Systemic senile amyloid in senescence-accelerated mice. A unique fibril protein demonstrated in tissues from various organs by the unlabeled immunoperoxidase method.

Tissue sections of senescence-accelerated mice (SAM) and of mice with normal aging characteristics (R series) were studied using a peroxidase-antiperoxidase (PAP) method with specific antisera against a unique amyloid fibril protein (AS/SAM) and against murine protein AA. Anti-AS/SAM antisera reacted with amyloid tissues of SAM and aged R series mice but not with normal tissues or amyloid tissues containing protein AA. Reactivity of this and serum could be abrogated by absorption with purified AS/SAM. Amyloid deposits in liver, kidney, spleen, gastrointestinal tract, lung, heart, gonads, pancreas, salivary glands, adrenal, thyroid, skin epineurium, and blood vessels were positive for AS/SAM in both the SAM and R series. Brain and bone medulla, however, were not stained. The amyloid deposited in gonads, papillary layer of dermis, and epineurium was exclusively AS/SAM. The amyloid observed in most of the P-1 series mice was AS/SAM. In the P-2 series and R series, protein AA frequently coexisted with AS/SAM as demonstrated using a double staining method. Each amyloid localized preferentially in the liver, spleen, and gastrointestinal tract.

[Treatment of pain caused by metastases of reactivated prostatic cancer with ifosfamide].

Ifosfamide (Z 4942) is an alkylating agent with the structure of a cyclophosphamide analog. Preliminary studies performed in our hospitals demonstrate the releasing effect of Ifosfamide for persistent pain caused by metastases of the prostatic cancer at various regions. Ifosfamide was given 2 grams a day intravenously with hydration and alkalization for consecutive 5 days. The regimen was performed every 3 or 4 weeks. Pain has disappeared in 7 of 10 cases within 2 or 3 courses. Pain of other 3 cases has also greatly reduced. The effective rate for the primary lesion of the prostatic carcinoma in stage C and D is 30.7%. Side effects were nausea, vomiting, hair loss and leukopenia.

[A case of renal lymphoma].

A 74-year-old man was admitted to our hospital, complaining of gross hematuria and complete urinary retention. Right nephrectomy and suprapubic prostatectomy were performed under general anesthesia. On the second postoperative day, acute renal failure developed and peritoneal dialysis was begun. But, unfortunately he died the next day. Pathological diagnosis was malignant lymphoma infiltrating to the perirenal and periadrenal adipose tissue.

[Prognostic analysis of malignant peritoneal effusions from gastric cancer].

The prognosis of 135 cases with malignant peritoneal effusion from gastric cancer was analyzed clinically and cytologically. The histologic type of the primary gastric cancer or therapeutic differences had no significant influence on the prognosis. Previous gastrectomy or absence of distant metastases had no marked effect on survival. The formation of large clusters of cancer cells on smear preparations of peritoneal effusion was interpreted as a sign of good prognosis. The cancer cell number and the presence of lymphocytes on smear preparations bore no relationship to prognosis.

Amino-acid sequence of a heat-stable enterotoxin produced by human enterotoxigenic Escherichia coli.

A heat-stable enterotoxin produced by a human strain of enterotoxigenic Escherichia coli was extensively purified by reverse-phase high-performance liquid chromatography. The minimum effective dose of the purified toxin to cause fluid accumulation in suckling mice was 2.5 ng. The amino acid sequence of the purified toxin was determined by Edman degradation and a combination of fast atom bombardment mass spectrometry and carboxypeptidase digestion to be Asn-Ser-Ser-Asn-Tyr-Cys-Cys-Glu-Leu-Cys-Cys-Asn-Pro-Ala-Cys-Thr-Gly-Cys-Tyr. This sequence was identical to that deduced from the nucleotide sequence encoding a human heat-stable enterotoxin, reported by Moseley et al., except for the C-terminal Tyr residue.
