Antitumor activities of human recombinant interferon-gamma against cervical tumor transplanted into nude mice.

This study examines the antitumor effects of recombinant interferon-gamma (ReIFN-gamma), by comparing various routes for its administration in nude mice transplanted with human cervical tumor cells. Daily intratumoral injection of ReIFN-gamma showed the best inhibitory effect, although intramuscular daily administration of this IFN also showed a significant antitumor effect. Furthermore, the earlier treatment was initiated after tumor inoculation, the better was the antitumor effect of the IFN. Histologic examination of the transplanted tumors after treatment with ReIFN-gamma showed a sparse or reticular arrangement of the tumor cells, and very large multinucleated giant cells. Unlike the original tumor, 'pearl' formation and isolated cell keratinization were frequently observed in the transplanted tumors obtained from IFN-treated groups.

Theca lutein cysts with maternal virilization and elevated serum testosterone in pregnancy.

A clinicopathological study was performed on a pregnant patient with bilateral multiple ovarian cysts, who presented with pre-eclampsia, marked virilization and exceedingly high levels of serum testosterone. The female fetus showed no evidence of virilization. Microscopic examination of the ovaries revealed theca lutein cysts with granulosa cells which showed the existence of testosterone by immunoperoxidase staining.

Successful treatment with bromocriptine of hypertension associated with polycystic ovarian disease.

The use of bromocriptine, 5 mg per day, markedly improved the severe hypertension (up to 180/120 mmHg) of a patient suffering from polycystic ovarian disease (PCOD). The response was rapid, with decreasing concentrations of serum testosterone and 11-deoxycorticosterone (DOC). Bromocriptine induced further renewal of the ovulatory cycle. Bromocriptine may thus be of considerable value in the treatment hypertensive patients with PCOD.

Effects of highly purified eicosapentaenoic acid on plasma beta thromboglobulin level and vascular reactivity to angiotensin II.

Eicosapentaenoic acid (EPA), the precursor fatty acid of three series of prostaglandins, has been reported to have an antiatherothrombotic potential. We gave highly purified EPA in a soft capsule (90% ethylester form of EPA; EPA-E) to 6 healthy male volunteers for 4 weeks. After the administration of 900 mg/day of EPA-E for two weeks or longer, a significant reduction in plasma beta thromboglobulin level was observed, and after 4 weeks' administration, significantly blunted pressor responsiveness to infused angiotensin II was observed. These changes were not observed 4 weeks after EPA-E had been discontinued. After 4 weeks' ingestion of EPA-E, the platelet count, mean platelet volume, platelet aggregation with adenosine diphosphate or collagen, plasma recalcification time, prothrombin time, plasma antithrombin III, fibrinogen or plasmin concentrations, serum concentrations of total cholesterol, triglycerides, total phospholipid, nonesterified fatty acid or high-density lipoprotein cholesterol were unchanged. From the data presented it can be said that EPA-E causes a mild depression of vascular contractility and of platelet aggregability in vivo and exerts a beneficial influence on several cardiovascular factors.

Regulation of IgE receptor expression on human peripheral blood lymphocytes by lymphocytosis promoting factor (LPF), lectins and dexamethasone.

Using a monoclonal anti-human Fc epsilon R antibody (H107), we found that lymphocytosis promoting factor (LPF), phytohaemagglutinin (PHA-P) and Concanavalin A (Con A) could induce Fc epsilon R, detected by immunofluorescence study, on normal human peripheral blood lymphocytes without IgE. The number of Fc epsilon R bearing lymphocytes was increased by stimulation with 3, 10 and 10 micrograms/ml of LPF, PHA-P and Con A, respectively, from 6.0 +/- 3.0/1000 cells to 26.0 +/- 7.9, 54.0 +/- 6.7 and 24.8 +/- 7.1/1000 cells, respectively. Although the induction of Fc epsilon R occurred neither in the separated T-enriched fraction (TEF) nor the T-depleted fraction (TDF), it recovered when the two fractions were mixed. The cell free supernatants from TEF stimulated with LPF or PHA-P could increase Fc epsilon R(+) cells in TDF, whereas those from TDF failed to increase them in TEF. The results suggest that the induction of Fc epsilon R occurs mainly on B lymphocytes by the soluble factor(s) formed by T cells stimulated with LPF or PHA-P. The induction of Fc epsilon R by stimulants was completely inhibited by 10(-6) M dexamethasone. It was demonstrated that the effects of dexamethasone on lymphocytes were dual: one was on B cells to inhibit responsive increases of Fc epsilon R, and the other was on T cells to suppress the formation of the soluble factor(s) which induced Fc epsilon R on B cells.

[A case of breast cancer with pleural metastasis responding to administration of intrapleural adriamycin and systemic chemotherapy with FEMP].

A 51-year-old woman with breast cancer metastatic to the pleura was treated with intrapleural instillation of adriamycin after thoracentesis, and systemic administration of FEMP, with complete response for more than 11 months. She underwent radical mastectomy and bilateral oophorectomy for advanced breast cancer, and 18 months later presented radiological evidence of pleural fluid contralateral to the affected breast. Initially, thoracentesis for symptomatic relief was done with a positive cytology in the pleural effusion. Intrapleural instillation of adriamycin was followed by partial resolution, and administration of FEMP achieved complete disappearance of both pleural metastasis and pleural effusion.

Scanning electron microscopic study on the angioarchitecture of glomeruli in experimental toxemia of pregnancy in the rabbit.

Experimental toxemia of pregnancy in late gestation in rabbits was induced by constricting the aorta below the renal arteries on day 21 of gestation. Light and transmission electron microscopic examination of glomeruli on day 29 revealed only a slight swelling of endothelial cells. The angioarchitecture was also investigated using scanning electron microscopic examination of methyl methacry corrosion casts of the glomeruli. The kidney in experimental toxemia was characterized by a reduction of glomerular vessel diameter compared to that of normal pregnant rabbits. This probably resulted from enlargement of the endothelial cells, suggesting a reduced blood supply to the glomerulus. These findings suggest that interference with the blood supply to the pregnant uterus is an important factor in the development of the renal lesion of experimental toxemia of pregnancy in the rabbit.

[A population survey on mass screening program for gastric cancer. Part 2. Why don't they receive mass screening examination?].

The attitude of non-examined residents to a mass screening program for gastric cancer has been surveyed by a self-administered questionnaire among 3660 residents in a small town near Kitakyushu City. Among 1159 subjects who had not received any type of examination during the past 3 years, the answers of 638 subjects who completed the questionnaire were analyzed in this study. Reasons for not being examined were classified into 4 types; "unnecessary", "rejected", "inconvenient" and "others". The most frequent reason for not taking part in the screening program was "inconvenient" which means that there were some difficulties for them to participate in the program even if they wanted to. This reason was followed by "unnecessary" and "rejected". "Inconvenient" was the main reason for not being examined among people in their forties and fifties and/or among those who have jobs. Among elderly people, "unnecessary" had the highest frequency. "Rejected" was the least important reason in all age groups. In each age group "rejected" was found more frequently among females than males. As the type of reason for not being examined was different among groups, it is essential to work out different countermeasures for each group in order to raise the participating rate.

A high molecular weight protease in the cytosol of rat liver. II. Properties of the purified enzyme.

The properties of a soluble endoprotease from rat liver were studied. The enzyme was purified in a latent form. It sedimented as a single component with a sedimentation coefficient (S(0)20,w) of 19.8 S. Measurement by quasi-elastic light scattering gave a diffusion coefficient (D(0)20,w) of 2.5 X 10(-7) cm2 X s-1 and an effective hydrodynamic radius of 85 A. The enzyme had an unusually high molecular weight, estimated as 743,000 by sedimentation equilibrium and 722,000 by sedimentation velocity and diffusion measurements and as 760,000 by a recently developed low-angle laser light scattering method. Judging from electron microscopic observation and the calculated frictional and axial ratios, the enzyme molecule is disc-shaped. Analysis of the far-ultraviolet circular dichroic spectrum showed that the enzyme contains 50% alpha-helical, 25% beta-sheet, and 15% unordered structures with 10% beta-turns. The isoelectric point of the enzyme is 5.0. These properties indicate that the purified enzyme is a homogeneous molecule. In addition, the enzyme is a simple protein since it contains no measurable amounts of nucleic acid carbohydrate or lipid.

Chemoattractant efficacy: oxidation of stimulus by responding cells.

Although all human neutrophils have receptors for formyl-methionyl-leucyl-phenylalanine, only 20-30% migrate in vitro at optimal attractant concentrations. Since this attractant can be oxidized by myeloperoxidase from stimulated neutrophils, we determined if its efficacy was increased by protection from oxidation. Efficacy was increased by reducing agents and by molecules with oxidizable sulfur, 10(-5) M methionine or 1.5 X 10(-6) M albumin. The antioxidant effect was on the attractant, not the cells: albumin in the cell compartment did not increase responses. Furthermore, antioxidants had no effect on efficacy of fNle-Leu-Phe-Nle-Tyr-Lys, an attractant that also stimulated superoxide release but had no oxidizable sulfur.

[A population survey on a mass screening program for gastric cancer. 1. Population characteristics related to examination rates for the screening program].

The types of examination received for gastric cancer during the past 3 years have been surveyed by a self-administered questionnaire among residents of a small town near Kitakyushu City. Among 3660 surveyed, 2205 subjects who completed the questionnaire are analyzed in this study. Examination rates of mass screening for males were higher than those for females. Males in their forties and fifties and females in their forties through sixties have higher examination rates than other age groups. Higher examination rates were seen in public officials and employees who have opportunities to receive the screening program at work site. On the other hand, among people without a job and housewives who have opportunities to receive a screening program offered only by the town hall, examination rates of the mass screening program were low. More people with a family history of cancer death were examined than those without a similar family history. There was no difference in the examination rates by the mass screening program between the people who took care of their health and those who did not. In order to raise examination rates for the mass screening program, it is essential to improve the systems of this program which can be easily accepted, and to educate residents in order to stimulate motivation for screening.

[Analyses of industrial society from ecology--modeling analyses of industrial ecological system].

A macroscopic industrial ecological model has been developed to investigate the effects of changes in industrial structure on social well-being in general and health in particular. The model is composed of the following sectors: industry and employment, environment and pollution, and population and health care. The model was calibrated using available data of the past 60 years for Japan. The simulation study was conducted with the time span of 1920 to 2000. The results indicated that the gradual shift, in the industrial structure of Japan, would improve the level of living standards resulting in enhanced health service capability and lower death rate although increasing adult disease rates in the aged society. Also the rapid future growths of service industry and information and communication were observed.

[A study of utilization review in hospitals--focusing on medical statistics].

It is a matter of course that health care is an important issue closely related to the daily life of residents. In this situation, university hospitals, although their main aim is research and education, also play a significant role as core hospitals in various regions of the community. By reviewing hospital utilization, extended to facilities in a given area, we first analyzed the medical service area and the length of stay in the hospital affiliated to the University of Occupational and Environmental Health, on the basis of discharge various directions of facility planning based on future bed using control, including addition of beds, and also attempted to evaluate the actual state of annual facility management with the aid of computer.

Characterization of a vasopressor substance generated in human plasma by incubation.

Incubation of human plasma at 37 degrees C for several hours leads to the formation of a non-dialysable vasopressor substance termed the active pressor principle. Some of the chemical and physical natures of active pressor principle were investigated in anesthetized and ganglion blocked rats. It was found to have properties characteristic of protein. The substance was crudely purified to about 25-fold in alcoholic trichloroacetic acid solution after placing the plasma in a boiling water bath for 5 minutes ("active fraction"). After treatment of the vasoactive plasma or "active fraction" with Pronase, the pressor activity was almost abolished. The molecular weight of this fraction as determined by gel filtration was about 68,000. With addition of diisopropyl fluorophosphate before incubation of the plasma, no vasopressor substance was generated. After treatment of the rat with captopril, an angiotensin converting enzyme inhibitor, the pressor effect of incubated plasma was not inhibited. These findings suggest that a vasoactive protein, which is clearly different from renin, is generated during simple incubation of plasma, and that a serine protease is involved in the formation of this substance.
