Transposon-like sequences in extrachromosomal circular DNA from mouse thymocytes.

Small polydisperse circular (spc) DNA was isolated from mouse thymocytes and cloned into the HindIII site of lambda vector Charon 7. Fifty-six recombinants from this spc DNA library were analyzed. R repeats, which were originally found near immunoglobulin genes, were enriched in spc DNA clones relative to their representation in the chromosome. In one clone, the R sequence was linked to Bam and MIF sequences and the contiguous arrangement was truncated from both ends. In another clone, composite Bam/R and R repeats existed as a pair in inverted repeat orientation. Truncation occurred from the 5' side without affecting the 3' ends. In both clones, short direct repeats flanked the repeated sequences. The possible role of R sequences in transposition and circular formation is discussed.

Evaluation of response to dopamine in idiopathic dilated cardiomyopathy by echocardiography and thallium-201 myocardial scintigraphy.

Twenty-six patients with idiopathic dilated cardiomyopathy (DCM) underwent thallium-201 myocardial scintigraphy. Nine patients (group A) showed a perfusion defect in the interventricular septum (IVS) and 7 patients (group B) showed a defect in the left ventricular posterior wall (LVPW). Hemodynamic responses and catecholamine levels were compared between 16 patients (DCM group) and 6 control subjects (control group) following dopamine infusion (6 micrograms/Kg/min). The end-diastolic thickness of the IVS and LVPW, and the percentage wall thickening were assessed by echocardiography. Plasma dopamine and norepinephrine concentrations in the DCM group were not different from those of the control group at rest. During the dopamine infusion, however, norepinephrine increased only in the control group. There were significant differences in the thickness of the IVS among the 3 groups [7.3 +/- 1.4 mm for group A (p less than 0.001 vs control and p less than 0.05 vs group B), 8.9 +/- 1.5 mm for group B (p less than 0.01 vs control), 10.2 +/- 1.5 mm for the control group]. The percentage thickening of the IVS increased during the dopamine infusion in group B only and the thickening of the LVPW increased in group A only. Thus, in the myocardium of DCM patients with thallium perfusion defects, the degree of thickening did not change during dopamine infusion, but in the myocardium of DCM patients with normal thallium uptake the percentage thickening increased more than in the control group.

Blood glucose kinetics in whole body and mammary gland of lactating goats exposed to heat.

Whole-body and mammary kinetics of blood glucose were measured in lactating goats exposed to thermoneutral, moderate hot, and severe hot environments for 4 d. Milk yields were reduced by 3 and 13% during moderate and severe heat exposure, but heat production was unchanged during the experiment. Concentrations of plasma glucose and free fatty acids did not change during heat exposure. Concentration of thyroxine tended to decrease and concentration of prolactin increased with increasing temperature. Whole-body turnover of blood glucose decreased significantly during both moderate and severe heat exposure. Blood glucose oxidation rate and contribution of blood glucose to total carbon dioxide production were not influenced by heat exposure. Mammary glucose uptake tended to decrease during heat exposure, and this reduction may account for the decreased whole-body turnover of blood glucose. The lactose concentration in milk was decreased on the 4th d of severe heat exposure. A relatively low production of milk lactose was apparently derived from blood glucose. These results suggest that the whole-body turnover of blood glucose decreases in step with a decrease in mammary glucose uptake during heat exposure in lactating goats.

[Percutaneous epidural electrical stimulation of the spinal cord for intractable pain--with special reference to deafferentation pain].

In a study of 44 patients with different types of chronic pain, mostly associated with deafferentation, chronic percutaneous epidural spinal stimulation has proved useful treatment achieving an initial 52% incidence of pain amelioration overall. Long-term result showed at six months in 86%, at 1 year in 90%, although technical problems, which included electrode displacement and required minor operative readjustment, affected 48% of those permanently implanted. No other complications were seen. Success bore no relationship to quality of pain reported by the patients or to duration of pain. The patients with denervation caused by nerve or root lesions responded better than those with cord lesions even though electrical paresthesia were delivered to the area of pain in each case. A decline in effectiveness with time was noted in small numbers of our cases despite persistence of paresthesia in the area of pain. It is suggested that late failure reflects plasticity of the nervous system in adapting to new inputs. Morphine study was carried out in some of these patients. Morphine did not help to ameliorate the pain in many cases with deafferentation pain. And also Naloxone was administered during successful pain-relieving stimulation. This did not result in recurrence of pain. The Somato Sensory Responses were recorded in 25 patients before and during neurostimulation. When stimulation was applied the late component was suppressed in most of those who enjoyed a good result. The early component was not changed in those patients even during stimulation. These results suggest that spinal cord stimulation would suppress the denervative hypersensitivity of dorsal horn in the patients with deafferentation pain.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of secretagogues on membrane potential and input resistance of pancreatic acinar cells of sheep.

The effects of acetylcholine, short-chain fatty acids and peptide hormones on membrane potential and input resistance of the pancreatic acinar cells of sheep were investigated using intracellular glass microelectrodes. The resting membrane potential and input resistance were -28.0 +/- 4.3 mV (mean +/- SD, inside negative, n = 23) and 2.4 +/- 1.7 M omega, respectively. The application of acetylcholine, short-chain fatty acids (acetate, butyrate and caprylate) and peptide hormones (caerulein and bombesin) always caused depolarisation accompanied by a marked reduction in input resistance. The equilibrium potentials of acetylcholine and caprylate were -13.5 and -12.8 mV, respectively.
