Clonal nature of murine hemopoietic blast cell colonies.

Murine hemopoietic blast cell colonies obtained from spleen cells of 5-fluorouracil (5-FU)-treated mice give rise to many multilineage colonies including granulocyte - erythrocyte - macrophage - megakaryocyte (GEMM) colonies in secondary cultures. Progenitor cells for blast cell colonies are considered to be more primitive than colony forming units (CFU)-GEMM. To determine whether they are clonal, we examined the phosphoglycerate kinase-1 (PGK-1) isozyme type of colonies originally grown from spleen cells of 5-FU-treated mice which had PGK-1 isozyme mosaicism. PGK assays of whole secondary colonies derived from one blast cell colony showed that they were either of type A or type B but not both. These results suggest that murine hemopoietic blast cell colonies are clonal.

Simultaneous determination of urinary creatinine and metabolites of toluene, xylene, styrene, ethylbenzene and phenol by automated high performance liquid chromatography.

An attempt was made to establish a method for the direct determination of urinary concentrations of creatinine, hippuric acid, methylhippuric acid and mandelic acid by automated high performance liquid chromatography (HPLC). Urine was diluted with distilled water or mobile phase, then the mixture was centrifuged and the supernatant was injected into HPLC. A stainless-steel column packed with octadecyl silanized silicate was used, and the mobile phase was a solution of [20 mM potassium phosphate monobasic containing 3 mM sodium 1-decanesulfonate]/acetonitrile (85/15). Another HPLC method for the determination of urinary concentration of phenol, metabolites of benzene and/or phenol is also described. Phenyl sulfate and phenyl glucuronide in urine were hydrolyzed enzymatically into phenol. The hydrolyzed mixture was injected into HPLC with the ODS column. The mobile phase was a solution of [20 mM potassium phosphate monobasic containing 1 mM sodium 1-decanesulfonate]/acetonitrile (85/15). The ratio of hippuric acid (HA) concentration to creatinine concentration determined by the urine of students after physical exercise was similar to that before exercise. Moreover, the coefficient of correlation found between the toluene concentration in a workshop and the HA concentration in workers' urine, corrected for creatinine, was higher than that obtained between the toluene concentration and the uncorrected HA concentration. For assays on stored urine samples, urine was spotted on filter paper, dried and kept several weeks, and then MA, HA, o-MHA, m-MHA and creatinine in the filter paper were eluted with 50% methanol and their concentrations determined by HPLC.

Percutaneous drainage for post-traumatic hepatic abscess in children under ultrasound imaging.

Hepatic abscess is rare in children. A patient with a post-traumatic hepatic abscess was successfully treated by ultrasound-guided percutaneous drainage. Although he underwent a drainage operation for the perihepatic abscesses, high fever continued. It was then considered that the traumatic hematoma had been infected and had become an abscess. Sonographically-guided percutaneous aspiration is useful for the definite diagnosis of hepatic abscess. Subsequent percutaneous drainage under ultrasound imaging is a safe and useful therapeutic method even in a poor-risk patient.

Technetium-99m pyrophosphate tomogram of nontransmural myocardial infarction: a case report.

Single-photon emission computed tomography (SPECT) with 99mTc-pyrophosphate (99mTc-PYP) has been reported to be useful in the detection and localization of acute nontransmural infarctions. Localized uptake of 99mTc-PYP has been shown in patients with nontransmural infarction in these studies. It is likely, however, that anatomically transmural infarctions could be mislabelled as nontransmural infarctions, since the absence of new abnormal Q waves has been used in differentiating between nontransmural and transmural infarctions. We report a case of relatively diffuse nontransmural infarction demonstrated by a semicircle of 99mTc-PYP activity which significantly overlapped 201Tl uptake on the SPECT study.

Effects of silver ion on the calcium-induced calcium release channel in isolated sarcoplasmic reticulum.

Ag+-induced Ca2+ release in isolated sarcoplasmic reticulum (SR) was studied by the stopped flow method monitoring chlortetracycline fluorescence change. After improving the experimental procedure, the initial rate of Ca2+ release could be determined more precisely than before. Micromolar concentrations of Ag+ specifically enhanced Ca2+ efflux from heavy fraction of SR vesicles (HSR). This specific effect was referred to as Ag+-induced calcium release. The Ag+-induced Ca2+ efflux was activated by caffeine and ATP, but was inhibited by Mg2+ and procaine. Further, Ag+ enhanced the Ca2+-induced Ca2+ release over the whole range of Ca2+ concentrations, similarly to ATP. Parallel to Ca2+ efflux, Mg2+ efflux, measured by the same method, was also activated by Ag+. Choline permeability determined by the light scattering method was also activated by Ag+. The results suggest that Ag+ binds to the activation site of the Ca2+-induced Ca2+ release channel and opens the channel. The Ag+ binding site is different from the Ca2+ binding site but similar to the ATP binding site.

Calcium release from isolated sarcoplasmic reticulum due to 4,4'-dithiodipyridine.

The effects of SH reagents on Ca2+ release from sarcoplasmic reticulum (SR) vesicles were examined by the tracer method using 45Ca2+. Among the various SH reagents tested, 4,4'-dithiodipyridine (PDS) was found to induce Ca2+ release most specifically from the heavy fraction of SR vesicles. Further, the following results were obtained. (i) PDS bound covalently to proteins in the SR membrane and induced Ca2+ release. (ii) The Ca2+ release was further enhanced by ATP and caffeine, but inhibited by procaine, ruthenium red and various divalent cations. (iii) PDS enhanced the Ca2+ release in the whole range of Ca2+ concentrations tested. (iv) Choline permeability was also enhanced by PDS. Further, the electrical conductance of the Ca2+-induced Ca2+ release channels was studied by incorporating them into lipid bilayers and it was found that PDS increased the probability of opening of the channels. These results suggest that PDS binds to certain SH groups of the Ca2+-induced Ca2+ release channels in the SR membrane and thus induces Ca2+ release.

Nerve root infiltration in the diagnosis of radicular pain.

Clinical and standard radiographic evaluation of patients with lumbosacral radicular symptoms may, on occasion, fail to delineate a cause. This study retrospectively reviews 62 patients who had undergone nerve root infiltration (NRI) and assesses the accuracy and indications for this diagnostic study. Surgical exploration of patients with a Group 1 response (typical pain reproduced by needle placement and then relieved by NRI) confirmed local root pathology in all. Exclusive of patients with arachnoiditis, a Group 1 response showed 85% accuracy in identifying a single symptomatic root. A Group 2 response (typical pain reproduced by needle placement but not relieved by local anesthesia) indicated multiple root involvement. Patients with a Group 3 or Group 4 response (typical pain not reproduced by needle insertion, with or without relief of pain by local anesthesia) were seldom relieved of radicular pain. NRI was most useful in investigation of patients with radicular symptoms in whom other investigations were 1) normal, 2) showed multiple level involvement, or 3) were difficult to interpret because of previous surgery.

"Osmiophilic elastolysis" of peripheral organ arteries in patients with Marfan's syndrome.

Electron microscopic examinations were performed on systemic muscle-type arteries from two teen-age girls with Marfan's syndrome. In the peripheral organ arteries, the internal elastic lamella revealed a feature tentatively termed "osmiophilic elastolysis". The lytic changes appeared segmentally in the form of electron-dense fragments, and were partly absorbed. Regenerated elastin had a normal structure and surrounded the elastolytic lesions. The same changes were observed in the newly formed small areas of elastin in the thickened intima. The remaining elastic lamella in the affected region contained numerous fine electron-dense granules. Intercellular substances, including collagen fibrils and proteoglycan, other than elastic fibers, were increased secondarily in the elastolytic lesions without any evidence of ultrastructural abnormality. Medical smooth muscle cells making contact with the elastolytic lesions were necrosed and absorbed, and smooth muscle cells in a state of synthesis were increased in the adjacent areas. Abnormal elastogenesis, which showed no remarkable changes macroscopically or light microscopically, was systemic but segmental in the peripheral organ arteries, and regeneration of the structurally normal elastin was concurrent with elastolysis. These events were diagnostic for Marfan's syndrome, and cannot be explained by the conventional theory of elastogenetic enzymic deficiency.

Effects of PSK on resistance to bacterial infection in splenectomized mice.

Nontumor-bearing C3H/He mice were splenectomized and intravenously inoculated 7 days later with Streptococcus pneumoniae, Pseudomonas aeruginosa, or Escherichia coli. The survival rate was reduced by splenectomy in the animals inoculated with S. pneumoniae, but did not change in those inoculated with P. aeruginosa or E. coli. When splenectomy was performed 2 days after transplantation of X5563, and bacteria were inoculated 7 days after the operation, the survival rate was reduced even in those inoculated with P. aeruginosa or E. coli, and elimination of the bacteria from the blood and liver was delayed. This reduction in resistance to infection was alleviated by oral administration of PSK after the splenectomy.

Copper metabolism in the macular mutant mouse: an animal model of Menkes's kinky-hair disease.

The tissue copper contents were measured in mutant (hemizygous macular male and homozygous macular female), heterozygous macular female and normal mice. The copper content in kidney and small intestine from 7-day-old mutant and heterozygote were extremely high compared to normal mice, whereas the copper content in other tissues (liver, brain, spleen and serum) was low. Copper content in whole body of mutant mice was extremely low at three stages (18 days gestation, 1 day old, and 7 days old) compared to normal mice with the exception of the mutant fetus at 14 days of gestation. Renal copper contents in the mutant fetus at 18 days of gestation and in the 1-day-old mutant were not changed compared to normal mice, whereas that in 7- and 14-day-old mutant mice was significantly elevated. Hepatic copper content of the mutant mice was extremely low at all stages compared to normal mice. Copper therapy was applied to 7-day-old mutant mice. At 1 day after injection, hepatic copper content in the mutants had increased slightly in comparison with the normal control mice, whereas renal copper content was extremely increased. At 7 days after injection, hepatic copper content in the mutants was decreased greatly in comparison with normal control mice, whereas an increase in renal copper content had remained.

The blood vascular architecture of the extra-adrenal chromaffin body in the adult rat: a scanning electron microscopic study of corrosion casts.

A well developed extra-adrenal chromaffin body with an axis of 200-400 micron was found in seven out of thirty adult male Wistar rats under a stereomicroscope. All seven bodies were located between the left and right kidneys. Blood vascular beds of the five bodies were reproduced with a methacrylate casting medium and observed with a scanning electron microscope. It was revealed that the extra-adrenal chromaffin body contained remarkedly numerous capillaries, which anastomosed with each other to form a conglomerated network. The blood capillaries were of small and uniform caliber and did not represent swollen sinusoids as in the adrenal medulla. The capillary network was denser than that in the adrenal medulla and had no direct vascular linkage with the adrenal cortex or an extra-adrenal cortical body. Histological examination of the two bodies treated with dichromate containing fixatives confirmed that they mainly consisted of chromaffin cells. These findings suggest that in the rat, extra-adrenal chromaffin bodies survive throughout life, actively producing catecholamines.

Collagen fibrillar networks as skeletal frameworks: a demonstration by cell-maceration/scanning electron microscope method.

A cell-maceration/scanning electron microscope (SEM) method was employed to demonstrate the arrangement of the collagen fibrillar network of various tissues. Immersion of fixed tissues in NaOH (25 degrees C) for 3-7 days, followed by rinsing in distilled water successfully removed the cellular elements, exposing collagen fibrils which were identified as such by transmission electron microscopy in their natural locations. SEM observations of the preparations are able to demonstrate the three-dimensional architecture of collagen fibrils much more precisely than other methods, including the silver impregnation method. Collagen fibrils, forming sheaths for housing individual cardiac myocytes, fused together, thus ensuring an equal stretch of contiguous myocytes and preventing the slippage of adjacent cells. Individual skeletal muscle fibers and nerve fibers were ensheathed by the meshwork of collagen fibrils running in two opposite helices. Such structures seem to play an important role in resisting the stretching impetus. At the epithelial-connective tissue junction of the tongue and fingertip skin, interwoven collagen fibrils formed numerous microridges which probably provide a broad anchorage for the epithelium. In the intestinal mucosa, the collagen fibrillar network immediately below the basal laminae of the villous epithelium possessed heterogeneous pores. As the collagen fibrillar network shows morphological features specific to individual organs and tissues, it is suggested that such formations not only constitute the skeletal framework but also provide those cells which are housed there with a microenvironment suitable for their activities.

Blood vascular architecture of the rat extra-adrenal cortical body: a scanning electron microscopic study of corrosion casts.

A well developed extra-adrenal cortical body with an axis of 200-700 micron was found in eighteen of thirty adult male Wistar rats under a stereomicroscope. All eighteen bodies were located between the kidneys. Blood vascular beds of sixteen of the eighteen bodies were reproduced with a methacrylate casting medium and observed with a scanning electron microscope. The extra-adrenal cortical body was found to contain remarkably numerous capillaries which anastomosed with each other to form a conglomerated network; it received one afferent vessel and issued one to three (usually, one) efferent vessels. The blood capillaries were of sinusoidal and uniform calibers, and resembled those of the adrenal cortex. The network with an axis of 300-700 micron possessed a typical, deep efferent rootlet which corresponds to the central vein of the adrenal gland. Histological examination of the two bodies treated with Orth's or Helley's fixative confirmed that they consisted of non-chromaffin cells similar to those of the adrenal cortex. These findings suggest that in the rat, the extra-adrenal cortical bodies persist throughout life, actively producing cortical hormones.

The blood vascular wreath of rat ovarian follicle, with special reference to its changes in ovulation and luteinization: a scanning electron microscopic study of corrosion casts.

Blood vascular casts of rat ovaries were prepared by injection of a low viscosity methacrylate medium, and observed by scanning electron microscopy. The proper vascular pattern of the rat ovarian follicle starts as a basket-like wreath of fine capillaries around the primary follicle. As the follicle grows larger, the wreath becomes more developed. The fully developed wreath around the Graafian follicle consists of sinusoidal capillaries. In ovulation, the apical area of the wreath is opened and the basal area of the wreath is elevated. After ovulation, the wreath shrinks and its opened area is closed, with markedly dilated vessels directly continuous with the arterial capillaries. The wreath then transforms, by intense new formation of capillaries, into a conglomerated sinusoidal capillary plexus with an avascular area within it. The avascular area soon disappears by continued intense formation of capillaries, in the plexus, and a complete and dense vascular network of the corpus luteum is formed. The capillaries of the fully developed corpus luteum are small in caliber and not sinusoidal in nature. Discontinuities of the capillaries and a flattening of the efferent branches are the initial signs of the degeneration of the corpus luteum. The small markedly degenerated corpus luteum contains only scattered thready capillaries. The corpus albicans contains few blood vessels.

The blood vascular architecture of the rat pineal gland: a scanning electron microscopic study of corrosion casts.

The blood vascular bed of the rat pineal gland was reproduced through a low viscosity methacrylate casting medium and observed with a scanning electron microscope. The pineal gland was located in front of the confluens sinuum which was formed by the confluence of the left and right transverse sinuses and the superior sagittal and great cerebral veins. The pineal gland was found to contain a rich vascular network of freely anastomosing capillaries. This network received a few afferent arteries from the posterior cerebral arteries while emitting ten to fifteen efferent veins. Most of these efferent veins drained into the upper segment of the great cerebral vein; the remaining one or two efferent veins continued directly into the confluens sinuum. A marked constriction, probably representing a venous valve or valve-like projection, was observed in the opening area of the great cerebral vein. Circular constrictions, probably representing sphincters, were imprinted in the efferent vessels. No direct vascular connection was noted between the pineal gland and the nervous tissues or between the pineal gland and the choroid plexuses of the third and lateral ventricles.
