[Phase II study of mitoxantrone for hematologic malignancies].

A phase II study of mitoxantrone (MIT) was performed in 21 patients with hematologic malignancies refractory to combination chemotherapy including anthracyclines. MIT was administered intravenously at doses of 8 to 13 mg/m2 on day 1 for 12 malignant lymphoma patients, and 1 to 3.3 mg/m2 on day 1 through 5 for 7 acute leukemia patients and 2 malignant lymphoma patients. Four malignant lymphoma patients (2 each of Hodgkin's disease and non-Hodgkin's lymphoma) achieved partial response lasting 19, 14, 8+, and 4 weeks, respectively. Although no definite response was obtained in acute leukemia patients, a marked cytoreduction was observed in 2 patients. Myelosuppression was a major toxicity, however, life-threatening toxicities were not observed in this study.

[Immunological significance of splenectomy in gastric cancer surgery. Production of immunosuppressive factor in the spleen].

We studied immunological significance of splenectomy in gastric cancer surgery with special reference to serum suppressor factor. The results showed that the sera from splenic venous blood in advanced gastric cancer was more suppressive to PHA blastogenesis of normal lymphocytes and also higher in the values of circulating immune complexes, as compared to the sera from peripheral blood. Furthermore, the media from cultures of spleen cells of patients with advanced gastric cancer indicated to suppress PHA blastogenesis of normal lymphocytes, as compared to the media from cultures of peripheral lymphocytes of the same patients. The suppressive activity was more increased in the media from cultures of nonadherent cell population obtained from the spleen cells. From these data, it was suggested that the spleen of patients with advanced gastric cancer was capable of producing serum suppressor factor. Therefore, gastrectomy combined with splenectomy may be reasonable in patients with advanced gastric cancer.

Molecular symmetry of the MoFe protein of nitrogenase. Structural homology/nitrogen fixation/x-ray crystallography.

X-ray diffraction data to 2.4-A resolution have been collected for native monoclinic crystals of the MoFe protein of nitrogenase from Clostridium pasteurianum. The MoFe protein is an alpha 2 beta 2 tetramer of 220,000 molecular weight with 1 molecule in the crystallographic asymmetric unit. A 6-A resolution rotation function shows the orientation of the crystallographic diad and pseudo mutually perpendicular diads representing 2-fold relationships between alpha and beta chains. Hence, at least at low resolution, there exists structural homology between these two polypeptide chains.

Physiologic status of the anorectum following sphincter-saving resection for carcinoma of the rectum.

The physiologic status of the anorectum after low anterior resections or pull-through operations of the rectum was evaluated clinically and by manometric studies. It was demonstrated that the presence of a normal anal resting pressure and an anorectal reflex were important to achieve postoperative continence after sphincter-saving operations. In patients with anastomotic leaks and poor function, a marked high-pressure zone in the anal canal and an anorectal reflex were not found, but as local inflammation resulting from the anastomotic leak disappeared, these parameters returned to normal. A normal anorectal reflex was found in one of three patients after Bacon-type pull-through operations, but the remaining two showed an increase of anal canal pressure during colonic distention. These results indicate that an elevation of anal-canal pressure in response to colonic distention plays a significant role in fecal continence at the time of "a sense of urgency" if internal sphincter function is impaired.

Amplitude of rippling in the P beta phase of dipalmitoylphosphatidylcholine bilayers.

We present x-ray diffraction results of dipalmitoylphosphatidylcholine (DPPC) multilayers in three structural phases. Using pure DPPC, precision temperature control, and high angular resolution methods, we have discovered splitting of the first diffraction order due to multilayering in the P(beta) phase. This splitting permits us to calculate the amplitude of ripples in this phase. The amplitude is large enough to suggest a structural mechanism for rippling.

Relationship between monensin and sulphur amino acid requirements in broiler chickens.

1. Diets containing 80, 100, 125, 150, 175 or 200 mg monensin/kg were fed to broiler chickens from 0 to 28 d in cages that prevented access to excreta. 2. Growth was depressed with 125 mg monensin or more/kg and food intake tended to decrease. Feathering was adversely affected with 175 mg or more/kg. 3. In a similar experiment, diets containing 8.8, 9.1, 9.4, 9.7 and 10.1 g total sulphur amino acids (SAA)/kg were supplemented with 125 or 80 mg monensin/kg and compared with a diet containing 8.8 g SAA and 33 mg robenidie/kg. 4. With 125 mg monensin/kg, body-weight gain was significantly less than that of birds receiving robenidine if dietary SAA content was 9.4 g or less/kg. With 9.7 g SAA or more/kg, gain in birds receiving monensin was similar to that of birds receiving robenidine. 5. Monensin at 125 mg/kg therefore appears to increase SAA requirement.

Histological and ultrastructural studies of nephroblastoma in rats induced transplacentally by ethylnitrosourea.

Histologic and ultrastructural features of nephroblastomas in Sprague-Dawley rats induced by a single transplacental injection of ethylnitrosourea (ENU) on the 15-16th day of gestation are described and their significance is discussed with respect to the histogenesis of the tumors. Many renal tumors were observed, in addition to predominant occurrence of tumors of the nervous system. Forty-two of 142 offsprings survived over 100 days developed renal tumors, of which 37 rats had nephroblastomas, three rats had cortical adenomas, and one rat had a cystadenoma. The nephroblastomas were found in 35 rats unilaterally and in two rats bilaterally. Most of these tumors had various amounts of hyalinized stroma. In the kidneys with nephroblastomas, isolated, small foci of renal blastema were also occasionally noted. Electron microscopy revealed that nephroblastomas consisted of two types of cells, namely, epithelial and mesenchymal cells. The epithelial cells showed cellular differentiation of varying degree; they are freely lying in the stroma, forming well differentiated tubules or immature tubule-like structures and solid cell nests. Most of the undifferentiated cells observed around the tubular structures and cell nests are demonstrated to have epithelial features. These observations seem to support the concept that nephroblastoma is originated from metanephric blastema.

DNA template activity in rat brain tumors induced by transplacental administration of ethylnitrosourea.

The ultracytochemical acridine orange (AO) method has been employed to demonstrate DNA template activity within experimental brain tumors induced by transplacental administration ethylnitrosourea (ENU) in Sprague-Dawley rats. Ultrastructural examination revealed that acridine orange binds to DNA exclusively within the active extended euchromatin portion of the cell nucleus of brain tumor and of foci of atypical cell proliferation. No AO reaction products were visible in the non-neoplastic cell nuclei adjacent to the brain tumors. The percentages of AO positive cells in brain tumor cells in rats given an intraperitoneal injection of 3H-thymidine 1 hr before sacrifice were between 2.4% and 8.0%. The average number of AO chromatin reaction products per single section of a cell nucleus varied between 7 and 21. The ratios of areas of euchromatin to heterochromatin were found to be slightly larger in AO positive nuclei then in AO negative nuclei in each of the tumors. The present results suggested that rat brain tumors induced by transplacental administration ENU exhibit de-repression of DNA template normally repressed in the adult state. The usefulness of this method for study on the development of experimental brain tumors is briefly discussed.

Proliferative response of mouse seminal vesicle epithelium to androgen and estrogen, assayed by incorporation of [125I]iododeoxyuridine.

The proliferation and death of androgen- and estrogen-responsive cells in the seminal vesicles of mice were investigated. Since 5-[125I]iodo-2'-deoxyuridine ([125I]IdUrd) was incorporated into seminal vesicle DNA, and since the steroid-induced change in [125I]IdUrd uptake by whole seminal vesicles paralleled the change in the mitotic index in epithelial cells, [125I]IdUrd uptake was used as an index in epithelial cells, [125I]IdUrd uptake was used as an index for the proliferation of epithelial cells. Injections of testosterone propionate (TP) in castrated mice induced a 50-fold increase in [125I]IdUrd uptake. When androgen-induced cell proliferation was labeled with [125I]IdUrd, the incorporated radioactivity was retained in mice given continuous TP injections but not in mice given continuous TP injections but not in mice given 17 beta-estradiol (E2) or vehicle. Although the injection of E2 into castrated mice induced a 10-fold increase in [125I]IdUrd uptake, E2 did not increase seminal vesicle weight, and when estrogen-induced cell proliferation was labeled with [125I]IdUrd, the incorporated radioactivity was not retained in mice given continuous E2 injections. The simultaneous injections of E2 and TP showed no significant additive effect on [125I]IdUrd uptake. The present results suggest that 1) androgen induces proliferation and differentiation of the seminal vesicle epithelium and maintains the differentiated cells; 2) estrogen induces epithelial cell proliferation without further differentiation or maintenance of the resulting cells. The seminal vesicle epithelial cells responsive to estrogen may be the same cells that are responsive to androgen.

Proliferative pattern of seminal vesicle cells and the production of testosterone and 5 alpha-androgens from birth to adulthood in mice.

Proliferation of seminal vesicle cells and the concentration of testicular and serum androgens (testosterone and 5 alpha-androgens) from birth to adulthood were investigated in mice. The weight of the seminal vesicles increased significantly from day 0 to day 10 after birth (approximately 0.1-1 mg), remained nearly constant in the next 10 days, and increased again (1-50 mg) thereafter. As an index of cell proliferation, two distinct peaks of 5-[125I]iodo-2'-deoxyuridine ([125I]IdUrd) uptake by the whole seminal vesicles were found at days 8 and 30. Types of proliferating cells examined by autoradiography after injection of [3H]thymidine were partially testis-dependent fibromuscular and epithelial cells in the first peak and largely testis-dependent epithelial cells in the second peak. The [125I]IdUrd uptake values found around day 20 and after day 40 were similar to those in neonatally castrated mice. Concentrations of testicular and serum androgens were relatively high on days 8, 30, 35, 40, and 60 (0.2-0.7 ng/mg tissue and 1.3-2.2 ng/ml, respectively) but were very low on day 18 (0.02 ng/mg tissue and 0.4 ng/ml). These findings lead to the hypothesis that androgens secreted from neonatal and prepubertal mouse testes play a major role in the proliferation of seminal vesicle cells and that the quiescent interval of androgen secretion and action occurs around the 20th day after birth in mice.

Growth of human hepatoma cells lines with differentiated functions in chemically defined medium.

A human hepatoma cell line, HuH-7, which was established from a hepatocellular carcinoma, was found to replicate continuously in a chemically defined medium when the medium was supplemented with Na2SeO3. The cells grew better in this medium than in serum-containing medium without any adaptation period. Other established human hepatoma and hepatoblastoma cell lines, HuH-6 cl-5, PLC/PRF/5, huH-1, and huH-4, also grew in the defined medium. Although HLEC-1 cells failed to proliferate continuously with Na2SeO3 alone, they grew if a cell-free conditioned medium from HuH-7 cells was added to the medium. These cell lines, except the HLEC-1 cell line, produced the following human plasma proteins among those examined: albumin, prealbumin, alpha 1-antitrypsin, ceruloplasmin, fibrinogen, fibronectin, haptoglobin, hemopexin, beta-lipoprotein, alpha 2-macroglobulin, beta 2-microglobulin, transferrin, lipoprotein, alpha 2-macroglobulin, beta 2-microglobulin, transferrin, Complement Components 3 and 4, and alpha 1-fetoprotein. Beside plasma proteins, the media from HuH-7, HuH-6 cl-5, PLC/PRF/5, and huH-1 contained anti-carcinoembryonic antigen-reactive proteins, and those from PLC/PRF/5, huH-1, and huH-4 medium contained hepatitis B surface antigen. These proteins were detected during periods of serial cultivation over 9 months under the above culture conditions. The hepatoma cell lines grown in the fully defined synthetic medium may provide a new approach for investigating the growth and metabolism of human hepatoma cells in vitro.

Intratumor immunochemotherapy with 5-fluorouracil emulsion and BCG in C3H/HE mice transplanted with MH134.

Responses to intratumor immunochemotherapy by intralesional injections of BCG and 5-fluorouracil (5-FU) emulsion, prior to resection of a transplanted primary tumor, were investigated using a host-tumor system consisting of C3H/He mice and MH134. A significant prolongation of survival and suppression of lymph node metastasis were attained by the combined use of BCG and 5-FU emulsion, compared to treatment with either BCG or 5-FU emulsion alone. However, this did not alter the survival rate of the combined regimen group as compared with those of the latter treatment groups. When rechallenged with the same tumor cells after resection of the primary lesion, mice treated with BCG alone exhibited a marked tumor growth suppression while the control and BCG + 5-FU emulsion group showed a less effective suppression. No such effects were observed in the group given 5-FU emulsion alone. Assessment by the splenic plaque-forming cell assay with sheep erythrocytes revealed a marked inhibition in the development of humoral immunity in the animals treated with 5-FU emulsion alone. Concomitant administration of BCG was effective to some extent in preventing the depression of these immune functions. The data obtained indicate that the clinical response to immunochemotherapy is determined by a balance between the antitumor effect of the antitumor agent administered and its influence on the host's immune functions.

[Experimental study on local immunochemotherapy].

The purposes of this work were twofold: firstly to determine whether intratumor chemoimmunotherapy was more effective than either treatment alone or systemic therapy and; secondly to study how the intratumor therapy affected on the development of the tumor-specific immunity. Inbred male C3H/He mice and mouse ascited hepatoma 134 (MH 134) of C3H origin were used as host-tumor system. Mitomycin C was used as the chemotherapeutic agent and BCG as the immunopotentiating agent. Intratumor treatment of MMC + BCG led to complete cure in 85 percent of the mice. The lymph node metastases were markedly inhibited in the group treated with MMC + BCG compared to the groups treated with MMC alone or BCG alone. The growth of rechallenged tumor was investigated; 79% of mice treated with MMC + BCG were immune to rechallenge, whereas 57% of mice treated with BCG alone. The number of PFC and DTH against SRBC of the mice treated with MMC intraperitoneally significantly decreased compared to that treated with MMC intratumorally.

[Clinical evaluation of the serum level of immunosuppressive acidic protein (IAP) and immunosuppressive substance (IS) in gastric cancer patients].

Recently, immunosuppressive acidic protein (IAP) and immunosuppressive substance (IS) were reported by two institutions, and they have been currently under extensive clinical investigations. As these two acidic proteins seemed to be very similar glycoproteins, we tried to elucidate clinically the dissimilarities between these two substances. The mean value of IAP of 106 healthy donors was 378 +/- 100 micrograms/ml, and as 95% of them was under 550 micrograms/ml, therefore, the upper limit of normal value of IAP was determined to be 550 micrograms/ml. On the other hand, the mean value of IS was 553 +/- 125 micrograms/ml and the upper limit of normal value was 750 micrograms/ml. In 110 gastric cancer patients, the incidence of the abnormal value of IS (43%) was higher than that of IAP (33%), and abnormal values were observed at earlier stage in IS than that observed in IAP. The reaction of skin tests was more highly correlated with the value of IS than the value of IAP. From these facts, it was suggested that the value of IS was more predictable than that of IAP as the parameter of the immunosuppression of the gastric cancer patients.
