[Studies on DNA amplification fingerprinting of cortex Magnoliae officinalis].

Total genomic DNA samples of Chinese traditional medicine(CTM) Cortex Magnoliae Officinalis ("Houpo"), its counterfeits and substitutes were amplified by DNA amplification fingerprinting(DAF). Their amplified DNA banding patterns were distinguished on PAGE. The results show that DAF can be used to identify CTM "Houpo", its counterfeits and substitutes, which is also quite valuable for correctly introducing original plant.

[A population-based surveillance system on birth defects and its application].

OBJECTIVE: To describe a unique surveillance system on birth defects in the People's Republic of China. METHODS: The system was instituted in 1992 as a component for the evaluation on the effectiveness of a community intervention program using periconceptional folic acid supplementation to prevent neural tube defects and currently carries out surveys on a birth cohort of approximately 150 000 infants per year. Information was collected in the form of detailed written descriptions by local health care providers and photographs of affected infants. The system allowed for detection of birth defects at the local level with later definitive classification and coding; however, information was limited to those structural anomalies which were visible on physical examination. RESULTS: This surveillance system on birth defects provides an extensive database of infants with major and minor external structural anomalies. From 1993 to 1996, more than one million births were monitored, with more than 10 000 birth defects and 30 000 photos submitted. Among them 5 000 were identified with major birth defects. Neural tube defects, oral clefts and extremity abnormalities were the first few defects with most cases. Data were used in birth defect surveillance reports and scientific papers. CONCLUSIONS: This system can be utilized for studies on etiology, descriptive epidemiology, and identification of unusual trends which is unique with multiple advantages in the People's Republic of China.

[Case-control study on sexual coercion and related risk factors in China].

OBJECTIVE: To find out the determinants of sexual coercion among adolescent abortion seekers in China. METHODS: Case-control study was adopted. Women seeking for abortion and ever having experienced sexual coercion were taken as case group and those who had never experienced sexual coercion as control group. Case-control study was carried out in 11 large hospitals in Beijing city from January 2000 to April 2000. RESULTS: The number of subjects in case group was 512, comparing to 517 in the control group. The mean age of subjects was 20.25 years with monthly income 881.2 RMB Yuan. The difference between case group and control group in income, occupation and age showed no statistical significance. The results of the study indicated that the factors which were more likely to be related to sexual coercion include lower educational level (chi(2) = 15.27, P < 0.01, OR: 1.71), not living with parents (chi(2) = 10.18, P < 0.01, OR: 1.50), mobile nature (chi(2) = 20.60, P < 0.01, OR: 1.78), experienced battery by partner (chi(2) = 6.79, P < 0.01, OR: 2.72), abused by her partner (chi(2) = 4.22, P < 0.05, OR: 1.71) multi-partners (chi(2) = 19.08, P < 0.01, OR: 2.17), sex after being drunk (chi(2) = 34.22, P < 0.01, OR: 5.04), and larger gaps of in age between male and female partners (chi(2) = 11.04, P < 0.01, OR: 2.17). CONCLUSION: Factors as, poor-educated, not living with parents, floating population, multi-partners, the inequality between male and female were more likely to increase the risk of sexual coercion.

[High efficient expression of recombinant human platelet factor IV in Pichia pastoris].

OBJECTIVE: To express recombinant human platelet factor 4 (PF4) in methanol yeast. METHODS: PF4 cDNA amplified by PCR was cloned into the mating factor alpha (MFalpha) signal sequence, downstream the alcohol oxidase 1 (AOX 1) promoter of the pPIC9 vector. The reconstructed vector was subsequently integrated into the methanol yeast P. pastoris strain GS115. The highly expressing strain was selected and the protein sequences and their biological activities were assayed. RESULTS: The amino acid sequence of rhPF4 was the same as that of natural one. The rhPF4 can neutralize the anticoagulation effect of heparin in a dose-dependent manner. CONCLUSION: The amount of rhPF4 expressed in methanol yeast meets well the scale for manufacturing. The biological activities of the rhPF4 were the same as that of natural PF4.

Increased peritoneal membrane permeability is associated with abnormal peritoneal surface layer.

OBJECTIVES: We recently showed that the peritoneal surface layer may be an important barrier in modulating peritoneal membrane permeability. In the present study, we investigated the relationship between an increased peritoneal transport rate and the peritoneal surface layer. METHODS: Male Sprague-Dawley rats (n = 8) received intraperitoneal injections of 4.25% glucose dialysate daily for 1 week. Forty-eight hours after the last injection, a 4-hour dwell study using 25 mL 4.25% glucose dialysate was performed in each rat. The results were compared with those from control rats that received no intraperitoneal injections (n = 8). The peritoneal fluid and small-solute transport characteristics were evaluated. The peritoneal surface layer was studied using an electron microscope. The phospholipids content of the dialysate was also evaluated. RESULTS: Peritoneal fluid removal was significantly reduced in the daily injection group (30.6 +/- 1.3 mL) as compared with the control group (38.2 +/- 0.6 mL). The peritoneal fluid absorption rate and small-solute transport rate were also significantly higher in the daily injection group as compared with the control group. The amounts of phospholipids in the dialysate were significantly lower in the daily injection group--especially the quantity of phosphatidylcholine. However, lysophosphatidylcholine increased significantly in the daily injection group. Electron microscopy showed that the peritoneal surface layer was almost completely gone in the daily injection group, but that a dense and thick (average 4 microm) peritoneal surface layer was present on the top of the mesothelial cells in the control group. CONCLUSIONS: Our results suggest that daily injection of hypertonic glucose dialysate significantly increased the peritoneal transport rate. The increased peritoneal transport rate was associated with a significant reduction in the peritoneal surface layer and the phospholipids content of the dialysis effluent.

Peritoneal dialysis solutions.

After several decades of experience of peritoneal dialysis therapy, we now understand the peritoneal membrane and the causes of its changes during long-term dialysis much better. Several new, more biocompatible solutions are available in clinic today, and the outcome of peritoneal dialysis therapy is expected to be further improved. However, limitations with the currently available peritoneal dialysis solutions still exist, and continual efforts are needed to develop solutions that are more efficient and more membrane-friendly. With the better understanding of the role of fluid balance in peritoneal dialysis, we believe that development of peritoneal dialysis fluid that protects the peritoneal surface layer (and thus the integrity of the peritoneum, thereby improving peritoneal fluid removal) may be an area of research in the near future.

[Protective effects of Chinese herb-compound on cellular immunological function (correction of funcion) in tail-suspended rats].

Objective. To observe the protective effects of two kinds of Chinese herb-compounds (Dan-huang-ci compound and Shen-chuan-shu compound) on cellular immunology in tail-suspended rats. Method. The rats were divided into: 1) normal control group; 2) tail-suspended group; 3) tail-suspended + Dan-huang-ci compound; and 4) tail-suspended + Shen-chuan-shu compound. Ability of lymphocyte proliferation and production of IL-2 in rats in the four groups were compared after 21 d. Result. The immunological function of tail-suspended control group decreased significantly as compared with normal control group. Shen-chuan-shu compound could improve immunological function of tail-suspended rats obviously. Conclusion. Shen-chuan-shu compound could enhance cellular immunological function in rats under simulated weightlessness.

[The expression of BDNF and NT-3 in dorsal root ganglion following peripheral and central axotomy--an immunohistochemical study].

OBJECTIVE: To explore the expression of brain-derived neurotrophic factor (BDNF) and neurotrophin-3(NT-3) in dorsal root ganglion (DRG) following peripheral and central axotomy. METHODS: Fifteen male adult cats were divided into intact group, peripheral axotomy group and central axotomy group. The ten cats of the latter two groups were subjected to unilateral peripheral cut and central processes cut respectively. The animals were allowed to survive 4 days long; L5 DRG was taken from each animal and made into frozen sections 20 microns thick. The sections were stained using antisera of BDNF and NT-3 respectively by immunohistchemical ABC method. The neuronal numbers of BDNF and NT-3 of the three groups were counted and compared. RESULTS: The positive products of BDNF were mainly distributed in small to medium sized neurons (13-57 microns) while those of NT-3 were mainly in large sized neurons (57-100 microns). The neuronal numbers of BDNF and NT-3 in peripheral and central axotomy groups (24.1 +/- 3.5, 29.4 +/- 5.2; 19.4 +/- 2.4, 20.2 +/- 1.9) increased significantly than those of intact group (12.1 +/- 2.2, 13.3 +/- 2.9), P < 0.01. And the neuronal numbers of BDNF and NT-3 in peripheral axotomy groups were more than those of central axotomy group(P < 0.05). CONCLUSION: The results indicate that peripheral axotomy and central axotomy lead to the increase of the expression of BDNF and NT-3 in DRG neurons. And the effect of peripheral axotomy is greater than that of central axotomy.

[Investigation of the tongue squamous cell carcinoma prognostic factors].

OBJECTIVE: The aim of this study was to investigate the factors which influence the prognosis of the tongue squamous cell carcinoma (TSCC). METHODS: The Cox's proportional hazard model was used to analyze 121 cases of TSCC treated by the affiliated dental hospital of West China University of Medical Sciences in the period from 1990 to 1994. Thirteen parameters (age, sex, occupation, primary tumor site, clinical stage, tumor differentiation, clinical therapy, cervical lymph node metastasis, surgical margin status of the primary tumor, amount of blood transfusion, smoking, drinking, reconstruction during operation) were evaluated. RESULTS: The monofactor analysis showed that factors influencing the postoperative survival of TSCC included primary tumor site, the surgical margin status of the primary tumor and the cervical lymph node metastasis (P < 0.05). Multifactor analysis demonstrated that two of these thirteen relevant factors influenced the postoperative survival of TSCC independently. They are successively the surgical margin status of the primary tumor and the cervical lymph node metastasis (P < 0.05). CONCLUSION: This study suggests that it is critical for surgeons to resect the primary tumor and the metastatic cervical lymph nodes thoroughly to improve the survival rate of TSCC patients.

[The influence of the hamulotome and interplatal incision on the middle ear function].

OBJECTIVE: The aim of this study is to explore the effect of hamulotome on the eustachian tube function and verify the function of the tensor veli palatili muscle on eustachian tube. METHODS: Single-sided hamulotome were carried out in the rabbits under the direct visual. And the middle ear cavity mucosa of the rabbits was collected at different intervals and dyed with PAS and PH to observe the middle ear changes under microscope. RESULTS: Middle ear function changed obviously after the hamulotome, with increase in globular cells, submucosal edema and inflammatory cells. And mucin was not found in several cavities. CONCLUSION: Hamulotome do affect on the middle ear function and should not be damaged in order to protect the middle ear function in some patients.

[Effect of 3'-UTR of EG I from Trichoderma reesei on its gene expression in Saccharomyces cerevisiae].

Several industrial yeast are developed as ideal expression hosts for the production of the commercially useful proteins. The expression levels in yeast cells of the heterologous proteins are affected by the regulation factors of the genes themselves. The full-length cDNA coding for EG I from Trichoderma reesei, the cellulose-degrading filamentous fungus, was expressed in Saccharomyces cerevisiae H158. EG I produced by the recombinant S. cerevisiae exhibits maximal activity at 50 degrees C-60 degrees C, pH 5.0. It was observed that removal of the 3'-untranslated region (3'-UTR) from EG I cDNA resulted in no active EG I produced by recombinant yeast. RT-PCR analysis indicated that unlike the yeast cells harboring full-length EG I cDNA, there was no detectable EG I mRNA in the yeast cells harboring EG I cDNA without 3'-UTR. The data suggested that 3'-UTR is important for the expression of EG I in Saccharomyces cerevisiae.

[Cloning and expression of Trichoderma reesei endoglucanase III (EG III) gene in Saccharomyces cerevasiae].

Using Congo red-staining method, one positive clone with CMCase activity was isolated from the Trichoderma ressei cDNA gene bank constructed in Saccharomyces cerevasiae. Sequencing result showed that the 1.5 kb-length DNA fragment inserted in the recombinant plasmid encoded EG III gene from T. reesei. Enzymatic characterization of the EG III produced by recombinant S. cerevasiae was analyzed. The experimental results indicated that the optimum pH and temperature for EG III are 5.0 and 60 degrees C, respectively. The effects of secretory system components SSO 2 and SEB1 of S. cerevisiae on EG III secretion were examined. The results indicated that the amount of EG III secreted by the strain with SSO 2-overexpression was highest among the different recombinant S. cerevisiae strains, showed that SSO 2 is a rate-limiting component of the secretory machinery in the process of EG III secretion. Furthermore, the EG III expression level was increased 5.3 times by deletion. Furthermore, the EG III expression level was increased 5.3 times by deletion of the 98 bp in 5' untranslated region of eg3 mRNA sequence. This result suggested that the regulation region could exist in the 5' untranslated region of EG III mRNA, which is recognized by the gene expression related factors of S. cerevasiae.

[The change of nerve growth factor and brain derived neurotrophic factor in neurons of cerebral cortex of adult rat following local ischemia].

OBJECTIVE: To acquire knowledge about the change of nerve growth factor (NGF) and brain derived neurotrophic factor (BDNF) in neurons of cerebral cortex of adult rat following local ischemia. METHODS: Using specific antiserums of NGF and BDNF by immunohistochemical ABC method. RESULTS: NGF-like and BDNF-like immunoreactions distributed mainly in the neurons of the third and fifth layers in cerebral cortex. After local ischemia, the average gray degrees of NGF and BDNF in neurons of cerebral cortex both decreased on the operated side more than on the un-operated side. CONCLUSION: This experiment demonstrated that the levels of NGF and BDNF in neurons of cerebral cortex following ischemia were upregulated apparently, suggesting that NGF and BDNF may play an important role in the process of neurons' reaction after ischemia.

[Significance and expression of transforming growth factor beta in human nasal polyp tissue].

OBJECTIVE: To explore the pathogenesis of nasal polyposis and the expression of transforming growth factor beta (TGF-beta) in human inflammatory nasal polyps. METHODS: TGF-beta 1-3 in nasal polyp tissues and inferior turbinate mucosa of twenty-five polyposis patients were detected with immunohistochemistry alkaline phosphatase and anti-alkaline phosphatase (APAAP) method. The inferior turbinate mucosa of eight healthy volunteers were selected as control. Six polyp tissues were estimated with double immunolabeling and Western-blot analysis to compare the characterization of the TGF-beta isoforms expression and the proportion of macrophages and eosinophils in nasal polyp tissues. RESULTS: The expression of TGF-beta 1-3 in nasal polyps was significantly higher than that in nasal mucosa and indetecable in nasal mucosa from healthy volunteers; TGF-beta 1 was the main isoform detected in nasal polyps; TGF-beta positively was accompanied by numerous macrophage and eosinophil infiltration. CONCLUSIONS: TGF-beta mainly TGF-beta 1 is strongly expressed in nasal polyps and its mucosa, where it could be produced by macrophages and eosinophils. TGF-beta could induce modification of epithelium and connective tissue and therefore be involved in the pathogenesis of nasal polyposis.

[Preservative surgery of medial wall pyriform sinus cancer].

OBJECTIVE: To study the surgical methods and functional outcome of preservative surgery for medial wall pyriform sinus cancer. METHODS: Seventy-one patients with medial wall pyriform sinus cancer, who were treated surgically between 1985 and 1997, were reviewed. Of the 71 cases, 49 underwent preservative surgery, and 22 total laryngectomy. Preservative surgical procedure was defined as follows: 1. Extent of resection: Supraglottic horizontal partial laryngectomy and resection of medial wall of pyriform sinus were performed in patients without fixation of the true vocal cord, and the section extended to paraglottic space, partial lateral wall of pyriform sinus, preepigottic space, superior-posterior of thyroid cartilage in those with restrained vocal cord motility. Supracricoid hemilaryngopharyngectomy and resection of medical wall and partial lateral wall of pyriform sinus were practiced in those with fixed hemilarynx. Cricoid ring and cervical esophagus were removed partially if the pyriform sinus apex was involved. 2. Reconstruction: Larynx: the remains of epiglottis, perichondrium of thyroid cartilage, the infrahyoid muscular fascia, and the platysmal flap were utilized to restore the defects of larynx. Pyriform sinus: Suturing the remaining pharyngeal mucosa directly to cover the wound if the defect was relatively small. For the large defect produced by extended resections, the pectoralis major myocutaneous flap and deltopectoral flap were used. 3. Surgical treatment of cervical lymphaden: Sixty five out of the 71 cases (91.5%) underwent neck dissection. Of which ipsilateral neck dissection were done in 39 cases, and bilateral neck dissection in 26 cases. 4. All patients received postoperative radiotherapy with doses of 60-75 Gy. RESULTS: In the group of preservative surgery the 3- and 5-year survival rates were 63.4% and 49.6% respectively, whereas those in the total laryngectomy group were 52.4% and 42.4% respectively. Of the 49 cases with preservative surgery, 71.4% (35/49) had all laryngeal functions restored and 28.6% (14/49) partially restored. CONCLUSION: Despite the fact that the medial wall pyriform sinus cancer tends to have laryngeal invasion, preservative surgery can be practiced for the majority of the cases with the lesions entirely removed.

[The result of testing psychotic disorder patients in general hospital with Minnesota Multiphasic Personality Inventory].

OBJECTIVE: To study the psychological characteristics of patients with psychotic disorder in general hospital. METHODS: 92 male and 158 female patients with psychotic disorder were tested with Minnesota multiphasic personality inventory (MMPI). They consist of 215 cases with neurosis (hypochondriasis, depression, anxiety, obsessive-compulsive disorder, hysteria, neurasthenia); 4 cases with schizoaffective disorder; 3 cases with manic-depressive insanity. 5 cases with schizophrenia and 23 cases of somatoform disease with affective disorder. RESULTS: The scores of F, K, Hs, D, Hy, Pd, Pa, Pt and Si scales in the patients were significantly higher than those in normal controls. The scores of Mf were significantly higher in the male han those in the female patients and the scores of F lower in the male than those in the female patients. CONCLUSIONS: The psychotic disorders as well as seen in patients in general hospital are hypochondriasis and somatoform disorder, and depression of the female.
