Maintenance of rat liver viability enhanced by cold flushing with oxygenated perfluorochemicals.

The possible benefit of oxygenation during initial cold flushing was investigated as means of improving the quality of live preservation in rats. In five group of animals (total 61 experiments), the lives were flushed with different perfusates. Non-oxygenated groups included controls, Collins' solution alone and Collins' solution containing perfluorotributylamine (FC-43). In the oxygenated groups, Collins' solution alone and collins' solution containing FC-43 were oxygenated by bubbling. The hepatic ATP level and histopathological changes were used to assess the quality of liver preservation. Oxygenation during the initial cooling process proved to be effective in maintaining energy metabolism and preventing the characteristic microscopic changes of ischemic damage. Oxygenated Collins' solution containing FC-43 showed a much longer lasting effect compared with oxygenated Collins' solution alone. Without FC-43. Under light microscopy, in integrity of the liver appeared to be well preserved up to eight hours with the former solution. It is concluded that enhanced oxygenation with FC-43 in the initial cold flushing period can improve the quality of liver preservation.

A simple method for the isolation of flagellar shape mutants in Salmonella.

During prolonged static cultivation of liquid cultures of Salmonella strains with normal flagella, flagellar shape mutants occurred spontaneously and their proportion in the cultures increased so that they were easily isolated by spreading dilutions of the cultures on semisolid medium. By this method various types of flagellar shape mutants, including curly, straight and polymorphous mutants, were obtained.

Anticardiolipin antibody-induced sudden profound sensorineural hearing loss.

INTRODUCTION: A link between sensorineural hearing loss (SNHL) and autoimmune disease is postulated. The association of SNHL in a patient with systemic lupus erythematosus is reported. METHODS: An adolescent female with left lateral medullary syndrome and right internuclear ophthalmoplegia was diagnosed with systemic lupus erythematosus. Treatment with prednisolone markedly improved her symptoms. Two years later, profound SNHL developed in the right ear. Extensive serologic testing was undertaken. RESULTS: Serologic tests for syphilis were false-positive. Enzyme-linked immunosorbent assay to the immunoglobulin (Ig)G anticardiolipin antibody was positive. The anticardiolipin antibody is strongly correlated with episodes of recurrent venous or arterial thrombosis. CONCLUSIONS: It appears that the anticardiolipin antibody may be associated with sudden profound sensorineural hearing loss in patients with autoimmune disease.

Effect of peripheral administration of arginine vasotocin on neonatal sleep in rats.

Vasotocin was recently reported to increase the neonatal amount of active sleep in kittens. In this study we examined the effects of arginine vasotocin (vasotocin), given intraperitoneally and at a wide dose range, on the sleep-wake behavior of 7-day-old and 14-day-old rat pups using a static charge sensitive recording system. It increased the percentage of quiet state relative to total sleep time at the doses of 0.01-1.0 ng/g of body weight but did not affect it at the dose of 0.001 ng/g. The percentage of active sleep was decreased by doses of 0.01-10.0 ng/g. The increase in the amount of quiet state and the concurrent suppression of active sleep lasted for 6 hours. Wakefulness was slightly increased at 10 ng/g of vasotocin. These findings indicate that, in contrast to the reports on kittens, in newborn rats peripheral administration of vasotocin increases the amount of quiet state relative to total sleep time and decreases that of active sleep.
