Evidence of an estrogen-like effect of dehydroepiandrosterone on lysyl oxidase activity in the mouse cervix.

The effect of dehydroepiandrosterone (DHA) on the lysyl oxidase activity in the mouse cervix was studied. Injection of animals ovariectomized 10 days before with 1 mg DHA was followed 24 h later by an increase in enzyme activity, which remained unaltered for 48 h, returning to control values by 96 h after injection. At least 100 micrograms of DHA was required to increase enzyme activity; the maximum effect occurred with 1 mg of the steroid and was comparable to that of 1 microgram estradiol-17 beta. These results demonstrate an estrogen-like effect of DHA on lysyl oxidase activity in mouse cervix. The onset of response was later and the duration of response was longer for DHA than for estradiol-17 beta.

Activated macrophage hybridomas secreting a cytotoxic factor.

Stable activated macrophage hybridomas were generated by somatic cell fusion between Propionibacterium acnes-induced peritoneal exudate cells and NS-1 myeloma cells. Five cell lines were obtained and each was cloned by limiting dilution; 59 clones were obtained. The cells of 2 clones (MP4-4 and MP4-8) which adhered to the culture dishes were selected for further analysis. These hybridomas exhibited non-specific esterase and beta-galactosidase intracellularly, and asialo GM1, Mac-1, Ia antigens and Fc-receptors on their cell surface. They did not, however, show phagocytic activity or secrete lysozyme. These hybridomas (MP4-4 and MP4-8) secreted the cytotoxic factor without any stimulation. Furthermore strong cytotoxic activity was found in ascites and sera from nude mice inoculated with these hybridomas. These activated macrophage hybridomas should be very useful in studies on cancer immunology and the physiology of macrophages.

Circulating atrial natriuretic polypeptide during exercise in patients with essential hypertension.

We studied the relationship between haemodynamic measurements and plasma atrial natriuretic peptide (ANP) levels during exercise in patients with essential hypertension. The exercise protocol consisted of three fixed work loads (25, 50 and 75 W) using a supine bicycle ergometer. Plasma ANP levels and haemodynamic indices were measured before and during each stage of exercise. Plasma ANP levels and pulmonary artery wedge pressure (PAWP) significantly increased at the maximum work load. Significant correlations of PAWP and mean arterial pressure (MAP) to plasma ANP levels were observed at four points obtained before and during each stage of exercise (PAWP, r = 0.83, P less than 0.001; MAP, r = 0.48, P less than 0.01). Furthermore, a significant inverse correlation between shortening fraction (SF), estimated by M-mode echocardiography, and plasma ANP levels at 75 W work load was also observed (r = -0.66, P less than 0.01). These results suggest that the increase of circulating ANP levels during exercise in essential hypertensive subjects may be associated with elevated left atrial pressure and arterial pressure, or left ventricular systolic dysfunction.

Clinical validity of washout time constant images obtained by digital subtraction angiography.

Functional images of left ventricular myocardial perfusion were obtained using the washout time constant obtained from the analysis of digital subtraction angiograms (DSA). The results were compared with those of left ventriculography to evaluate its clinical validity. DSA examinations were performed in eight patients with old anterior myocardial infarction and in 10 control subjects. Washout time constant images of the left ventricular wall were nearly homogeneous in normal cases. On the contrary, regional heterogeneity on the washout time constant images was observed in cases of anterior infarction. The abnormal region in the washout time constant image corresponded well to the area of abnormal percent wall thickening, whereas the extent of the abnormal wall motion area tended to be broader than that of the abnormal washout time constant area or area of abnormal percent wall thickening. Thus, the washout time constant images obtained by DSA may comprise a reliable means of estimating the extent of ischemia in the myocardium.

[Left ventricular wall motion abnormalities evaluated by factor analysis as compared with Fourier analysis].

Factor analysis was applied to multigated cardiac pool scintigraphy to evaluate its ability to detect left ventricular wall motion abnormalities in 35 patients with old myocardial infarction (MI), and in 12 control cases with normal left ventriculography. All cases were also evaluated by conventional Fourier analysis. In most cases with normal left ventriculography, the ventricular and atrial factors were extracted by factor analysis. In cases with MI, the third factor was obtained in the left ventricle corresponding to wall motion abnormality. Each case was scored according to the coincidence of findings of ventriculography and those of factor analysis or Fourier analysis. Scores were recorded for three items; the existence, location, and degree of asynergy. In cases of MI, the detection rate of asynergy was 94% by factor analysis, 83% by Fourier analysis, and the agreement in respect to location was 71% and 66%, respectively. Factor analysis had higher scores than Fourier analysis, but this was not significant. The interobserver error of factor analysis was less than that of Fourier analysis. Factor analysis can display locations and dynamic motion curves of asynergy, and it is regarded as a useful method for detecting and evaluating left ventricular wall motion abnormalities.

Digital subtraction angiography: image-sequence analysis for regional myocardial perfusion dynamics.

Digital subtraction angiography with selective coronary injections of contrast media has enabled us to obtain clear images, not only of the artery, but of the capillary and venous phases of the myocardial perfusion. In the present study, densitometry was used to estimate regional myocardial perfusion dynamics in 10 control cases and 11 anterior myocardial infarction cases. The time density curve showed that contrast material increased rapidly in the arterial phase and appeared to be washed out monoexponentially in the venous phase. The time from the onset of contrast medium injection to the maximal density of the contrast medium (Tp), and the time constant obtained from the washout curve (Tc) were analyzed. In the control group, Tp in the apical region was slightly prolonged as compared with Tp in the anterobasal region, but the difference was not significant (5.2 +/- 0.5 vs 4.2 +/- 0.4 sec: mean +/- SEM). Tc did not definitely change in any portion of the myocardium (anterobasal 5.1 +/- 0.5, anterior 4.8 +/- 0.5, apex 4.6 +/- 0.5 sec, respectively). In anterior myocardial infarction, Tp in the marginal region was significantly prolonged compared to Tp in the control region (6.0 +/- 0.3 vs 4.7 +/- 0.3 sec, p less than 0.01). Tp was prolonged for more than 10 sec in the infarcted region. Tc in the marginal region was markedly prolonged compared to Tc in the control region (7.4 +/- 0.9 vs 4.4 +/- 0.5 sec, p less than 0.025). Tc could not be determined in the infarcted regions because data acquisition time of our apparatus was inadequate.(ABSTRACT TRUNCATED AT 250 WORDS)

Induction of tumor necrosis factor by administration of OK-432 in cancer patients.

We examined whether OK-432, a streptococcal preparation, could induce tumor necrosis factor in cancer patients. OK-432 was administered at a dose of 100 KE intratumorally to 4 advanced gastric cancer patients and 10KE intracavitary to 8 patients with malignant pleuroperitoneal effusion. The cytostatic activity of the sera and malignant effusions was assayed by the growth inhibition of L929 cells. OK-432 induced significant cytostatic activity in the sera and malignant effusions. The activity was partially neutralized by the monoclonal antibody against human recombinant tumor necrosis factor. These data suggest that OK-432 induces tumor necrosis factor in the sera and malignant effusions of cancer patients.

[Clinical studies on the biliary excretion of mezlocillin, especially in cases with liver dysfunction].

To investigate efficacy of mezlocillin (MZPC) in the treatment of biliary tract infection, the time course concentrations of MZPC in the bile of patients with, in particular, liver dysfunction were measured. MZPC concentrations in the bile decreased with the increase in the severity of liver dysfunction. However, the bile concentration was maintained more than 50 micrograms/ml even in cases of severe cholangitis with obstructive jaundice. These results indicate that MZPC is an useful antibiotic for the treatment of biliary tract infection.

[Childhood liposarcoma of the eyelid--a case report].

An 11-year-old girl noted a small mass on the left upper eyelid. The tumor was excised and diagnosed as round cell-type liposarcoma. In spite of intensive multimodal therapy, the tumor tended to recur, metastasizing to the central nervous system and soft tissues. The patient died of progressive disease three years after diagnosis. Features unique to this case include the ophthalmological primary site, the age at onset, the histological subtype and the mode of metastasis.

[Intra-arterial chemotherapy of soft tissue sarcoma in children].

As to the treatment of soft tissue tumor in children, we have performed intraarterial infusion chemotherapy in 3 children, two of whom had rhabdomyosarcoma and one of whom had infantile fibrosarcoma, at Sapporo National Hospital. The former two patients had large and unresectable solid tumor. In the latter case, an uncommon disease of childhood, local excision was considered while sacrificing a significant degree of function in the affected part. We describe here these three cases and the effectiveness of this therapy. Intraarterial chemotherapy reduced the tumor remarkably and facilitated second-look surgery. Although transient hematuria, slight articular contracture and vasculitis were observed in each case as side effects during the therapy, they were only minimal and tolerable. Unfortunately, one patient relapsed 6 months after surgery, but two patients are still in good health without recurrence. According to the results of our study, we believe that this method can be effective for controlling unresectable primary malignant soft tissue tumor.
