Non-linear accumulation of gentamicin in guinea-pig kidney.

The dose dependence of gentamicin renal uptake in guinea-pigs was studied. The concentration of gentamicin in the kidneys was determined 2 h post administration. The plot of gentamicin concentration vs. dose over the range 0.3-48 mg/kg iv revealed nonlinearity of the uptake. As the delivery of gentamicin to proximal tubule cells apparently obeys first order kinetics, the nonlinear process should occur at the cellular uptake level.

Determination of ethimizol and two of its metabolites in serum by high-pressure liquid chromatography.

A liquid chromatographic assay has been developed for the determination of ethimizol and two of its metabolites in serum. The analysed compounds are preseparated from serum by a micro-column packed with an octadecylsilanized silica gel. Components of the micro-column eluate are analysed on a silica-gel-packed column by means of a high-performance liquid chromatograph fitted with a photometric detector. At 262 nm the detection limit of the injected amount of ethimizol is about 5 ng. The mass spectra of two ethimizol metabolites isolated from rat serum are presented. The suitability of the developed assay for pharmacokinetic studies of ethimizol is demonstrated.

Hepatic and plasma lysosomal enzyme activity in shock-like state following administration of polysaccharide-protein complex isolated from Candida albicans.

Polysaccharide-protein complex (PPC) derived from the cell walls of Candida albicans exerts several pathophysiological effects in laboratory animals similar to the symptoms of shock found at acute disseminated candidiasis. The present study evaluated the presumed fragility of hepatic lysosomes in rats in shock-like state following a single dose of PPC (25 mg/kg) administered intravenously, in terms of temporal changes in lysosomal enzymatic activity (cathepsin D, N-acetylglucosaminidase) in four sedimentable fractions and supernatant of liver homogenate after differential centrifugation, as well as in plasma. A reduction of sedimentable lysosomal activity (13,000 X g) was observed during the first hours (3-5 h) after administration of PPC with both lysosomal hydrolases, accompanied by a concomitant increase in the unsedimentable activity that represented the enzymatic activity released from disrupted lysosomes. Plasma levels of cathepsin D and N-acetylgucosaminidase had the same pattern of changes, reaching the maximum level 2-3 h after administration of PPC with a tendency of returning to the control levels by 12 h. Thus PPC from Candida albicans appears to have similar effects upon integrity of hepatic lysosomes as bacterial endotoxins have by labilizing the lysosomal membrane.

Radiation-induced changes in gentamicin pharmacokinetics.

Decreased clearance of i.v. or intratracheally administered gentamicin was observed in rats following single whole-body irradiation. 6 Gy 60Co. reaching its lowest rate on the 7th post-irradiation day. Simultaneously the absorption rate of gentamicin from the lungs was found to be increased.

The effect of polysaccharide-protein complex isolated from Candida albicans on regional blood flow in rats.

After i.v. administration to rats of polysaccharide-protein complex, isolated from Candida albicans, a decrease of cardiac output was observed from 20 sec to 240 min postinjection, followed by a recovery at 360 min. Concomitantly the regional blood flow was maintained in heart and lungs, moderately decreased in intestine, liver and adrenals and markedly reduced in skin, muscle, spleen and kidney.

Kinetics of 131 I-o-iodohippurate in rats.

The organ distribution of 131I-o-iodohippurate (OIH) was studied in rats in the time interval of 1--1440 min. The mean decrease of OIH in plasma was fitted with a function composed of three exponential terms. A three-compartment model describing kinetics of OIH in rats was suggested and its parameters were computed. Comparison of organ and plasma concentrations of OIH showed that the liver, up to 30 min, and the heart and lungs, up to 15 min after administration, form with plasma a common central compartment.

The distribution in mice of intravenously administered labelled Candida albicans.

The time dependence of the distribution of intravenously injected radiolabelled Candida albicans in the body of mice was studied. The Candida cells were labelled by cultivating them 7 days at 28 degrees C in a medium containing one of the following radionuclides: 46Sc, 95Nb, 59Fe, 144ce, 89Sr, 60Co, 65Zn, 54Mn, 45Ca, 51Cr and 91Y, which are listed in decreasing order in respect to amount bound. The labelled cells were killed by heating them 120 min at 60 degrees C, without loss of immunologic properties. Owing to the amount and strength of binding, 144Ce labelled Candida, together with 14C labelled cells was used in animal kinetic study. A rapid disappearance of the labelled cells from blood, interrupted by a small peak, was paralleled by a transient uptake in lungs and followed by a long lasting accumulation in the liver. The kidneys and spleen revealed only small uptakes of the labelled material.

Pharmacokinetics of gentamicin administered intratracheally to rats.

The pharmacokinetics of intratracheally instilled and intravenously injected gentamicin were compared in the rat and analyzed by a one-compartment open model. The maximum concentration of gentamicin in plasma occurred within 10 min after intratracheal instillation. Considerable amounts of gentamicin were absorbed from lungs after intratracheal instillation, as shown by its concentrations in plasma and elimination in urine. The data suggest that the absorption of gentamicin from the pulmonary system would be sufficient to maintain therapeutic levels of this agent in plasma.
