in dog bile duct and prevent the billiary narrow when dog bile duct was injured by balloon.
