Two distinct CCR5 domains can mediate coreceptor usage by human immunodeficiency virus type 1.

The chemokine receptor CCR5 is the major fusion coreceptor for macrophage-tropic strains of human immunodeficiency virus type 1 (HIV-1). To define the structures of CCR5 that can support envelope (Env)-mediated membrane fusion, we analyzed the activity of homologs, chimeras, and mutants of human CCR5 in a sensitive gene reporter cell-cell fusion assay. Simian, but not murine, homologs of CCR5 were fully active as HIV-1 fusion coreceptors. Chimeras between CCR5 and divergent chemokine receptors demonstrated the existence of two distinct regions of CCR5 that could be utilized for Env-mediated fusion, the amino-terminal domain and the extracellular loops. Dual-tropic Env proteins were particularly sensitive to alterations in the CCR5 amino-terminal domain, suggesting that this domain may play a pivotal role in the evolution of coreceptor usage in vivo. We identified individual residues in both functional regions, Asp-11, Lys-197, and Asp-276, that contribute to coreceptor function. Deletion of a highly conserved cytoplasmic motif rendered CCR5 incapable of signaling but did not abrogate its ability to function as a coreceptor, implying the independence of fusion and G-protein-mediated chemokine receptor signaling. Finally, we developed a novel monoclonal antibody to CCR5 to assist in future studies of CCR5 expression.

Human papillomavirus and cervical neoplasia: a case-control study in Taiwan.

As part of a large-scale, community-based cervical neoplasia screening project in rural Taiwan, a case-control study was undertaken to evaluate the etiologic role of human papillomavirus (HPV) infection in this mainly monogamous (2% reported having multiple sexual partners) female population. A total of 88 biopsy-confirmed cases and 261 cytologically normal controls were selected for the study. The case group included 40 cases of cervical intraepithelial neoplasia (CIN) 1, 9 of CIN 2, 36 of CIN 3 and 3 cases of invasive cancer. Cervical swabs collected at screening from study subjects were tested for HPV DNA by an LI consensus primer polymerase chain reaction (PCR)-based technique. HPV DNA was found in 92% of high-grade cases (CIN 2-3 and invasive cancer); 54% of low-grade cases (CIN 1); and 9% of controls. HPV was significantly associated with both high-grade and low-grade cervical neoplasia. As reported in Western countries, HPV 16 was the predominant type among HPV-positive high-grade cases. However, HPVs 52 and/or 58 combined were the most common types among HPV-positive low-grade cases and controls. Among women without any high-risk HPV infection (types 16, 18, 31 or 45), those with multiple-type HPV infection had a higher risk for high-grade cervical neoplasia than those with single-type infection. Overall, 91% of high-grade cases and 50% of low-grade cases could be attributed to HPV infection. Our results show that, even in this monogamous population, HPV is the major risk factor for high-grade cervical neoplasia.

Purification of (-)-epigallocatechin from enzymatic hydrolysate of its gallate using high-speed counter-current chromatography.

Epigallocatechin gallate (EGCG) was hydrolyzed at various concentrations of tannase under pH 6.0 at 35 degrees C, and the reaction mixtures were separated by high-speed counter-current chromatography with a two-phase solvent system composed of hexane-ethyl acetate-water (1:13:20). The best results were obtained when 2 mg/ml of the enzyme buffer solution was added to 0.3 M EGCG buffers at a rate of 0.1 ml/min. Using 10 mg of the enzyme, 342 mg of epigallocatechin were obtained at a purity of 99.1%.

Epidemiology and partial nucleotide sequence of four novel genital human papillomaviruses.

Polymerase chain reaction (PCR)-based genital human papillomavirus (HPV) detection methods that use consensus primers have enabled the broad-spectrum detection of most characterized HPV types. In addition, these techniques have allowed the identification of potentially novel viral sequences in clinical specimens. These methods were used to determine the partial L1 nucleotide sequence (the region generated by L1 consensus primers MY09 and MY11) of four novel viruses. The prevalence of these viruses in cytologically normal and dysplastic cervical specimens and in invasive cervical cancer was also determined. The partial DNA sequences of W13B (MM4), PAP291 (MM7), PAP155 (MM8), and PAP238a (MM9) are most similar to HPV-51, -61, -61, and -34, respectively. Prevalence studies suggest that W13B and PAP238a are cancer-associated, while PAP155 and PAP291 appear to be lower-risk viruses.

The 0.7 to 3.3 megabase chromosomes from Candida, Kluyveromyces, and Pichia provide accurate size standards for pulsed field gel electrophoresis.

Pulsed field gel electrophoresis was used to size intact chromosomal DNAs from Candida albicans, Kluyveromyces lactis, Pichia scolyti, and Pichia mississippiensis by optimization methods using, as size standards, concatenated bacteriophage lambda DNA, and intact and NotI digestion products of Schizosaccharomyces pombe chromosomal DNAs. These newly sized fungal DNAs can now serve as convenient and accurate size standards for DNA molecules between 0.7 and 3.3 megabases (Mb). These size standards are valid over a wide range of different electrophoretic conditions.

[The spectroelectrochemical characterization of electrochemical oxidation mechanism of vitamin E].

The electrochemical oxidation mechanism of vitamin E(VE) on pyrolytic graphite electrode (PGE) was studied by cyclic voltammetry. The results of experiment showed that the oxidative mechanism of VE was a quasi-reversible ECE process controlled by diffusion on PGE and the same on platinum electrode (PtE). The results of absorption spectroelectrochemistry experiment verified the oxidative mechanism of VE. It is clear that the oxidation of VE is a quasi-reversible 2e process to a divalent cation and followed by hydrolysis reaction to the quinone or semi-quinone compounds in acidic solution.

Secondary pulsed field gel electrophoresis: a new method for faster separation of larger DNA molecules.

A novel technique, which we call secondary pulsed field gel electrophoresis (SPFG) has been developed. In SPFG, short pulses are applied in the direction of net migration of the DNA in addition to the reorienting pulses used in conventional pulsed field electrophoresis (PFG). Experimental results show that SPFG extends and improves the electrophoretic resolution of DNA for molecules from 0.5 megabase pairs to over 10 megabase pairs in size. This improved resolution is obtained with dramatically shorter run times. Thus SPFG appears to circumvent a number of the key limitations in previous PFG protocols.

Studies on the preparative capability of the horizontal flow-through coil planet centrifuge and high-performance liquid chromatography in the separation of polar compounds from Oxytropis ochrocephala Bunge.

Horizontal flow-through coil planet centrifuge (CPC) and high-performance liquid chromatography (HPLC) techniques were used for separation of polar compounds from a crude ethanol extract of Oxytropis ochrocephala Bunge, a poisonous legume plant widely distributed in northwestern China. The performance of these two chromatographic methods was compared in terms of column efficiency, peak resolution, separation time, sample loading capacity, etc. The results indicated that two polar compounds in the crude extract were equally well separated by these two methods. HPLC gave comparable peak resolution in shorter separation time while its sample loading capacity was limited to the mg range. The CPC method required a long separation time, but yielded a higher purity of fractions with a much greater capacity.

Cross-axis synchronous flow-through coil planet centrifuge for large-scale preparative counter-current chromatography. III. Performance of large-bore coils in slow planetary motion.

A preparative capability of the present cross-axis synchronous flow-through coil planet centrifuge was demonstrated with 0.5 cm I.D. multilayer coils. Results of the model studies with short coils indicated that the optimal separations are obtained at low revolutional speeds of 100-200 rpm in both central and lateral coil positions. Preparative separations were successfully performed on 2.5-10 g quantities of test samples in a pair of multilayer coils connected in series with a total capacity of 2.5 l. The sample loading capacity will be scaled up in several folds by increasing the column width.

Cross-axis synchronous flow-through coil planet centrifuge for large-scale preparative counter-current chromatography. I. Apparatus and studies on stationary phase retention in short coils.

Using a new cross-axis synchronous flow-through coil planet centrifuge with a 20-cm revolutional radius, the retention of the stationary phase for nine solvent systems was studied with short coils mounted at two different locations on three holders with 5-, 15- and 25-cm hub diameters. Coils mounted 10 cm to the left of the center of a holder produced a much improved retention of most of the solvent systems compared with the same coils mounted at the center of the holder. In the lateral coil position the retention was found to be affected by the direction of the planetary motion and the head-tail elution mode. This phenomenon may be attributed to the effect of the lateral force field acting asymmetrically between the upper and lower halves of the coil.

Rapid separation of flavonoids by analytical high-speed counter-current chromatography.

A commercial model of the analytical high-speed counter-current chromatography instrument was used for separation of flavonoids from a crude ethanol extract of dried fruits of sea buckthorn (Hippophae rhamnoides). Using a two-phase solvent system of chloroform-methanol-water (4:3:2), a five-fold increase in flow-rate of the mobile phase from 60 to 300 ml/h resulted in a rapid separation of five components in less than 15 min without significant loss in peak resolution. Major flavonoid component, isorhamnetin, was identified in its pure state by mass spectrometric analysis.

Multi-stage mixer-settler planet centrifuge. Preliminary studies on partition of macromolecules with organic-aqueous and aqueous-aqueous two-phase solvent systems.

A rotary-seal-free planetary centrifuge holds a separation column which consists of multiple partition units (ca. 200) connected in series with transfer tubes. In the cavity of each partition unit the transfer tube extends to form a mixer which vibrates to stir the contents under an oscillating force field generated by the planetary motion of the centrifuge. Consequently, solutes locally introduced at the inlet of the column are subjected to an efficient partition process in each partition unit and separated according to their partition coefficients. The mixer tube equipped with a flexible silicone rubber joint was found to produce excellent results for partition with viscous polymer phase systems. The capability of the method was demonstrated on separation of cytochrome c and lysozyme using a PEG-aqueous dibasic potassium phosphate-aqueous two-phase solvent system.

Separations of flavonoids and alkaloids in medicinal herbs by high-speed counter-current chromatography.

Counter-current chromatography is a new liquid-liquid partition chromatography without using solid support. Recently, the technique has been remarkably improved in both partition efficiency and separation time. In this paper the capability of this high-speed counter-current chromatography was demonstrated on separation of two sets of samples obtained from medicinal herbs: a synthetic mixture of 3'-hydroxygenkwanin, luteolin and apigenin was separated on a two-phase solvent system composed of chloroform-methanol-water (4:3:2, v/v/v) and a crude ethanol extract from Anisodus tangulicus (Maxin) Pasch on chloroform-0.07 M sodium phosphate (pH 6.4) (1:1, v/v). In the light of chromatograms obtained from these samples, advantages of high-speed counter-current chromatography over other chromatographic methods were discussed in terms of partition efficiency, peak resolution, separation time, sample loading capacity, etc.
