[The localization and production of interleukin-1 in pharyngeal epithelium].

Interleukin-1 (IL-1) has been thought to be one of the essential cytokines mainly produced by macrophage. The function of IL-1 are not only activates T-lymphocytes, but also regulates immune responses to several antigens and effects on local and systemic immune reactions. Recently, it is reported that epidermal keratinocytes produces IL-1, and attention has been paid to local immune reaction mediated with this cytokine. In this study, we performed immunohistochemistry and immunoblotting on the pharyngeal epithelial keratinocytes and we evaluated the production of IL-1 by these cells. Pharyngeal mucous membrane of each region reacted strongly against anti-IL-1 alpha antibody, on the contrary, anti-IL-1 beta antibody comparatively only weakly. Strong reactivity was often observed on the basal layer, then it was seemed that some important relationship exists between the production of IL-1 and maturation of pharyngeal keratinocytes. Infiltration of T-lymphocytes was observed in proportion to the increase of epithelial reactivity against IL-1. There seemed to be no correlation between expression of HLA-DR antigen on keratinocytes and reactivity of IL-1. In immunoblotting study, the 31kD band, intracellular immature molecule, was identified besides 17 and 52kD. From this finding, it was suggested that IL-1 is derived from pharyngeal keratinocytes. In conclusion, the present study is helpful for well-understanding of local cellular immunity of pharyngeal mucous membrane.

[A case of left atrial giant thrombus which appeared in a short period].

We reported a case with left atrial giant thrombus (LAGT) which appeared over a period of six months. The patient was a 65-year-old woman who came to our hospital complaining of syncopal attack. Her electrocardiogram showed atrial fibrillation with very slow ventricular response, and there was cardiac arrest for 1500 ms, mitral stenosis and regurgitation, aortic and tricuspid regurgitations were recognized in the echocardiogram. By coronary angiography, 90% stenosis in the left circumflex artery (LCX) was found, and using transseptal left atrium heart catheterization, pressure gradient from the left atrium to the left ventricle was also measured. We diagnosed that her valvular diseases were mild. To protect her from unconsciousness and heart failure, a permanent pacemaker was implanted and PTCA for LCX lesion was performed. After six months, we found LAGT by echocardiography. It is very rare that LAGT is formed in a short period. We suggested that the changes of hemodynamics caused by transseptal left atrial catheterization or the permanent pacemaker implantation effected the formation of the LAGT.

[The step to improve the operative outcome in patients with severe valvular heart disease].

In an effort to improve the operative outcome in severe valvular heart disease, we investigated the effects of low-dose infusion of dopamine and dobutamine up to the day of operation. New York Heart Association Functional Class was improved significantly before operation, as compared with at the time of admission. It was possible to perform operation on individual patients when their physical condition seemed to be at their best. In patients with fear of postoperative renal failure, hemodialysis was combined during cardiac surgery to prevent failing to renal dysfunction after operation. As the result of these procedures, we were able to obtain satisfactory result. In patients with severe valvular heart disease, the preoperative combined infusion of dopamine and dobutamine, and hemodialysis during cardiac surgery to prevent postoperative renal dysfunction seemed to improve the operative outcome.

Pulpal and cutaneous inputs to somatosensory neurons in the parabrachial area of the cat.

The majority of somatosensory neurons recorded from the mesencephalic parabrachial area and pontine parabrachial nucleus of the cat responded exclusively to noxious mechanical stimuli to the skin. Their receptive fields were very large. Two-thirds of the neurons tested responded to electrical stimulation of the tooth pulp. These results suggest that neurons in this area have extensive convergence of spinal and trigeminal inputs, and contribute to the affective or autonomic aspects of pain.

Cell cycle-regulated gene expression in transgenic plant cells.

A majority of histone genes are expressed in the S phase during the cell cycle. Using the gene expression system of transformed sunflower cells into which wheat histone H3 gene was introduced by the Ti-plasmid gene transfer technique, we determined three cis-acting control sequences (hexameric, octameric, and nonameric motifs) which seemed to confer the S-phase-specific transcription of wheat histone genes. Furthermore, as candidates for regulatory transcription factors, three nuclear DNA-binding proteins HBP-1a, HBP-1b, and HBP-2 that interact with the hexameric and nonameric motifs were identified. The structural analysis of the cDNA of HBP-1a revealed that a nuclear protein has the leucine-zipper structure and a DNA-binding motif. The hexameric motif in the H3 gene was also seen in cauliflower mosaic virus 35S (CaMV 35S) promoter and shown to function as a regulatory element of this promoter. The wheat HBP-1b can interact with the hexameric motif of the CaMV 35S promoter. Much attention has been paid to the significance of the hexameric sequences within the H3 and CaMV 35S promoters and the DNA-binding proteins HBP-1a and HBP-1b.

Properties of an acid cholesteryl ester hydrolase inhibitor from rat serum.

The inhibitory effect of a protein isolated from rat serum on lysosomal acid cholesteryl ester hydrolase (acid CEH; EC.3.1.1.13) activity was studied. An inhibitor was purified from rat serum following ultracentrifugation and heat treatment using column chromatography on Sephacryl S-200 and ultrafiltration. The purified inhibitor appeared as a single protein band in sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. The molecular weight of the inhibitor was 28,000 Daltons as judged by gel filtration on Sephacryl S-200 and SDS-polyacrylamide gel electrophoresis. The purified inhibitor was shown to be apolipoprotein A-I (apo A-I), the major apolipoprotein of high-density lipoprotein (HDL), using immunoprecipitation with rat anti-apo A-I immunoglobulin (Ig)G. Inhibition of acid CEH activity by apo A-I was dependent on the concentration of apo A-I. The values of Vmax obtained were similar with or without apo A-I. Apo A-I of various other mammalian species, including human, bovine and rabbit, also inhibited acid CEH activity. Other apolipoproteins, such as apo A-II and apo B, also showed inhibiting activity. On the other hand, apo A-I had no effect on the activity of other enzymes found in lysosomes, such as cathepsin D, beta-glucuronidase and acid phosphatase. The results suggest that apolipoproteins may play a role in the regulation of hydrolysis of cholesteryl esters in lipoproteins, that have been transferred to the liver, and that the inhibition of acid CEH activity by apo A-I may be a characteristic of the lipid-binding protein or be due to changes of the lipid/water interface.

Changes in parathyroid hormone in diabetic patients on long-term hemodialysis.

Changes in parathyroid hormone (PTH) and osteocalcin over 3 years were studied in hemodialyzed patients with diabetic nephropathy (HD/DM) and hemodialyzed patients without diabetes (HD/non-DM). In HD/DM patients, concentrations of the carboxyl terminal regions of PTH and osteocalcin in the serum did not change significantly, but in HD/non-DM patients, both concentrations increased significantly. In patients in both groups, the mean concentration of the mid-region of PTH increased significantly. Secondary hyperparathyroidism in HD/DM develops slower than in HD/non-DM.

Inhibitory effect of heparin and/or antithrombin III on intraperitoneal fibrin formation in continuous ambulatory peritoneal dialysis.

The intraperitoneal fibrin formation and its inhibition by intraperitoneal heparin and/or antithrombin III (AT III) were examined in 8 patients on continuous ambulatory peritoneal dialysis (CAPD). With 1,000 and 2,000 U/L of heparin added to inflow dialysate, the concentration of fibrinopeptide A (FPA) in plasma decreased from 39.43 +/- 5.30 (mean +/- SEM) to 8.00 +/- 2.20 and to 0.74 +/- 0.12 ng/ml, respectively. The FPA concentration in outflow dialysate decreased from 34.20 +/- 5.75 to 12.94 +/- 2.10 ng/ml (1,000 U/l of heparin) and to 4.54 +/- 0.79 ng/mg (2,000 U/l of heparin). The AT III concentration was 0.47 +/- 0.07 mg/dl in dialysate and that in plasma was 24.20 +/- 2.76 mg/dl. With 100 U/bag of AT III added to inflow dialysate, the AT III concentration increased from 0.47 +/- 0.07 to 3.36 +/- 0.17 mg/dl in outflow dialysate but did not increase in plasma. The inhibition of fibrin formation of intraperitoneal heparin was increased by addition of AT III without a systemic inhibitory effect on fibrin formation. These data suggest that intraperitoneal administration of heparin without AT III would be sufficient for the purpose of preventing fibrin formation in CAPD patients without any trouble, and additional AT III might increase inhibitory effect of heparin.

[Primary coronary artery dissection: its incidence and genesis studied by clinical and coronary angiographic findings].

The incidence and genesis of primary coronary artery dissection which occurred spontaneously in the coronary artery system were elucidated in this study. We examined clinical and coronary angiographic features of the patients who presented this dissection. Primary coronary artery dissection was observed in 23 (0.43%) of 5,400 consecutive patients with ischemic heart disease undergoing coronary angiography. The basic diseases were myocardial infarction in 17 cases and angina pectoris in 6. Sixteen patients (70%) had smoking habits, which might be related to this disease. The dissection of the right coronary artery in 12 cases, of the left anterior descending artery in 10, and of the left circumflex artery in only one was found. All lesions occurred in the proximal positions of the coronary branches. Seven (30%) of all cases had fixed coronary stenosis in other coronary branches. The other cases had "one-vessel" disease. Seventeen patients were treated medically, 5 had PTCA therapy and one had CABG operation. All the patients, except one who died of cancer are surviving in a 3.8-year follow-up. Patients can be divided etiologically into three groups. The first was comprised of those in the post- or peri-partum period. The second was those associated with coronary vasospasm, and the third was those with atherosclerotic coronary artery disease. The third group was the most frequent.

Wheat nuclear protein HBP-1 binds to the hexameric sequence in the promoter of various plant genes.

HBP-1 is a sequence-specific DNA-binding protein that interacts with the hexameric sequence ACGTCA, the putative cis-acting element of the wheat histone H3 gene. Gel mobility shift and DNase I footprint analyses showed that this protein interacts with homologous sequences in the regulatory regions for the transcription of the cauliflower mosaic virus (CaMV) 35S RNA and nopaline synthase (NOS) genes, evidence that HBP-1 may bind to hexameric sequences in the regulatory regions of various genes. An HBP-1-like protein, indistinguishable from wheat HBP-1 in its the DNA-binding specificity, is present in sunflower nuclear extract, an indication that HBP-1-like DNA-binding proteins also exist in dicots.

Function of the hexameric sequence in the cauliflower mosaic virus 35S RNA promoter region.

The hexameric sequence ACGTCA functions in transcriptional regulation of wheat histone genes. The cauliflower mosaic virus (CaMV) 35S RNA promoter has the same hexameric sequence, and mutation analyses confirmed that the hexamer contributed greatly to transcription from the 35S promoter when a test gene with this promoter was introduced into sunflower cells. Electrophoretic mobility shift assays revealed the existence of a nuclear protein(s) in sunflower cells which is homologous to the HBP-1b that has been identified as binding to the 35S promoter in wheat. These results provide evidence of the involvement of the hexameric sequence and the HBP-1b-like DNA binding protein(s) in transcription from the 35S promoter.

Multiplicity of the DNA-binding protein HBP-1 specific to the conserved hexameric sequence ACGTCA in various plant gene promoters.

A novel DNA-binding protein that specifically interacts with the hexameric sequence ACGTCA in the regulatory region of the wheat histone H3 gene has been identified in wheat nuclear extract and designated HBP-1a. The nuclear protein HBP-1 previously identified as a DNA-binding protein that interacts with hexameric sequences in the H3, cauliflower mosaic virus (CaMV) 35 S RNA, and nopaline synthase (NOS) promoter regions therefore has been renamed HBP-1b. The flanking sequences that surround the hexameric sequence may account for the difference in the binding properties of HBP-1a and HBP-1b.

A protein that binds to a cis-acting element of wheat histone genes has a leucine zipper motif.

The structure and function of transcription factors of higher plants was studied by isolating cDNA clones encoding a wheat sequence-specific DNA binding protein. A hexameric nucleotide motif, ACGTCA, is located upstream from the TATA box of several plant histone genes. It has been suggested that this motif is essential for efficient transcription of the wheat histone H3 gene. A wheat nuclear protein, HBP-1 (histone DNA binding protein-1), which specifically binds to the hexameric motif, has previously been identified as a putative transcription factor. A cDNA clone encoding HBP-1 has been isolated on the basis of specific binding of HBP-1 to the hexameric motif. The deduced amino acid sequence indicates that HBP-1 contains the leucine zipper motif, which represents a characteristic property of several eukaryotic transcription factors.

Plasma endothelin levels in patients with uraemia.

Plasma immunoreactive endothelin concentrations were measured in patients with uraemia and in non-uraemic controls. Concentrations were beneath the detection limit of the assay in most patients without uraemia but were readily detectable in those undergoing haemodialysis. Plasma endothelin concentrations in patients with uraemia who were not undergoing haemodialysis were lower than in those on maintenance dialysis.
