Cholinergic amacrine cells in the rat retina.

Staining of rat retinal wholemounts with a monoclonal antibody against choline-acetyl-transferase (ChAT) reveals two matching populations of amacrine cells in pigmented and albino rat retinae. One population has cell bodies in the inner nuclear layer (INL). Their dendrites are confined to a narrow stratum in the outer half of the inner plexiform layer (IPL). The other, displaced, population has cell bodies in the ganglion cell layer (GCL) with dendrites stratifying in the middle of the IPL. The density changes with eccentricity, ranging from 1,700 cells/mm2 centrally to 600 cells/mm2 in the periphery. Presumptive cholinergic cells were filled with the fluorescent dye Lucifer yellow. Both subpopulations have the same "starburstlike" morphology as described for rabbit cholinergic amacrine cells (Famiglietti, '83; Tauchi and Masland, '84; Masland et al., '84b). Their dendritic tree sizes change with eccentricity and range from 160 to 300 microns in diameter. Counterstaining of Lucifer yellow-filled cells by ChAT immunohistochemistry did not yield an unequivocal double staining. Nevertheless, indirect evidence of same soma size, same number and form of primary dendrites, same level of stratification, and the good fit into the cholinergic mosaic makes it very likely that the "starburstlike" amacrine cells in the rat use acetylcholine as their transmitter. A comparison with the rabbit cholinergic system strengthens this assumption and reveals a striking similarity between both species.

Analysis of a glycinergic inhibitory pathway in the cat retina.

Incubation of cat retinas with 3H-glycine in vitro, followed by horizontal sectioning and autoradiography, showed labeling of 10-12% of bipolar cells and 45% of amacrine cells. To ascertain the effects of glycine-accumulating bipolar and amacrine cells on the response properties of retinal ganglion cells, in vivo iontophoretic studies were performed in the cat eye. Glycine inhibited all ganglion cells, and this action was blocked by strychnine. Aminophosphonobutyric acid (APB) suppressed ON-ganglion cells, but activated OFF-ganglion cells. The influence of APB upon OFF-ganglion cells could be completely blocked by strychnine. In the mudpuppy, APB suppressed ON-bipolar cells without affecting OFF-bipolar cells and without direct effects on other retinal neurons (Slaughter and Miller, 1981, 1985). If APB has the same mode of action in the cat retina, the present results suggest a glycinergic inhibitory input of the ON-channel upon OFF-ganglion cells.

[Long-term anticoagulation of the post-thrombotic syndrome].

Observations of the course in 20 patients who underwent a long-term anticoagulation due to postthrombotic syndromes resulted in a clinical improvement according to the degrees of severity of the GDR section phlebology as well as in an increase of the plethysmographically measurable venous regurgitation. Thus the late sequelae of the postthrombotic syndrome may possibly reduced by long-term anticoagulation. In one third of the patients despite the anticoagulant therapy a decreased antithrombin-III-value was found.

Action and localization of gamma-aminobutyric acid in the cat retina.

The effects of iontophoretically applied GABA (gamma-aminobutyric acid) and bicuculline on retinal ganglion cells were studied in the optically intact eye of the anaesthetized cat. GABA suppressed both the spontaneous activity and light-evoked discharge of all retinal ganglion cells, regardless of their type and regardless of the visual stimulus used. Bicuculline antagonized the action of iontophoretically applied GABA. Bicuculline enhanced the spontaneous activity of on-centre cells, but suppressed the spontaneous activity of most off-centre cells. The light-evoked response of on-centre cells was increased by bicuculline. A more complicated picture emerged for off-centre cells. Weak light responses were suppressed by bicuculline, but during strong light responses the initial transient phase of the response was dramatically enhanced. Amacrine cells of the inner nuclear layer and displaced amacrine cells of the ganglion cell layer were labelled, using glutamic acid decarboxylase (GAD) immunohistochemistry and [3H]muscimol uptake. GAD-positive dendrites were found throughout the inner plexiform layer and no sign of dendritic stratification was detected.

Action and localization of glycine and taurine in the cat retina.

The effects on retinal ganglion cells of iontophoretically applied glycine, taurine and strychnine were studied in the optically intact eye of the cat. Glycine and taurine suppressed the light-evoked discharge of all on-centre and off-centre brisk ganglion cells, regardless of the visual stimulus used. Strychnine blocked the action of externally applied glycine and taurine. The light-evoked response of all ganglion cells was raised by strychnine. The tonic discharge of the light response was suppressed or raised by the drugs more than the phasic response. A population of amacrine cells, which was heavily labelled by [3H]glycine, did not take up [3H]taurine. [3H]taurine was only weakly accumulated by inner nuclear layer neurones and was predominantly located in the outer retina.

[Hemostaseologic studies of maternal blood in intrauterine retardation and threatened premature labor].

Changes in coagulation of maternal blood in the case of severe gestosis, which indicated a relation to disturbed uteroplacental blood flow, gave rise to study other risk pregnancies and to do specific coagulation analyses. The following parameters were determined in 102 women showing a distinct tendency for premature labor, 20 women patients with suspicion of intrauterine growth retardation, and 18 normal pregnant women: Number of thrombocytes in whole venous blood, fibrinogen, plasminogen, reptilase time, thrombin time. Compared to the control group, a statistically significant increase in the number of thrombocytes was found only in the cohort of women showing signs of imminent premature labor during the 34th to 36th weeks of pregnancy. Both cohorts of patients showed markedly higher levels of plasminogen than the control group over the entire period of gestation studied in this investigation. All other parameters of coagulation showed no significant changes of pathological pregnancies in comparison with normal gravidas.

[Step-wise diagnosis of primary and secondary lymphedema].

The diagnosis of the lymphatic oedema is possible by means of non-invasive methods. The degree of severity of the lymphatic oedema can already be assessed within the basic diagnostics (anamnesis, clinical findings). The recognition of secondary lymphatic oedemas is important, particularly of those due to tumours of pelvic organs. Indications and contraindications of other diagnostic methods (plethysmography, dye test, computer tomography, sonography, lymphography) in diseases of the lymphatic vessels are presented.

Retinotopic scatter of optic tract fibres in the cat.

The retinal fibers in the optic tract (OT) of the cat have a roughly retinotopic arrangement. The retinotopic scatter of OT fibres at the rostral pole of the superior colliculus (SC) and at the lateral margin of the lateral geniculate nucleus (LGN) was investigated using the retrograde transport of horseradish peroxidase. Small lesions and injections were made in the tract and from the distribution of labelled ganglion cells in the retina the retinotopic precision was measured. At an eccentricity of 30 degrees along the horizontal meridian the vertical scatter was 20-30 degrees. This is similar to the precision of the map in the SC. However a comparison with the retinotopic map of the LGN shows the precision there to be much better.

[Analysis of eclampsia cases of the years 1957-1982 at the University Gynecologic Clinic in Leipzig].

Of the total of 104 873 patients who have been delivered between 1957 and 1982, 99 women (0.094 per cent) were diagnosed as having eclampsia. Maternal lethality was 4.0 per cent, and perinatal lethality was 20.8 per cent. Delivery was spontaneous in 34.3 per cent, and caesarean section, vacuum extraction, and forceps delivery were performed in 45.5 per cent, 8.1 per cent and 12.1 per cent respectively. The incidence of eclampsia decreased generally. The prognosis for both mother and fetus could be improved by modern methods of intensive care in close connection with termination of pregnancy.

[Thrombosis therapy and the prophylaxis of thrombosis recurrences in obstetrics].

IN general the therapy of thrombosis during pregnancy is carried out with heparin, in which case the initial hospital treatment may be continued later on with double subcutaneous applications of heparin in the out-patient department. Apart from coagulation-physiological parameters plethysmography and/or ultrasound Doppler method serve for the control of the course. Within the prophylaxis of recidivations in earlier thromboembolism during pregnancy regular clinical and apparative controls are performed, in which cases in the last third of the pregnancy the possibilities of the consideration work are used. Immediately pre-, intra- and postpartally heparin is injected with low dose up to the optimum phenprokoumon regulation (beginning with the 2nd day post partum). Thromboses in the anamnesis are no essential risk, when an optimum prophylaxis of thrombosis is guaranteed during pregnancy and puerperium

[Prophylaxis of thrombosis in surgical gynecology and obstetrics (author's transl)].

An account is given of possible approaches and means available today to cope with physical and medicamentous prophylaxis of thrombosis in gynecologico-obstetric practice. Literature is evaluated, and preference is given to methods which have been safely established by verifiable diagnostic techniques as being applicable to prophylaxis of thrombosis. Due consideration is given to individual inclinations to thrombosis and haemorrhage. Therefore, emphasis is laid on low doses of heparin or dextran.

[Gynaecological and obstetric operations in coagulative disorders (author's transl)].

Evaluated in this paper are 16 gynaecological operations which had been performed on 14 women for haemostatic disorders, between 1973 and 1979. Von Willebrand's disease had been recorded from ten patients, thrombocytopenia from two, and hyperfibrinolysis from another two. Haemorrhagic complications were observed in six cases, although specific haemotherapy had been applied to them. - Twelve analysable courses of labour of patients with known coagulative disorders were recorded from among those who had been under obstetric care, between 1963 and 1979. Two of the above three diseases ranked high on the list again, with von Willebrand's disease being diagnosed in six cases and thrombocytopenia in four. Hyperfibrinolysis was recorded from one patient and an unelucidated plasmatic coagulation disorder from another one. All twelve women had spontaneous deliveries without complications. Foetal death had occurred in two women with thrombocytopenia, prior to their admission to hospital. One of the deaths had been caused by umbilical cord strangulation. Secondary haemorrhage was observed in two cases, both late in the puerperium. - Diagnosis and therapy are discussed. Cases of the above kind should be treated in clinical centres staffed with experienced personnel.

[Coagulation disorders in obstetric patients at gynaecological hospital of Leipzig University, 1961 through 1978 (author's transl)].

Intrapartum coagulopathy was secured from 41 in 69,336 obstetric patients, between 1961 and 1978. Haemostatic disorders, such as v. Willebrand-Jürgens syndrome, idiopathic thrombocytopenia, plasmatic coagulation defects, and hyperfibrinolysis, had been recorded from ten patients even before parturition. The courses of pregnancy, childbirth, and puerperium of those patients are reported, with reference being made to the need for prepartum hospitalisation as well as for target-oriented substitutional therapy and non-invasive delivery control. - Acute intrapartum haemorrhagic syndrome occurred to 31 patients, among them 13 patients with loss coagulopathy. Prophylaxis and therapy of such coagulation disorders are discussed. The deaths of two mothers are analysed. Consumptive coagulopathy, triggered by typical gestational complications, was manifest in the other 18 patients. Prophylaxis and therapy of that severe obstetric coagulopathy are discussed.

Characterization of microsomal electron transport components from control, phenobarbital- and 3-methylcholanthrene-treated mice. II. Improved resolution and quantitation of major components in ammonium sulfate fractions from total liver microsomes.

Quantitation of microsomal components in ammonium sulfate fractions using a high-resolution sodium dodecyl sulfate-polyacrylamide gel electrophoresis system, and a comparison of these results with those from similar experiments on total liver microsomes has enabled us to identify and better characterize the interactions between microsomal electron transport components. It was found that: (1) phenobarbital decreased the amount of one protein component of approximately 50 000 molecular weight while increasing a component of very similar molecular weight; (2) only two proteins appeared to be associated with CO binding; (3) another protein of approximately 68 000 molecular weight, one of the glycoproteins found in liver microsomes, appears to be induced by phenobarbital pretreatment; (4) the induction of NADPH-cytochrome c reductase activity after phenobarbital pretreatment is not dependent on an increase in the known NADPH-dependent flavoprotein, but rather on the increase in some component found predominately in our most soluble sub-microsomal fraction. A very good separation of the above components was achieved by ammonium sulfate fractionation, e.g. simply on the basis of their solubility. This and the fact that the more-or-less soluble proteins were induced by phenobarbital or 3-methylcholanthrene respectively indicate that the solubility of membrane proteins plays a major role in the structure and function of microsomal membranes.
