The extraction and partial purification of the deoxycholate-soluble matrix protein from human plantar horny layer.

The electron-dense, amorphous matrix protein in human horny cells was extracted with deoxycholate. The deoxycholate-soluble material was partially purified by differential centrifugation and molecular sieve chromatography. Approximately 44% of the total protein was extracted into the 270,000 g supernatant fraction. Macroaggregates with histochemical characteristics indentical with those in situ keratohyalin granules were formed upon dialysing the 270,000 g supernatant fraction against redistilled water. Antibody was raised to the purified material of the initial peak from the Sepharose 6B column. The resulting antiserum yielded a single precipitin line when tested against the immunizing antigen. Electrophoresis in the presence of sodium dodecylsulfate showed this antigenic material to be heterogeneous. It was immunologically homogeneous when this antigenic material was examined by immuno-electrophoresis. Since the antibody was localized on the keratohyalin granules of human planter skin by indirect immunofluorescence, the partially purified, deoxycholate-soluble material from human plantar horny layers is considered to originate from keratohyalin granules. Amino acid analysis revealed a high content of glycine, glutamic acid and aspartic acid, and a low content of histidine and leucine.

Polynucleotides. XXIV. Synthesis and properties of a dinucleoside monophosphate derived from uridine 6,2-cyclonucleoside.

A dinucleoside monophosphate, 6,2'-anhydro-6-oxy-1-beta-D-arabinofuranosyluracil-phosphoryl- (3'-5')-6,2'-anhydro-6-oxy-1-beta-D-arabinofuranosyluracil (I) was synthesized by the condensation reaction using DCC from 5'-monomethoxytrityl derivative(VII) and 3'-acetyl-5'-phosphate(X) of the monomer units. Yield was ca. 70%. UV of compound I showed no hypochromicity and its CD spectrum showed only a trough at 252 nm. No temperature dependency of epsilon and [Q] values was observed. These results suggested an unstacked random conformation for I. Complex formation of I with cycloadenosine oligomer could not be detected under the experimental condition.

Polynucleotides. XXVI. Complex formation of polynucleotides derived from formycin and laurusin with cyclonucleoside oligonucleotides.

Poly(formycin phosphate) and poly(laurusin phosphate) were synthesized by polymerizing formycin and laurusin 5'-diphosphate by means of E. coli polynucleotide phosphorylase. The complex formation of these polynucleotides with cyclonucleoside polynucleotides were investigated. While poly(formycin phosphate) did not form the complex with an octanucleotide of 6,2'-anhydro-6-oxy-1-beta-D-arabinofuranosyluracil, poly(laurusin phosphate) did form a 1: 1 complex with octanucleotide of 8,2'-anhydro-8-mercapto-9-beta-D-arabinofuranosyladenine in the presence of 0.15M Na ion at neutrality and 3(o). CD spectrum of this complex showed a couple of a trough at 286 nm and a peak at 262 nm. This fact suggests that the complex has a left-handed helical conformation, which is opposite to the natural double helical polynucleotides. The cause of this phenomenon was discussed in connection with the complex of cyclonucleoside oligonucleotides.

Kinetics of Activation of Nicotinamide Adenine Dinucleotide Kinase by Phytochrome-Far Red-absorbing Form.

The effects of exogenous redox cofactors and purine analogues on the activation of the NAD kinase by Pfr were examined. Addition of phenazine methosulfate, flavin mononucleotide, or methylene blue increased the activation of NAD kinase by red light in a partially purified preparation of phytochrome. Phenazine methosulfate and flavin mononucleotide do not absorb light in the red or far red region, so they do not act as light receptors in this activation. Thus they probably intervene in electron transfer between phytochrome and NAD kinase. Addition of kinetin with these compounds increased photopotentiation further. In the presence of phenazine methosulfate and kinetin, the activation of NAD kinase by red light was counteracted by illumination with far red light immediately after red light. The relation between the dose of illumination with red light and the degree of activation of NAD kinase was measured. Illumination for 5 minutes with 800 ergs cm(-2) sec(-1) of red light was sufficient to obtain maximal activation. In the dark, the activated state of NAD kinase was maintained for about 10 min and then rapidly lost. The time of preservation of the activated state was greatly shortened by illumination with far red light, indicating the participation of phytochrome.

Photoregulation of Nicotinamide Adenine Dinucleotide Kinase Activity in Cell-free Extracts.

This article gives evidence that NAD kinase activity is controlled by the action of phytochrome. The NADP level rapidly increased in the cotyledons of seedlings of Pharbitis nil strain Violet (a short day plant), when the inductive dark for flowering was interrupted with a 5-minute illumination of red light. Illumination with far red light immediately after illumination with red light counteracted partly the effect of the latter.A partially purified phytochrome preparation from the uppermost internodes including the hook of Pisum sativum var. Alaska contained NAD kinase activity, and this increased greatly on illumination with red light. The effects of red and far red light on NAD kinase activity were reversible. The Km value of NAD kinase in the phytochrome preparation for NAD was 1.84 mm in the dark and 0.90 mm under red light. A broad optimum of the NAD kinase reaction and its photopotentiation appeared in the region of pH 6.6 to 8.6.The NAD kinase activity in a phytochrome preparation from the coleoptiles of Avena sativa was also activated by red light. It was suggested that the Pfr type of phytochrome may contribute to the decrease of the Km value of NAD kinase reaction by causing a conformational alteration of NAD kinase.

The successful treatment of diffuse neonatal hemangiomatosis with flashlamp pulsed dye laser.

Diffuse neonatal hemangiomatosis is a rare disease with the distinctive features of multiple hemangiomas of the skin and visceral organs. These lesions have been treated with systemic corticosteroids, interferon-alpha, and their combination. We report a patient with diffuse neonatal hemangiomatosis who had multiple cutaneous and hepatic hemangiomas. Single therapy with the flashlamp pulsed dye laser was effective for cutaneous hemangiomas, whereas the hemangiomas of the liver remained stable and no liver dysfunction or hemorrhage has occurred so far, even with no treatment.
