Oral immunization with attenuated vaccine strains of Vibrio cholerae expressing a dodecapeptide repeat of the serine-rich Entamoeba histolytica protein fused to the cholera toxin B subunit induces systemic and mucosal antiamebic and anti-V. cholerae antibody responses in mice.

Entamoeba histolytica is a significant cause of morbidity and mortality worldwide. The serine-rich E. histolytica protein (SREHP) is a surface-expressed trophozoite protein that includes multiple hydrophilic tandem repeats. A purified fusion protein between the dodecapeptide repeat of SREHP and cholera toxin B subunit (CTB) has previously been shown to be immunogenic in mice after oral inoculation when cholera toxin is coadministered as an immunoadjuvant. We engineered a live attenuated El Tor Vibrio cholerae vaccine strain, Peru2, to express the SREHP-12-CTB fusion protein to the supernatant from either a plasmid [Peru2 (pETR5.1)] or from a chromosomal insertion (ETR3). Vector strains were administered orally to germfree mice that were subsequently housed under nongermfree conditions; mice received one (day 0) or two (days 0 and 14) inoculations. No immunoadjuvant or cholera holotoxin was administered. Mice that received two inoculations of Peru2(pETR5.1) had the most pronounced antiamebic systemic and mucosal immunologic responses. Less marked, but significant, anti-SREHP serum immunoglobulin G antibody responses were also induced in mice that received either one or two oral inoculations of strain ETR3. Anti-V. cholerae responses were also induced, as measured by the induction of serum vibriocidal antibodies and by serum and mucosal anti-CTB antibody responses. These results suggest that V. cholerae vector strains can be successful delivery vehicles for the SREHP-12-CTB fusion protein, to induce mucosal and systemic antiamebic and anti-V. cholerae immune responses. The magnitude of these responses is proportional to the amount of SREHP-12-CTB produced by the vector strain.

Interleukin-12 (IL-12) and IL-18 synergistically induce the fungicidal activity of murine peritoneal exudate cells against Cryptococcus neoformans through production of gamma interferon by natural killer cells.

We examined the ability of interleukin-12 (IL-12) and IL-18 to induce the production of gamma interferon (IFN-gamma) and nitric oxide (NO) by murine peritoneal exudate cells (PEC) and to stimulate the growth-inhibitory activity of these cells against Cryptococcus neoformans. PEC produced IFN-gamma and NO when stimulated with a combination of IL-12 and IL-18 but little or no IFN-gamma or NO when either cytokine was used alone. PEC anticryptococcal activity was mediated by IFN-gamma and NO production, since it was completely inhibited by a neutralizing anti-IFN-gamma monoclonal antibody (MAb) and N(G)-monomethyl-L-arginine, a competitive inhibitor of NO synthesis, respectively. To identify the IFN-gamma-producing cells among PEC stimulated with IL-12 and IL-18, we depleted NK cells, gammadelta T cells, or CD4+ T cells by treating PEC with specific Abs and complement. NK cell depletion strongly suppressed IFN-gamma production and almost completely inhibited NO production and anticryptococcal activity, while depletion of other cells had no such influence. Alternatively, purified NK cells by two cycles of glass adherence and magnetic separation with anti-CD3, -CD4, -CD8, and -B220 MAbs produced a greater amount of IFN-gamma by stimulation with IL-12 and IL-18 than unseparated non-glass-adherent PEC. Our results demonstrated that IL-12 and IL-18 synergistically induced NO-dependent anticryptococcal activity of PEC by stimulating NK cells to produce IFN-gamma.

Neutrophils play a critical role in early resistance to amebic liver abscesses in severe combined immunodeficient mice.

Animal models of liver abscess formation with Entamoeba histolytica suggest that the neutrophil is the first cell of the host immune system to interact with the invading ameba. In vitro studies have suggested that lysis of neutrophils by virulent amebae may exacerbate the damage seen in amebic liver abscesses. To investigate the role of neutrophils in vivo, we used the severe combined immunodeficient (SCID) mouse model of amebic liver abscess formation and compared liver damage in neutrophil-depleted and control mice. We found that neutrophil-depleted animals have significantly larger amebic liver abscesses at early stages of infection and that abscesses in neutrophil-depleted SCID mice lack the prominent inflammatory cell ring seen in amebic liver abscesses in control SCID mice. These data suggest that neutrophils play a protective role in the early host response to amebic infection of the liver.

Somatosensory influences on renal sympathetic nerve activity in anesthetized Wistar and hypertensive rats.

This study compared the cardiovascular and renal nerve activity responses to somatosensory stimulation with capsaicin in normotensive and hypertensive rats. The importance of the cardiopulmonary receptors in these two states was examined with the use of phenylbiguanide (PBG) infusion. Subcutaneous capsaicin increased blood pressure (BP), heart rate (HR), and renal nerve activity (RNA) 6-35% (P < 0.01), and total power (TP) and %power at HR (%PHR) rose two- to threefold (P < 0.001). PBG reduced basal RNA, TP, and %PHR (20-70%, P < 0.05). PBG did not change the cardiovascular, but attenuated the TP and %PHR increases due to capsaicin (P < 0.001-0.01). PBG given to vagotomized normotensive rats normalized the cardiovascular and RNA responses to capsaicin. In hypertensive rats, capsaicin increased BP, HR, RNA(10-20%), TP, and %PHR (50-70%, P < 0.001). PBG infusion into hypertensive rats decreased RNA (20%, P < 0.01) and the capsaicin-dependent rise in RNA was smaller (P < 0.05). TP and %PHR were unchanged, except in vagotomized hypertensive rats given PBG, in which these responses were minimally affected. Somatosensory modulation of RNA power spectra was suppressed by the cardiopulmonary receptors in normotensive rats, but in hypertensive rats their impact was much smaller.

Neural regulation of kidney function by the somatosensory system in normotensive and hypertensive rats.

This investigation examined the renal sympathetic nerve and renal excretory responses to somatosensory stimulation in normotensive and stroke-prone spontaneously hypertensive rats (SHRSP). Somatosensory activation was achieved by either subcutaneous capsaicin administration or exposure of the airways tract to irritant fumes from acetic acid in chloralose-urethan-anesthetized animals. In Wistar rats, blood pressure increased between 10 and 20% (P < 0.001-0.01), renal perfusion pressure was maintained unchanged, renal hemodynamics were unaltered, and urine flow and sodium excretion were decreased by 25 to 50% (P < 0.001-0.05). In the SHRSP, the somatosensory-induced increases in blood pressure were slightly larger (approximately 15-20% P < 0.05) than those of the Wistar rats, whereas the excretory responses were one-half those of the normotensive animals (P < 0.05). The somatosensory challenges reflexly increased integrated renal sympathetic nerve activity in both normotensive and hypertensive rats. The power spectral analysis demonstrated that the increases in percentage power at heart rate frequency and total power were two to three times more (P < 0.05) in the Wistar rats compared with the SHRSP. The reduced ability of the SHRSP to modulate the energy in the renal sympathetic nerve signal at heart rate frequency might explain in part the attenuated functional responses to the somatosensory challenges.

Kinetics of the thermal inactivation of alkaline phosphatase from green crab (Scylla serrata).

The kinetics of thermal inactivation of alkaline phosphatase from green crab (Scylla Serrata) has been studied using the kinetic method relating to the substrate reaction during irreversible inhibition of enzyme activity previously described by Tsou. The results show that the thermal inactivation of the enzyme is an irreversible reaction. Comparison of the microscopic rate constants for thermal inactivation of free enzyme and the enzyme-substrate complex shows that the presence of substrate has a certain protective effect against thermal inactivation.

[Outward character and inner quality of Bombyx Batryticatus, Carapax Trionycis, Squama Manitis and endothelium corneum Gigeriae Galli after frying].

According to the requirement for outward character in processing traditional Chinese medicines, Bombyx Batryticatus, Carapax Trionycis, Squama Manitis and Endothelium Corneum Gigeriae Galli, which are four kinds of often used traditional Chinese animal medicines, were fried, the contents of amino acids in each kind were determined, and investigate the inner quality of each kind of the processed product was investigated with reference to other standards.

[Synthesis of S-9-(2,3-dihydroxypropyl)adenine (DHPA) analogs and their inhibition of S-adenosyl-L-homocysteine (SAH) hydrolase].

In order to search for new antiviral agents with high potency and low toxicity, eleven new acyclonucleosides were synthesized. Nucleic-acid bases were condensed with 3-chloro-2-methylpropene to give 5-8a. b, which were oxidized by N-methylmorpholine-N-oxide in the presence of OsO4 to give vicinal dihydroxy acyclonucleosides 1-4a.b. Four DHPA analogs have been tested for the inhibitory activities on s-adenosyl-L-homocysteine hydrolase (SAH). Only compound 1 showed some enzyme inhibitory effect with IC50 of 1.1 mmol.L-1.

[Effect of wind tunnel noise on left ventricular function of heart].

A two-dimensional color doppler ultrasonic imaging instrument was used to detect the left ventricular function in 36 wind tunnel technicians before and after their work in the wind tunnel. The results showed that under the noise between 91.5 and 101.5 dB(A) inside the wind tunnel SV, EF, Fs, MVCF, PE, SE and EAC decreased while SA, PA/PE increased. It indicates that high level wind tunnel noise has certain harmful effect on the left ventricular function of wind tunnel technicians.

Gravitation model for the magnitude of migration and its application.

"The magnitude of migration is one part of the feasibility study on migration, its result being critical to the yield of positive or negative effect.... The key to the success of migration is that the magnitude of migration must be limited within the capacity of the place of destination with generous room for flexibility. It is therefore necessary to determine the appropriate magnitude by scientific methods." The geographical focus is on China.
