[Intra-arterial infusion of CDDP in advanced gastrointestinal cancer (combination of general chemotherapy)].

We attempted to use CDDP for patients with advanced cancers of the gastrointestinal system by intra-arterial infusion, giving consideration to the side effects of CDDP. Of 19 cases treated with CDDP, 17 cases were evaluable. These 17 cases comprised 10 cases of gastric cancer, 1 of pancreatic cancer and 6 of colon cancer. Therapeutic effects were as follows. According to the criteria for judgement of solid cancers, there were 10 evaluable cases which comprised 1 case of PR, 2 of MR, 4 of NC and 3 of PD. According to the criteria for judgement of malignant ascites, there were 6 evaluable cases which comprised 4 effective and 2 non-effective cases. As to side effects, nausea and vomiting were observed in 10 cases, numbness in 1, fever in 1 and aggravation of diabetes in 2. From the above results, intra-arterial infusion of CDDP is considered to be an effective method for the treatment of advanced cancers of the gastrointestinal system, especially of malignant ascites.

Two modes of amber codon read-through in vitro.

Read-through translation of bacteriophage R17 amB2 coat cistron carrying an amber mutation at the seventh codon was studied in vitro using the crude cell extract (S30) derived from an Escherichia coli nonsuppressor strain. Despite the presence of termination factors as well as ribosome-releasing factor (RRF) which prevent the read-through translation [M. Ryoji, J. W. Karpen, and A. Kaji (1981) J. Biol. Chem. 256, 5798-5801], synthesis of coat-like protein still persists at a low level in this system. Characterization of this protein by peptide fingerprinting and amino acid sequencing was performed to reexamine the generally accepted notion that it is produced by amino acid misinsertion to the amber mutation codon. The results indicated, however, that the major population of this coat-like protein is produced as a result of reinitiation of translation from the eighth codon. Read-through by amino acid misinsertion in this system becomes predominant only when the Mg2+ concentration is higher than 16 mM.

Clinical effects of total-body hyperthermia combined with anticancer chemotherapy for far-advanced gastrointestinal cancer.

The authors treated 17 patients with far-advanced gastrointestinal cancer with extracorporeally induced total-body hyperthermia (TBHT) combined with anticancer chemotherapy. Although all patients' tumors were clinically resistant to prior chemotherapy with 1-(2-tetrahydrofurlyl)-5-fluorouracil or 5-fluorouracil and mitomycin C, three patients had an objective response to TBHT plus the same anticancer agents with or without cyclophosphamide. That is, a partial response was obtained in 2 of 12 patients with recurrent gastric cancer and in 1 of 5 patients with recurrent large bowel cancer. However, their survival time was not markedly prolonged. As a characteristic complication of TBHT, the authors noted reversible weakness of the muscles of the lower extremities. Two patients died due to hepatorenal syndrome; preoperatively these patients had manifested hepatic or renal dysfunction. Therefore, in patients with hepatic or renal dysfunction, the application of TBHT must be considered carefully.

Alteration of adenosine deaminase levels in peripheral blood lymphocytes of patients with gastric cancer.

The present study was undertaken to determine the adenosine deaminase (ADA) activity of peripheral lymphocytes in patients with gastric cancer, with respect to the cancer progression, the effect of surgery and/or immunotherapy. The gastric cancer patients showed lower lymphocyte ADA activity than did the normal control. The lymphocyte ADA activity did not decrease with the cancer progression. There was a significant correlation between lymphocyte ADA activity and blastogenesis of lymphocyte by phytohemaglutinin or concanavalin A. Six months following gastrectomy, the lymphocyte ADA activity was increased, as compared with the preoperative value. The ADA activity of patients on post-operative OK-432 showed a greater increase, as compared to that of patients not given this treatment. In conclusion, decreased lymphocyte ADA activity in gastric cancer patients might be due to either the cancer bearing status or to the immunological suppression.

Age-dependent change in sensitivity of oestrogen-induced uterine cell proliferation of mice, estimated by incorporation of [125I]iododeoxyuridine.

The stimulative effects of both oestradiol-17 beta (E2) and diethylstilbestrol (DES) injections on the proliferation of uterine cells of mice were investigated by using 5-[125I]iodo-2'-deoxyuridine [( 125I]IdUrd) incorporation as an index. Female mice of (WB X C57BL/6)F1 were neonatally castrated, and the [125I]IdUrd uptake by the whole uterus was determined on days 1, 5, 10, 20, 30 and 40 after birth. The relative minimal dose (a minimal dose expressed per body weight, microgram/g b.wt) of E2 necessary for the maximal [125I]IdUrd uptake was much higher than the relative minimal dose of DES on days 1, 5, and 10, when the serum concentration of alpha-fetoprotein was relatively high. However, the relative minimal dose of E2 was comparable to that of DES on days 30 and 40, when the concentration of alpha-fetoprotein was negligible. The difference between the effect of injected E2 and DES during neonatal and suckling periods seems to be attributable to the presence of alpha-fetoprotein in serum, which binds to E2 but not to DES with high affinity. When the relative minimal dose of DES necessary for the maximal [125I]IdUrd uptake was examined in detail, the value was lower on days 10 and 20 of age than on days 30 and 40 of age. This might represent the increased sensitivity of uterine cells to oestrogen on days 10 and 20.

Combining solid-state and solution-state 31P NMR to study in vivo phosphorus metabolism.

Otherwise unavailable information concerning the distribution of phosphorylated compounds in biological systems is obtained by a combined solid-state/solution-state NMR approach, illustrated here for oocytes from Rana pipiens. General methodology is developed, and further extensions are proposed. The following conclusions pertain to the specific system under examination. (i) Nucleoside phosphates can be observed by magic-angle sample spinning of the lyophilized material. (ii) The solid-state NMR technique of dipolar decoupling provides no additional resolution of the phospholipid and phosphoprotein components of the yolk. However, cellular death produces sufficient pH changes to cause the phospholipid and protein phosphate peaks to become resolvable. The concentration of nucleoside phosphates also decreases. (iii) The phospholipid and phosphoprotein components are shown by computer simulation to be present in a ratio of 40:60, respectively. (iv) The amounts of inorganic phosphate, nucleoside phosphates, and sugar phosphates are determined by solution-state NMR observation of the perchloric acid extract of the oocytes.

Quantitative studies on translocation and metabolic conversion of lysophosphatidylcholine incorporated into the membrane of intact human erythrocytes from the medium.

The fate of palmitoyl-lysophosphatidylcholine (lysoPC) incorporated into the membrane of intact human erythrocytes from a medium was investigated under nonhemolytic conditions at 37 degrees C by means of 14C-labeled tracers. The lysoPC was first incorporated into the outer half of the membrane lipid bilayer and then gradually translocated into the inner half during the incubation. At the same time it was metabolically converted into phosphatidylcholine (PC) and free fatty acid (FFA) plus glycerophosphorylcholine by the actions of acyltransferase and lysophospholipase, respectively. The half times of the conversion were about 14 h, while the value of 0.5 h was obtained when the half time was measured with the hemolysate of the lysoPC-loaded erythrocytes. Chymotrypsin treatment of unsealed ghosts caused a definite decrease in lysophospholipase activity, while similar treatment of resealed ghosts did not. This together with other evidence already reported in the literature suggests that both lysophospholipase and acyltransferase may be located in the inner surface of the membrane. The above findings strongly suggest that the most of the lysoPC loaded to the membrane is gradually translocated from the outer to the inner half of the bilayer and soon converted to either PC or FFA.

Trans-bilayer movement of added phosphatidylcholine and lysophosphatidylcholine species with various acyl chain lengths in plasma membrane of intact human erythrocytes.

14C-Labeled phosphatidylcholine (PC) and lysophosphatidylcholine (lysoPC) species with two homologous saturated acyl chains and of a saturated acyl chain of various lengths, respectively, were each incorporated into the outer leaflet of the membrane lipid bilayer of intact human erythrocytes, and the transbilayer movement into the inner leaflet during incubation at 37 degrees C of the lipid-loaded erythrocytes was followed. The labeled PC and lysoPC molecules present in the outer leaflet were extracted with egg-yolk PC liposome suspension and BSA solution, respectively, and the amount which moved into the inner leaflet during the incubation was measured by determining the residual amount of the labeled lipid in the membrane. Translocation of lysoPC molecules was also measured by assaying the decrease in the amount of the added labeled lysoPC in the membrane during the incubation on the basis of the previously reported fact that lysoPC molecules are all converted metabolically to PC or glycerylphosphorylcholine plus fatty acid as soon as they are translocated from the outer to the inner leaflet. Every lipid tested showed significant transbilayer movement during the course of the incubation for up to 10 h. With the C8, C10, and C12 species of PC the rate of the transbilayer movement increases with decreasing acyl chain length. The same is true with the C14, C16, and C18-lysoPC species.

Estrogen-induced increase in eosinophil number and peroxidase activity in uterus of genetically mast cell-deficient W/Wv mice.

The role of mast cells in induction of uterine eosinophilia was investigated by using genetically mast cell-deficient (WB X C57BL/6)F1-W/Wv (hereafter called WBB6F1-W/Wv) mice. The injection of estradiol-17 beta (0.16 micrograms/g body weight) increased the peroxidase activity and eosinophil number in the uteri of castrated WBB6F1-W/Wv and WBB6F1-+/+ mice. Since no significant differences were detectable between these two type of mice, mast cells did not seem to be essential for the estrogen-induced uterine eosinophilia, at least in mice.

Heterogeneity in mouse seminal vesicle epithelial cells responding to androgen as evaluated by incorporation of [125I]iododeoxyuridine.

When the uptake of 5-[125I]iodo-2'-deoxyuridine ([125I]IdUrd) into the seminal vesicle of castrated mice was measured 3 days after starting injections of various doses of testosterone propionate (TP), logarithmic values of [125I]IdUrd uptake were proportional to the logarithmic doses of TP in the range of 0.04-2 micrograms/g BW. The [125I]IdUrd uptake values correlated well with the labeling and mitotic indices of epithelial cells. Since daily injections of 0.4 microgram TP/g BW did not increase significantly the weight or DNA content or protein content of the seminal vesicle, the [125I]IdUrd uptake is a sensitive index of androgen action. Moreover, this suggests that low doses of androgen induce division of epithelial cells without resulting in the increase in cell number. The [125I]IdUrd radioactivity in the seminal vesicle was measured 2-15 days after the injection of [125I]IdUrd, since the value represented the fraction of surviving cells synthesizing DNA at the time of [125I]IdUrd injection. When injections of 4 micrograms TP/g BW were continued, the incorporated radioactivity was retained. In contrast, continuous injections of 0.2 microgram TP/g BW did not maintain the radioactivity, of which incorporation was induced by the same dose of TP. Thus, the present result suggests the presence of heterogeneity in androgen-responsive epithelial cells of the seminal vesicle.

Hormonal control of alpha-fetoprotein secretion in human hepatoma cell lines proliferating in chemically defined medium.

The regulation of alpha-fetoprotein (AFP) secretion and growth rate by various hormones in established human hepatoma (HuH-7, PLC/PRF/5, huH-1, huH-4, and KIM-1/c-4) and hepatoblastoma (HUH-6 Clone 5) cell lines was studied. These 6 cell lines replicated continuously in a chemically defined medium and secreted 84 ng (HuH-7) to 23 pg (huH-4) AFP per 24 h per 1 X 10(4) cells into the culture medium. The addition of insulin increased the growth rate of all examined cell lines and partially inhibited the AFP secretion in those cell lines except KIM-1/c-4, while the addition of dexamethasone inhibited the growth and stimulated the AFP secretion in all of the cell lines. The addition of 3,3',5-triiodothyronine inhibited the growth of all cell lines; however, different effects on the AFP secretion were observed depending on the cell lines used. Obviously, the AFP secretion was unrelated to the change in growth rate. When dexamethasone and N6-O2-dibutyryl cyclic AMP were added together, the AFP secretion was further stimulated. On the other hand, when dexamethasone and insulin were added simultaneously, the dexamethasone-mediated stimulation of AFP secretions was diminished. The data indicated that the regulatory mechanisms of AFP secretion by the hormones in the established human hepatoma and hepatoblastoma cell lines cannot be deduced according to the results of one cell line.

Effects of aphidicolin and alpha-amanitin on visualization of acridine orange binding to DNA in rat astrocytoma C6 cells.

The purpose of this study was to examine effects of aphidicolin and alpha-amanitin on visualization of acridine orange (AO) binding to DNA in rat astrocytoma C6 cells and to discuss briefly the significance of AO chromatin interaction products. Aphidicolin inhibited DNA synthesis but percentage of AO positive cells was approximately 60% of that of the untreated control cells. alpha-Amanitin caused a slight inhibition of RNA synthesis and 3H-uridine incorporation in the treated cells was about 64% of that of the untreated cells, whereas a distinct decrease of the number of AO positive cell nuclei was observed. The results suggest that activity of RNA polymerase II and mRNA synthesis is mainly concerned in visualization of AO chromatin interaction products.

[The effect of vagotomy on carcinogenesis in the glandular stomach of rats, induced by N-methyl-N'-nitro-N-nitrosoguanidine].

The effect of hydrochlorhydria caused by vagotomy on carcinogenesis in the glandular stomach of male rats was studied. Group A (35 rats): After N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) 80 mg/l solution was orally administered during the first 15 weeks of life, vagotomy was performed. Group B (35 rats): After the oral administration of MNNG 80 mg/l solution during the first 15 weeks of life, laparotomy was done. Group C (10 rats): As the control, vagotomy was undertaken at the 15th week of life. Group D (10 rats): As the control, laparotomy was done at the same time. At the 52nd week, all surviving rats were autopsied, and gastrin cell counts and body weight were ascertained. The incidence of adenocarcinoma was 62% in Group A, 32% in Group B (p less than 0.05) and nil in Group C and D. These results strongly support the view that the hypochlorhydria plays the role of a promoting factor in producing gastric carcinoma.

Establishment and characterization of cell lines derived from a human osteosarcoma.

Continuously growing cell lines have been established in vitro from a human osteosarcoma after transplantation into athymic nude mice. These cell lines grew as an adherent monolayer and consisted of various types of cells. Twelve clones were colonially isolated from a cell line, HuO-3N1, and subdivided into three groups depending on the morphologic features. The cells of HuO-3N1 and its clones had alkaline phosphatase (ALP)-positive granules in the cytoplasm. Cytogenetic studies showed that these cell lines were human aneuploid lines. A tumor was produced by injection of HuO-3N1 cells into an athymic nude mouse. ALP activity increased in a clonal cell line, HuO-3N1 cl-2, when cells were treated with 1,25-dihydroxy-vitamin D3. The proliferation of cells was inhibited when the cells were cultured in a medium supplemented with L-homoarginine, which is an inhibitor of bone and liver-specific ALP. This cell line has an osteoblastic phenotype and provides a useful model for studies of human osteosarcoma and phenotypical expression of human osteoblastic cells.
