Probucol decreases total body fat loss in VX2-carcinoma-induced cachectic rabbits.

We previously reported the continuous decrease of total body fat in VX2-carcinoma-bearing rabbits after tumor implantation, as well as changes in the serum lipid profile. Probucol, an antioxidant drug, has a cholesterol lowering effect against hyperlipidemic subjects. VX2-carcinoma-bearing rabbits fed with a diet containing 1% probucol did not show any difference in serum lipid compositions as compared with rabbits fed with a control diet. Similarly serum lipolytic activity showed no differences, whether probucol was administered or not, while the decrease in total body fat was significantly less when probucol was administered.

Balloon-occluded arterial infusion chemotherapy, simple total hysterectomy, and radiotherapy as a useful combination-therapy for advanced cancer of the uterine cervix.

We first performed 3 or 4 courses of balloon-occluded arterial infusion (BOAI) chemotherapy in 19 patients with advanced cancer of uterine cervix classified as stage III-IV according to the International Federation of Gynecology and Obstetrics. In the patients in whom the first BOAI resulted in insufficient drug accumulation in tumor tissue, the ovarian arteries and veins were ligated to improve the therapeutic effect. Ultimately simple total hysterectomy and radiotherapy were performed in 9 patients in whom the uterus was resectable (the combination group), and radiotherapy alone was conducted in the other 10 patients in whom hysterectomy was not practicable (the incomplete group). The survival rates were significantly higher in the combination group (2-year survival 88. 9%; 5-year survival 66.7%) than in the incomplete group (p=0.0013).

Effects of cyclic plasma perfusion on adipocyte apoptosis and lipolytic activity in VX2 carcinoma-bearing rabbits.

BACKGROUND: In an animal model we found that total body fat (TBF) decreased with progression of tumors, but that it rebounded after cyclic plasma perfusion (CPP). In this study, we investigated changes in adipocyte apoptosis and serum lipolytic activity (LA) after CPP. MATERIALS AND METHODS: TBF, LA, and adipocyte apoptotic index (AAI) in VX2 carcinoma-bearing rabbits were measured every ten days after implantation. CPP was performed every 5 days from post-implantation day (PID) 14. RESULTS: TBF decreased as the days passed and reached 43.2 +/- 10.0% on PID 50 in the non-CPP group, while it decreased 22.7 +/- 8.8% in the CPP group. LA on PID 50 was 17.8 +/- 14.0, 33.2 +/- 6.3 mumol glycerol/10(5) adipocytes in the non-CPP group, while LA did not increase in the CPP group. AAI was 28.8 +/- 16.4, 27.8 +/- 17.7, 10.3 +/- 9.9 and 0.4 +/- 1.1 on PID 10, 20, 30, and 40, respectively, in the non-CPP group. No change was noted in AAI after CPP. CONCLUSIONS: CPP removed a humoral factor that possesses TBF-decreasing LA, but it did not suppress adipocyte apoptosis.

Histochemical analysis of glycoconjugates in the eccrine glands of the raccoon digital pads.

The distribution and selectivity of complex carbohydrates in the eccrine glands of the digital pads in the North American raccoon (Procyon lotor) were studied using light and electron microscopic histochemical methods, particularly lectin histochemistry. In the eccrine glands, the dark cells exhibited neutral and acidic glycoconjugates with different saccharide residues (alpha-L-fucose, beta-D-galactose, beta-N-acetyl-D-glucosamine and N-acetyl-neuraminic acid); the clear cells contained numerous glycogen particles and showed a distinct reaction of alpha-L-fucose. The presence of complex carbohydrates with various terminal sugars was evident in the excretory duct cells. In addition, beta-D-galactose and N-acetyl-neuraminic acid residues were mainly observed in the luminal secretion. The glycoconjugates produced by the eccrine glands of the raccoon digital pads may protect the epidermis against physical damage or microbial contamination. In this way, the normal functioning of the sensory apparatus of the foot pads is ensured.

Morphological, histochemical and immunohistochemical characterization of secretory production of the ciliary glands in the porcine eyelid.

In addition to performing general histology and cytology of the ciliary glands of the miniature pig, we studied the localization of glycoconjugates and beta-defensins in these glands with the use of carbohydrate histochemical and immunohistochemical methods. The secretory cells of the glands were equipped with non-homogeneous secretory granules, a well-developed Golgi apparatus and rough endoplasmic reticulum. The secretory epithelium and luminal secretion of the glands contained large amounts of neutral and acidic glycoconjugates with various saccharide residues (alpha-L-Fuc, beta-D-Gal, alpha-D-GalNAc and sialic acid). The sebaceous glands and tarsal glands also exhibited positive reactions to most of the histochemical methods. Additionally, the antimicrobial peptide group of beta-defensins was demonstrated to be products of the ciliary glands, as well as the sebaceous glands and tarsal glands. The results obtained are discussed with regard to the specific function of the ciliary glandular secretions. These secretory products may be related to the moistening and general protection of the skin surface of the eyelid and ocular surface.

Effects of androgen, fibroblast growth factors or other various growth factors on growth of Shionogi carcinoma cells in a protein-free medium.

Shionogi carcinoma 115 (SC115) has been accepted for 20 years as an androgen-responsive mouse mammary tumor. We have recently established an androgen-dependent cloned cell line (SC-3) from a SC115 tumor. We found in the present study that the growth of SC-3 cells can be stimulated by 10(-8) M testosterone (up to 100-fold) even in a protein-free medium beginning from plating [Ham's F-12: Eagle's minimum essential medium (1:1, v/v)]. In the protein-free culture, the proliferation of SC-3 cells was also found to be stimulated by acidic or basic fibroblast growth factor (FGF) alone (up to 50-fold) among various growth factors examined such as FGFs, insulin, insulin-like growth factor (IGF)-I, IGF-II, nerve growth factor, platelet-derived growth factor, epidermal growth factor, transforming growth factor (TGF)-alpha and TGF-beta. The testosterone (10(-8) M)- or FGF (10 ng/ml)-induced growth of SC-3 cells was abolished only by TGF-beta (greater than or equal to 1 ng/ml) among various growth factors examined. We show for the first time in this study an androgen-dependent growth of cancer cells (SC-3) in a protein-free medium. In the protein-free medium, growth of SC-3 cells is also stimulated by FGFs, and the androgen- or FGF-induced growth is inhibited only by TGF-beta.

MR imaging: possibility of tissue characterization of brain tumors using T1 and T2 values.

To evaluate the usefulness of T1 and T2 values for tissue characterization of brain tumors, 37 histologically confirmed brain tumors were examined with a 0.5-T superconductive MR system. With spin-echo and inversion-recovery imaging sequences, computed T1 and T2 images were reconstructed, and T1 and T2 values of the tumors were calculated. Relaxation rates (1/T1 and 1/T2), T1/T2 ratios, and malignancy indexes, which were originally designed for gastrointestinal tumors, were also calculated. Values of all these parameters were so wide-ranged that it was impossible to characterize the tumor tissue types.

MR imaging of blood flows in the cavernous sinus.

To evaluate blood flows in the cavernous sinus (CS), seven normal CSs and a postmortem CS were evaluated using T1-weighted spin echo sequences. After the noncontrast study, contrast study with Gd-DTPA was performed in all cases except the postmortem one. In the normal CSs, arterial flows of the intracavernous carotid artery (ICA) were disclosed as no-signal areas in the noncontrast study and were not enhanced with Gd-DTPA. Most venous flows in the CS were seen as low-intensity areas in the noncontrast study and were markedly enhanced with Gd-DTPA. However, these low-intensity areas were heterogeneous in intensity and several were higher and lower in intensity compared with most venous spaces. In the postmortem case, the CS showed homogeneous low intensity due to the stasis of blood flow. It was thought that the heterogeneous appearance of venous spaces in the normal CSs was due to flow-related phenomena. This heterogeneous pattern of signal intensity suggested the distribution of flow velocities. MR demonstrated both arterial and venous flows, and this was facilitated by Gd-DTPA. MR is a promising modality for the demonstration of blood flows in the CS.
