Increased urine level of amino-terminal peptide derivatives of type III procollagen in patients with liver diseases.

The amino-terminal peptides of type III procollagen (PIIIP) in the urine of 40 patients with various liver diseases were determined with a commercial radioimmunoassay kit. The level of urinary PIIIP (uPIIIP) was correlated well with serum PIIIP (sPIIIP) in 9 patients, the coefficient of correlation being r = 0.836 (p less than 0.01) and the regression line being y = 1.42x + 24. Urinary PIIIP consisted of at least 4 different molecular species with molecular weights of 49 k, 18 k, 10 k and 4.6 k as estimated by column chromatography on Sephadex G-100. Furthermore. uPIIIP was found to be significantly elevated in acute hepatitis, chronic hepatitis, liver cirrhosis, hepatocellular carcinoma and other liver diseases, in which the elevation of sPIIIP has been reported by others. The mean values +/- standard deviations of uPIIIP were 44.0 +/- 32.0, 60.4 +/- 32.0, 62.0 +/- 46.5, 53.0 +/- 27.1 and 48.1 +/- 22.8 ng/ml for the respective liver diseases, and 13.2 +/- 4.5 for the non-hepatic disease group.

[Teratological test of 10% nitroglycerin (NT-1 ointment) in rabbits].

A teratological test of 10% nitroglycerin (NT-1 ointment) was carried out in New Zealand White rabbits. Pregnant rabbits were treated percutaneously with NT-1 ointment from day 6 to 18 of gestation at dose levels of 15, 60 and 240 mg/kg/day as nitroglycerin itself. All pregnant rabbits were killed on day 29 of gestation, and the influences of NT-1 ointment upon the performances of dams and fetuses were examined. During the treatment period, erythema was observed on the treated dorsal skin in all groups excluding the nontreatment control group. However, it disappeared soon after the cessation of NT-1 ointment administration. Influences of NT-1 ointment administration were not found at any dose levels on the food consumption and body weight changes of pregnant rabbits. In addition, influences of NT-1 ointment on the reproductive performance of dams and the development of fetuses were not observed at any dose levels. Therefore, the non-effect dose of NT-1 ointment on reproductive performance of dams and fetal development in rabbits was estimated to be 240 mg/kg/day and more as nitroglycerin itself.

[A study of itch intensity in relation to two-point tactual discrimination].

This study was designed to investigate whether or not the areas of the human body most sensitive to itch stimuli are identical with the body parts least sensitive to tactual discrimination, a hypothesis which arouse from our previous studies of itching. Subjects were 12 college students (six males and six females). The itching was induced by grated Japanese yam. The body areas were the upper arm (with large two-point thresholds), the lower leg (with intermediate two-point thresholds), and the middle finger (with small two-point thresholds). An experimental session consisted of a stimulus-application period (200 s), and a post-application period (600 s) during which the itching increases markedly. (After wiping off the grated yam, itching is increased because of the needle-crystals of calcium oxalate contained in the yam peel which penetrate slightly into the skin in the process of wiping.) Self-ratings on itching and the volume of dermal blood flow were recorded during two periods. The results support our hypothesis and suggest that tactile sensation has some interaction with itching.

Inhibition of liver acetyl-coenzyme-A carboxylase by 2-tetradecanylglutarate.

In a previous study (A. ENDO, et al., J. Antibiotics 38: 599 approximately 604, 1985), 2-alkyl glutarate and its derivatives isolated from cultures of Gongronella butleri were shown to inhibit animal acetyl-CoA carboxylase. In the present communication, the inhibition of liver acetyl-CoA carboxylase was investigated with several 2-alkyl glutarate and 2-alkyl succinate analogs. Their inhibitory potency increased with the chain length of the alkyl moiety, and 2-tetradecanylglutarate was most potent among the inhibitors tested. Kinetic analysis indicated that inhibition by 2-tetradecanylglutarate was non-competitive with respect to the substrates, ATP, HCO3- and acetyl-CoA, and competitive with respect to the allosteric regulatory citrate, giving a Ki value of 40 microM. Sucrose density gradient centrifugation analysis showed that the citrate-induced polymerization of the enzyme was inhibited by 2-tetradecanylglutarate.

The active immunotherapy of a 3-methylcholanthrene-induced rat tumor with Friend virus-infected (xenogenized) tumor cells.

We investigated the various conditions necessary for potentiating the immunogenicity of viable Friend virus (FV)-infected rat fibrosarcoma KMT-17 cells. The immunogenicity was most potent among cells passaged through three generations of FV-tolerant rats; a single subcutaneous immunization induced an increase in the transplantation resistance (LTD50) of 10,000 or more. It was also found that intradermal immunization of the tumor cells induced stable and potent resistance to the transplantation of non-xenogenized tumor cells soon after immunization, and that irradiated non-xenogenized tumor cells were effective in supplying booster immunization. Active immunotherapy against KMT-17 cells pre-transplanted subcutaneously was carried out on the basis of the above findings with the results that the active anti-tumor immunization alone caused a complete regression in 30% of the tumor-burdened rats. These findings indicate the possibility of achieving active immunotherapy with xenogenized tumor cells.
