Monoclonal antibody to the streptococcal preparation OK-432: tissue OK-432 localization and analysis of interaction between OK-432 and macrophages or NK cells in human salivary adenocarcinoma-bearing nude mice given OK-432.

An immunoglobulin M mouse monoclonal antibody (MAb) to the streptococcal preparation OK-432, TS-1, was generated. The TS-1 antigen is a carbohydrate epitope. This antigen is stable upon fixation and embedding in paraffin. The tissue and cellular OK-432 localization in human salivary adenocarcinoma-bearing nude mice given OK-432 intratumorally was examined by various methods according to the immunological procedures using the purified TS-1 MAb. The presence of OK-432 antigen recognized by TS-1 MAb was clearly observed in the tumor as well as the spleen and lung 24 or 48 h after OK-432 administration, whereas transfer of OK-432 from the site of injection to the organs, such as liver and kidney, was rarely seen. The presence of OK-432 antigen in some immunocompetent cells, as defined by Mac-1 antigen or asialo GM1 antigen, was observed by the double-antibody labeling technique in the tumor and spleen from tumor-bearing nude mice. Moreover, interaction between OK-432 and macrophages or natural killer (NK) cells in relation to expression of interferon (IFN) in tumor-bearing nude mice given OK-432 was observed. Consequently, significant increases of Ia-positive or Ia-negative macrophages, NK cells as well as IFN-alpha/beta- or IFN-gamma-positive cells in the tumor and/or spleen were found when compared with those without OK-432 administration.

Small cell undifferentiated carcinoma of the minor salivary gland containing exocrine, neuroendocrine, and squamous cells.

The light microscopic, electron microscopic and immunohistochemical features of a small cell undifferentiated carcinoma of the minor salivary gland are presented. The tumor was composed predominantly of undifferentiated small cells with focally admixed neuroendocrine, exocrine and squamous cells, occasionally arranged in an organoid manner. The presence of vasoactive intestinal polypeptide in the tumor was found immunohistochemically. In addition, the tumor cells stained with Grimelius' impregnation. Immunohistochemically the tumor contained cells that reacted positively with the antibodies to carcinoembryonic antigen, 66K keratin polypeptide, or human salivary amylase. These findings indicate that a small cell undifferentiated carcinoma of the minor salivary gland, reported here, exhibits focally multidirectional differentiation as well as neuroendocrine cell derivation.

Induction of cells with acinar cell phenotype including presence of intracellular amylase. Treatment with 12-O-tetradecanoyl-phorbol-13-acetate in a neoplastic human salivary intercalated duct cell line grown in athymic nude mice.

The adenocarcinoma produced by transplantation into nude mice of a neoplastic human salivary intercalated duct cell line was treated with 0.1 ml of minimal essential medium (MEM) containing 12-O-tetradecanoyl-phorbol-13-acetate (TPA) at a final concentration of 10(-7) mol/l daily for 28 days and examined morphologically and immunohistochemically. The TPA treatment resulted in an enhancement of tumor growth. In addition, tumor cells containing secretory granules positively reactive to antiamylase serum were observed in the treated tumors, but not in untreated controls. These findings lead us to suggest that neoplastic intercalated duct cells treated with TPA can be induced to differentiate into acinar cells in heterotransplanted athymic nude mice.

Immunoreactive vasoactive intestinal polypeptide in acinic cell carcinoma of the parotid gland.

Vasoactive intestinal polypeptide (VIP) in the neoplastic cells of acinic cell carcinomas arising in the human parotid gland was found immunohistochemically, whereas other parotid gland tumors, such as pleomorphic adenoma, Warthin's tumor, oxyphilic adenoma, mucoepidermoid carcinoma, adenocarcinoma, and adenoid cystic carcinoma, did not show positive immunoreactivity for VIP. The acinic cell carcinoma stained with Grimelius impregnation and had dense core granules immunoreactive with anti-VIP serum. Moreover, a comparatively high concentration of immunoreactive VIP was detected by radioimmunoassay in an acinic cell carcinoma, whereas VIP concentration of the other tumors was undetectable.

5-Azacytidine induction of stable myoepithelial and acinar cells from a human salivary intercalated duct cell clone.

A neoplastic human salivary intercalated duct cell clone was cultured in 5 microM 5-azacytidine for 5 days at 37 degrees C; then the cells were trypsinized and subcultured in growth medium without 5-azacytidine. Thereafter, subclones were cloned from the subculture. Of 12 subclones isolated, 7 clonal cell lines were established and characterized. The two subclones composed of cells which were spindle shaped or stellate exhibited phenotypes similar to those of myoepithelial cells such as microfibrils and myosin and formed a myoepithelioma upon transplantation of the cells into nude mice. The other five subclones were composed of polygonal cells with numerous secretory granules in their cytoplasm and containing amylase that seems to be specific to acinar cells; transplantation of these cells into nude mice resulted in production of acinic cell carcinoma. These findings indicate that a neoplastic human salivary intercalated duct cell is capable of at least bidirectional differentiation.

The role of epithelial cell differentiation in the expression of herpes simplex virus type 1 in normal human oral mucosa in culture.

We have examined by immunofluorescent antibody staining technique the expression of herpes simplex virus type 1 (HSV-1) in organ cultures of the normal human oral mucosa. The expression of HSV-1 antigen was found selectively in the epithelial cell layers in relatively undifferentiated states such as basal layer and lower prickle cell layer in addition to the basement membrane. When the epithelial cells dissociated from the oral mucosa were infected with HSV-1 and association of the HSV-1 expression with the cellular differentiation was examined, the epithelial cells containing laminin in an undifferentiated state were permissive for the expression of HSV-1 antigen whereas terminally differentiated epithelial cells with the cornified envelope did not express HSV-1 antigen. These findings indicate that the expression of HSV-1 antigen is restricted in the mucosal epithelial cells in a differentiated state, although the possibility that the cornified envelope might protect the cells from infection is not excluded.

Macromolecular synthesis at the early stage of herpes simplex virus type 2 (HSV-2) latency in a human neuroblastoma cell line IMR-32: repression of late viral polypeptide synthesis and accumulation of cellular heat-shock proteins.

We have shown that a latent infection of herpes simplex virus type 2 (HSV-2) can be established in a human neuroblastoma cell line IMR-32 if the infected cells are cultured at 40 degrees C. In the present study, viral polypeptides and cellular heat-shock proteins which were synthesized in HSV-2 infected IMR-32 cells cultured at 40 degrees C were analyzed by polyacrylamide gel electrophoresis. It was found that the synthesis of late viral polypeptide ICP 5 was markedly reduced in the infected cells at 40 degrees C as compared with those at 37 degrees C. Although infection of IMR-32 cells with HSV-2 at 40 degrees C resulted in shutoff of cellular protein synthesis, it was found that some cellular heat-shock proteins (90, 72 and 70 kd polypeptides) were synthesized and accumulated intracellularly. These findings suggest that modification of cascade regulation of HSV-2 polypeptide synthesis and/or accumulation of heat-shock proteins may be involved in the incomplete arrest of virus growth and in survival of the infected cells, leading to the establishment of HSV-2 latency in IMR-32 cells.

Expression of vasoactive intestinal polypeptide and amylase in a human parotid gland adenocarcinoma cell line in culture.

A neoplastic epithelial cell line initially established in culture from a human parotid gland adenocarcinoma grown in athymic nude mice with a BALB/c genetic background, which has an ultrastructure similar to that of the intercalated duct cell of the salivary glands, was examined for the expression of amylase and vasoactive intestinal polypeptide (VIP). The cultured cells were found by the peroxidase-antiperoxidase (PAP) method to express amylase and ultrastructurally to have secretory granules showing positive immunoreaction with anti-amylase serum. In addition, the cells were found to secrete amylase into the culture medium. The expression of VIP in the cultured cells was observed by the PAP method, immunofluorescent staining method, or immunoelectron microscopy. Moreover, the presence of the polypeptide reactive to antibodies directed against VIP in the cultured cells was confirmed by immunoblotting and radioimmunoassay. These findings indicate that the cells proliferating in culture express both amylase and VIP.

Effect of dibutyryl cyclic AMP on morphologic features and biologic markers of a human salivary gland adenocarcinoma cell line in culture.

Dibutyryl cyclic AMP (dB-cAMP) has marked effects on the growth, morphologic features, and biologic markers of a human salivary gland adenocarcinoma cell line in culture. A cell line with ultrastructure and biologic markers specific to the intercalated duct cells of human salivary glands was cultivated in the presence of dB-cAMP. Major alterations, such as process formation and expression of myofilaments and oxytocin receptor in addition to myosin and S-100 protein, were observed in those cells with a phenotype similar to myoepithelial cells. Both the anchorage-independent and anchorage-dependent growths were markedly suppressed in the presence of dB-cAMP. After the removal of dB-cAMP from the culture, the treated cells returned rapidly to the phenotype of the untreated cells. These findings indicate that reversible differentiation into the myoepithelial cells of a human salivary gland adenocarcinoma cell line occurs in growth medium containing dB-cAMP.

A latent infection of herpes simplex virus type 2 in a human neuroblastoma cell line IMR-32.

Human neuroblastoma (IMR-32) cells were infected with herpes simplex virus type 2 (HSV-2) at a multiplicity of infection (MOI) of 2 plaque-forming units (PFU)/cell and were cultured at 40 degrees C for 14 days. Then neither infectious virus particles nor virus capsids were detected in these cells whereas the presence of virus-specific antigens was observed by immunofluorescent antibody staining technique in 16.9 +/- 3.2 per cent of the infected cell population. When the cultivation temperature was shifted down from 40 degrees C to 35 degrees C, reactivation of virus growth occurred after lag periods of 2-9 days. These findings indicate that the IMR-32 cells can be latently infected with HSV-2 at 40 degrees C and that virus growth may be inhibited at the level of synthesis of virus-specific macromolecules or at some step preceding nucleocapsid formation.

Generation of cells with phenotypes of both intercalated duct-type and myoepithelial cells in human parotid gland adenocarcinoma clonal cells grown in athymic nude mice.

A neoplastic epithelial cell line was established from nude mice tumors grown after transplantation of surgical specimens from a human parotid gland adenocarcinoma. This cell line, which had ultrastructure similarities to salivary intercalated duct cells, was found by immunohistochemical techniques to contain amylase, but myosin was not detected. Ultrastructurally, cells of an intermediate type between intercalated ductal and myoepithelial cells were found in the transplanted tumors. Moreover, the expression of myosin in addition to the presence of amylase was detected in the tumors. These findings indicate that some transplanted tumor cells appear to be differentiating towards myoepithelial cells.

Recurrent intraoral herpes simplex virus infection.

A 48-year-old female had primary herpetic gingivostomatitis, followed by recurrent intraoral herpes simplex virus (HSV) disease; HSV isolates were obtained from the swabs of primary and recurrent lesions; restriction endonuclease cleavage analysis of the viral DNAs extracted from Vero cells infected with the HSV isolates according to the method of Hirt was carried out. The viral DNAs were cleaved by restriction endonucleases such as BamHI, KpnI and SalI and resolved by agarose gel electrophoresis, followed by staining with ethidium bromide. Consequently, their cleavage patterns were very similar to one another and were identified as HSV type 1. From these findings, it can be concluded that primary and recurrent lesions of this case are caused by the same virus.

Immunohistochemical demonstration of carcinoembryonic antigen (CEA) on tissue sections from squamous cell head and neck cancer and plasma CEA levels of the patients.

45 squamous cell head and neck cancers including 36 with carcinoma of the oral cavity and 9 with carcinoma of the maxillary sinus were examined immunohistochemically for the presence of CEA. 12 of 30 carcinomas of the oral cavity and 7 of the 9 carcinomas of the maxillary sinus had tumors containing CEA. This difference in the occurrence of CEA was statistically significant. The mean plasma CEA level of 36 patients with carcinoma of the oral cavity and 7 patients with carcinoma of the maxillary sinus was 1.95 +/- 1.72 ng/ml and 3.70 +/- 3.53 ng/ml, respectively. Significant elevation of plasma CEA levels was found only in the stage-IV group patients with carcinoma of the oral cavity as compared with the stage-I group patients. In the 3 patients having plasma CEA values exceeding 5 ng/ml at the time of pretherapy, plasma CEA levels were decreased to below 2.5 ng/ml with the cancer treatment followed by the complete remission. These findings indicate that plasma CEA as a tumor marker in squamous cell head and neck cancer is meaningful in a small proportion of the cancer patient population.

Interferon activity and its characterization in the sera of patients with premalignant lesions arising in oral mucosa.

The interferon (IFN) assay of the sera from the 26 patients with premalignant lesions such as lichen planus and leukoplakia arising in oral mucosa was performed by the plaque-reduction assay with vesicular stomatitis virus in FL cells derived from human amniotic membrane. When the serum IFN activity was characterized by acid treatment, significant increase of acid-stable IFN in the patients was found as compared with those in the normal controls. The titers of gamma-like IFN defined by anti-HuIFN-alpha and anti-HuIFN-beta in the sera of patients of 50-79 years age group (n = 17, P less than 0.002) showed a highly significant increase as compared with the relevant normal controls (n = 20). All of the 26 patients were treated with topical administration of HuIFN-beta. When the correlation between prognosis of the disease and titers of serum IFN was investigated by measuring gamma-like IFN and acid-stable IFN in the sera of patients, all of 13 patients with good prognosis after the HuIFN-beta therapy showed significantly decreased levels of gamma-like IFN (P less than 0.01), whereas the serum level of acid-stable IFN after HuIFN-beta therapy showed a significant increase compared to that before the therapy (P less than 0.05). These findings indicate that the endogenous IFN system may be associated with the pathophysiology in patients with the oral mucosal lesions.

Quantitative analysis of global and regional cardiac performance in normal subjects and patients with coronary artery disease by rest/exercise radionuclide blood-pool study.

This study was undertaken to assess regional and diastolic left ventricular (LV) function in relation to LV asynchronous contraction during exercise (Ex). Rest and serial Ex (25w-75w) gated blood-pool study (Tc-99m-RBC) of 6 normal subjects (N), 12 patients with stable angina pectoris (AP) and 8 with LV aneurysm (LVan) were evaluated in regional LV ejection fraction (EF) and Fourier analysis using higher-order harmonics. In the patients with Ex and regional EF showed various responses to Ex. Global LVEF in the patients with AP indicated almost no change during Ex and regional EF declined apparently in anteroseptal segment, corresponding to stenotic left anterior descending artery. Diastolic indices of regional and global cardiac performance, PFR (peak filling rate), TPF (time to PFR) and standard deviation (SD) of TPF histogram, were obtained by Fourier analysis using two-order harmonics. Patients with coronary artery disease (CAD) showed significantly lower PFR, longer TPF and higher SD of TPF than normal group, suggesting asynchronous relaxation. We conclude that regional EF response and diastolic indices of cardiac performance to exercise provide useful and sensitive information detecting the severity of CAD.

An adenoid squamous carcinoma-forming cell line established from an oral keratinizing squamous cell carcinoma expressing carcinoembryonic antigen.

A neoplastic epithelial cell line, TYS, was isolated from a well-differentiated squamous cell carcinoma expressing carcinoembryonic antigen (CEA) that arose in human oral mucosa. Expressions of CEA and amylase as well as ample tonofilaments were detected in cultured TYS cells. Transplantation of the cells into athymic nude mice resulted in production of adenoid squamous cell carcinoma containing CEA and amylase. Cultivation of TYS cells in the presence of sodium butyrate resulted in suppression of cell growth and production of secretory granules with amylase in the cytoplasm of the cells. When the sodium butyrate-treated cells were transplanted into nude mice, a small mass developed transiently at the inoculation site and then disappeared. This mass was histopathologically interpreted as acinic cell carcinoma with squamoid lesion. These findings suggest that we have established a human adenoid squamous carcinoma cell line presumably derived from a minor salivary gland present in oral mucosa.

Immunoreactive somatostatin in Warthin's tumor.

The presence of somatostatin (SRIF) in the neoplastic epithelial cells of certain Warthin's tumors arising in the human parotid gland was found immunohistochemically, whereas the other parotid gland tumors, such as pleomorphic adenoma, oxyphilic adenoma, mucoepidermoid tumor, adenocarcinoma, and adenoid cystic carcinoma, did not show positive immunoreactivity for SRIF. The SRIF-positive Warthin's tumor had dense core granules immunoreactive with anti-SRIF serum. Moreover, a comparatively high concentration of immunoreactive SRIF was detected by radioimmunoassay in an SRIF-positive Warthin's tumor, although the other tumors also contained low levels of immunoreactive SRIF.
