Diverse effects of heparin on mitogen-activated protein kinase-dependent signal transduction in vascular smooth muscle cells.

Proliferation of vascular smooth muscle cells (SMCs) is implicated in pathological events, including atherosclerosis and intimal hyperplasia following angioplasty. The glycosaminoglycan heparin is a growth inhibitor of SMCs in vitro and in vivo. The underlying mechanism, however, is still poorly understood. In the present study, we report that heparin inhibited the activation of the mitogen-activated protein kinase (MAPK) in baboon SMCs by serum but not by platelet-derived growth factor (PDGF). When fibroblast growth factor was used, heparin had a stimulatory effect on MAPK. The only MAPK-activating kinase found in SMCs was MAPK kinase (MAPKK)-1, although MAPKK-2 was present in comparable amounts. Activation of MAPKK-1 and DNA synthesis were affected by heparin in a similar fashion. Heparin does not appear to exert its effects through members of the protein kinase C family, which are downregulated by phorbol esters, because it was still capable of inhibiting MAPK/MAPKK-1 stimulation by FCS in phorbol ester-pretreated cells. The present findings support the conclusions that the effects of heparin depend on the nature of the mitogen and that heparin inhibits SMC proliferation by preventing activation of MAPKK-1.

The effects of artificial lung inflation on pulmonary blood flow and heart rate in the turtle Trachemys scripta.

As for most ectothermic vertebrates, the breathing pattern of turtles is episodic, and pulmonary blood flow (Qpul) and heart rate (fH) normally increase several-fold during spontaneous ventilation. While some previous studies suggest that these cardiovascular changes are caused by stimulation of pulmonary stretch receptors (PSRs) during ventilation, it has been noted in other studies that blood flows often change prior to the initiation of breathing. Given the uncertainty regarding the role of PSRs in the regulation of central vascular blood flows, we examined the effect of manipulating lung volume (and therefore PSR stimulation) on blood flows and heart rate in the freshwater turtle Trachemys scripta. Turtles were instrumented with blood flow probes on the left aortic arch and the left pulmonary artery for measurements of blood flow, and catheters were inserted into both lungs for manipulation of lung volume. In both anaesthetized and fully recovered animals, reductions or increases in lung volume by withdrawal of lung gas or injection of air, N2, O2 or 10% CO2 (in room air) had no effect on blood flows. Furthermore, simulations of normal breathing bouts by withdrawal and injection of lung gas did not alter Qpul or fH. We conclude that stimulation of PSRs is not sufficient to elicit cardiovascular changes and that the large increase in Qpul and fH normally observed during spontaneous ventilation are probably caused by a simultaneous feedforward control of central origin.

Ventilation and gas exchange in lizards during treadmill exercise.

The extent to which lizards ventilate their lungs during locomotion is controversial. Direct measurements of airflow across the nostrils suggest a progressive reduction in tidal volume and minute ventilation with increased running speed, while other studies have demonstrated that arterial PO2 remains constant during exercise. To resolve these conflicting findings, we measured minute ventilation and gas exchange rate in five specimens of Varanus exanthematicus and five specimens of Iguana iguana during treadmill locomotion at speeds between 0.14 and 1.11ms-1 at 35 degrees C. These speeds are much lower than maximal running speeds, but are greater than the maximal aerobic speed. In both species, the ventilatory pattern during locomotion was highly irregular, indicating an interference between locomotion and lung ventilation. In Varanus exanthematicus, treadmill locomotion elicited a six- to eightfold increase in minute ventilation from a pre-exercise level of 102mlkg-1min-1, whereas the rate of oxygen uptake increased approximately threefold (from 3.9 to 12.6mlkg-1min-1). After exercise, both minute ventilation and gas exchange rate decreased immediately. Because minute ventilation increased more than did oxygen consumption, an increase in lung PO2 during exercise is predicted and, thus, Varanus exanthematicus appears effectively to ventilate its lungs to match the increased metabolic rate during locomotion at moderate speed. In Iguana iguana, both minute ventilation and gas exchange rate increased above resting values during locomotion at 0.28ms-1, but both decreased with further increases in locomotor speed. Furthermore, following exercise, both minute ventilation and oxygen uptake rate increased significantly. Iguana iguana, therefore, appears to be unable to match the increased oxygen demand with adequate ventilation at moderate and higher speeds.

A comparison of two long-acting vasoselective calcium antagonists in pulmonary hypertension secondary to COPD.

UNLABELLED: STUDY OBJECTIVES AND PATIENTS: Pulmonary hypertension (PH) is common in COPD and may predict mortality in this disorder. We have compared the pulmonary vasodilator effects, dose-response characteristics, and tolerability of two calcium channel blockers, amlodipine and extended-release (ER) felodipine, in 10 patients (seven men, age 68+/-4.8 [SD] years) with clinically stable COPD and PH. DESIGN: Drugs were given in equal single daily oral doses (2.5, 5, and 10 mg), increasing weekly for 3 weeks, in a randomized investigator-blinded crossover manner with a 1-week wash-out period between the two treatments. MEASUREMENTS: Doppler measurements of pulmonary hemodynamics were made on the seventh day of treatment at each drug dose. Lung function, arterial blood gases, and adverse events were also monitored weekly. RESULTS: A dose-dependent decline of pulmonary artery pressure (PAP) was observed with each drug. A dose of 2.5 mg produced a significant decrease in PAP compared with baseline (20% amlodipine, 17% felodipine ER). Additional decreases in PAP were observed at 5 mg and 10 mg that were similar for both drugs, but did not reach statistical significance compared with 2.5 mg. There was a dose-related decrease in pulmonary vascular resistance and increase in oxygen delivery with amlodipine and felodipine ER. Lung function and blood gas values were stable throughout. Side effects (headache and ankle edema) were less frequent during amlodipine treatment (p<0.05). CONCLUSIONS: Both amlodipine and felodipine ER, given as a single daily oral dose of > or = 2.5 mg, are effective pulmonary vasodilators in COPD patients with PH. Their dose-response characteristics are similar, but amlodipine treatment was associated with fewer side effects.

Hyaluronan decreases peritoneal fluid absorption in peritoneal dialysis.

Hyaluronan, exhibiting a high resistance against water flow, acts in the tissue as a barrier against rapid changes in water content. To test whether hyaluronan has any effect on the peritoneal fluid and solute transport, and, in particular, on the peritoneal fluid absorption, a 4-h dwell study with an intraperitoneal volume marker (radiolabeled human serum albumin [RISA]) was conducted in 21 male Sprague Dawley rats (three groups, seven rats in each group). Each rat was injected intraperitoneally with 25 ml of 1.36% glucose solution alone (control group), with 0.005% hyaluronan (HA1 group), or with 0.01% hyaluronan (HA2 group). Dialysate and blood samples were taken frequently for analyses of fluid and solute (urea, glucose, and protein) transport. The intraperitoneal volume was calculated from the dilution of RISA with a correction for RISA disappearance from the peritoneal cavity. This study shows that adding hyaluronan to peritoneal dialysis solution significantly (P < 0.01) increased the net peritoneal fluid removal, mainly due to a significant decrease in the peritoneal fluid absorption rate (P < 0.01). The diffusive mass transfer coefficients for glucose, urea, and protein did not differ between the three groups. The peritoneal clearance of urea increased significantly in the two hyaluronan groups compared with the control group, due to the increased net fluid removal in the hyaluronan groups. These results suggest that intraperitoneal administration of hyaluronan during a single peritoneal dialysis exchange may significantly increase the peritoneal fluid and solute removal by decreasing peritoneal fluid absorption.

Vertical impact forces during bench-step aerobics: exercise rate and experience.

The purpose of this study was to determine the effects of two bench-step exercise speeds on vertical impact forces and to explore this variable between novices, and instructors. 12 women (mean age 24 yr.) randomly performed 8-min. protocols of the "basic" bench-stepping technique and a more advanced "travel" technique at 30 and 33 cycles.min.-1. Analysis showed that the faster exercise rate yielded significantly higher vertical impact forces on a reference (B-8) step height (20.3 cm). At 33 cycles.min.-1, the instructors, and novices' responses were both higher than those at 30 cycles.min.-1. The mean peak vertical impact force ranged from 1.54 times the body weight for the novice group at 30 cycles.min.-1 to 1.87 times the body weight for instructors at 33 cycles.min.-1. A comparison of the groups' force curves showed a distinctive pattern in the loading of the impact forces. Specifically, the instructors consistently produced a transitory decrement in force prior to attaining peak force. In addition, the novices exhibited nonuniform increases in the production of vertical impact force across other step heights at the faster (33 cycles.min.-1) speed. Thus, experience with bench-step exercise may afford an ability to make uniform and force-absorbing adjustments in the resultant vertical impact forces at increased speeds.

[Expression and antagonist role of endothelin and nitric oxide synthase in atherosclerotic plaque].

OBJECTIVE: To study the pathogenetic mechanism of atherosclerotic plaque, the action of mediation and antagonism of endothelin (ET) and nitric oxide synthase (NOS) was investigated. METHODS: In situ hybridization, RT-PCR on endothelin and NOS, cytochemistry on NOS were measured using the rabbit atherosclerosis model and cultured vascular smooth muscle cells (VSMC) from normal rabbit. RESULTS: Transcription of endothelin mRNA increased and transcription of NOS mRNA decreased in astherosclerotic plaque: compared with normal aorta, expression of ET gene in plaque was increased by 1.2 times and the expression of NOS gene was decreased by 22.2%; cytochemistry combined with image pattern analysis showed that ET could inhibit NOS protien synthesis in VSMC; type A receptor antagonist of ET could inhibit the role of ET which causes a decrease of NOS protein in VSMC. CONCLUSION: The imbalance between NOS and ET, namely abnormal increase of ET and/or obvious decrease of NOS, is related to atherosclerotic plaque formation.

[Study on the relationship between intratumor microvessel density and neck metastasis of laryngeal and hypopharyngeal squamous cell carcinomas].

Sixty-one laryngeal and hypopharyngeal squamous cell, carcinoma (LC, HPC) tissue slides were immunochemically stained using LSAB method to study epithelium cells. The results demonstrated that (1) intratumor microvessel density (ITMD) in LC and HPC group was higher than that of the benign group (P < 0.05). ITMD was higher in the subgroup of LC and HPC with positive lymph node positive than that with negative lymph nodes. This result suggest that ITMD is relevant not only to the nature of the tumor, but also to lymph node metastasis. The level of ITMD is an important predictive sign of metastasis. (2) The relationship between ITMD and the clinical staging had no statistic significance (P > 0.05). (3) The analysis on the relationship between ITMD and pathologic differentiation indicated that the level of ITMD raised gradually with the lowering of the pathologic differentiation.

[Early observation of healing of rabbit mandibular defects with porous blocks consisting of polylactic acid and recombinant human bone morphogenetic protein-2].

The effects of bone morphogenetic protein (BMP) which can induce bone formation have been surely proved, Porous polylactic-acid blocks were prepared by a freeze-drying method. During preparation of the blocks, recombinant human bone morphogenetic protein-2(rhBMP-2) was mixed with the polylactic-acid(4 mg per block). PLA-rhBMP-2 was implanted in the mandibular defects of rabbit and retrieved after 2 or 4 weeks. The results showed that: with the PLA-rhBMP-2 blocks, new bone formation was observed in 2 weeks after implantation, and plain PLA blocks produced a little of new bone formation in the edges of defect even in 4 weeks after implantation. Calcium content of the retrieved PLA-rhBMP-2 block was statistically higher than that of plain PLA blocks. These results suggest that PLA may be effective delivery system for BMP and the PLA/rhBMP-2 block may potentially have usefulness as a bone graft substitute.

Integrated regulation in response to simulated weightlessness.

To investigate physiological effects of tail suspension, Ca2+ concentration, immune factors, erythrocyte rheological properties, and growth hormone were determined in rats suspended for 15 and 30 d. The results showed that inhibitory changes of both local factors (proteins secreted by bone cell, Ca(2+)-ATPase in sarcoplasmic reticium) and integrated regulative factors (immune factor, growth hormone) were observed simultaneously with the decrease of bone mineral content, calcium transportation in skeletal muscles as well as erythrocyte deformability. It suggests that both local and integrated regulative processes are functioning in response to the effects of weightlessness.

[Investigation on the elimination of organic substances in urine by supercritical water oxidation].

Application of Supercritical Water Oxidation (SCWO) to processing of human metabolic wastes of Controlled Ecological Life Support System was studied for organic substance elimination in urine and recover potable water. The SCWO of simulated urine was investigated with oxygen as the oxidant at 400-500 degrees C and 24-30 MPa in a continuous flow reactor. The organic substance elimination efficiency was obviously effected by temperature, pressure and reaction time. It is lower than 10% at 400 degrees C, but more than 95% at 550 degrees C with reaction time of 2 min.

Effect of simulated weightlessness on TNF-alpha production and the response of bone marrow cells to GM-CSF in rats.

The purpose of this experiment was to investigate the effect of 15 and 30 d simulated weightlessness (SWL) on tumor necrosis factor-alpha(TNF-alpha) production secreted by macrophage stimulated with lipopolysaccharide(LPS) and the ability of bone marrow cells to respond to granulocyte/monocyte colony-stimulating factor(GM-CSF) in tail-suspension rats. The TNF-alpha production was assessed by radioimmunoassay. The ability of bone marrow cells to respond to GM-CSF was measured by 3H-TdR incorporation method. The results showed that after 15 d SWL, TNF-alpha production showed a tendency to decrease; and that after 30 d SWL, the decrease became significant (P < 0.05). The results also demonstrated that the ability of bone marrow cells to respond to GM-CSF decreased significantly after 15 d SWL (P < 0.01); and there was no significant change after 30 d SWL. It suggested that SWL could depress immune function, and that reduction of TNF-alpha production might be related to bone marrow cell proliferation function.
