Quantitative analysis of the action of Taka-amylase A on maltotriose.

The action of Taka-amylase A from Asp. oryzae was studied quantitatively by the product analysis method using unlabeled maltotriose and maltotriose labeled at the reducing end as substrates. It was found that the ratio of the unlabeled products, maltose (G2) and glucose (G1) exceeded unity, and that the ratio of the labeled products, G2/G1 was strongly dependent on the initial substrate concentrations. The results can only be explained by a transglycosylation or condensation mechanism or both. Analysis of maltose labeled at the nonreducing or reducing end reveal that the ratio of the transglycosylation to the condensation mechanism was about 20:1 with about 80 mM maltotriose. A computer simulation was made on a reaction scheme involving the termolecular-shifted complex, transglycosylation and condensation besides hydrolysis, by using reported subsite affinities as modified by the authors. The simulation reproduced the experimental results satisfactorily.

[Nursing procedures for cynomolgus monkey infants (author's transl)].

This paper describes the results of the artificial nursing of 8 infant cynomolgus monkeys (Macaca fascicularis) weighing 230-350 g at birth. The infants were separated from their mothers just after or 2 days after birth and were kept in a specially deviced small cage (Photo 1 & 2). They were fed on 6.4-16.6% solution of a commercially available powdered milk for the human baby. Apple juice also was given to them from the 3rd or 4th week on. In about the 6th week, the infants received solid foods such as small pieces of apple, biscuit and monkey chow. The main part of the dietary regimen was changed to the solid diet around the 12th week. The volume of milk taken was recorded every day, and the daily intake ff three major nutrients was estimated with the milk intake (Fig. 1,2,5,6). The body weights were between 600 and 750 g on the day of weaning, that is at about 12 weeks of age, except No. 2 infant showing a slight growth retardation (Fig.4). Their physical growth can be judged as normal as that of breast-nursed infants of the saame species from our laboratory. On the basis of these results, an example of the artificial feeding schedule for the infant cynomolgus monkeys was introduced (Fig. 7).

Crystallization and preliminary x-ray investigation of soybean beta-amylase.

Beta-Amylase [1, 4-alpha-D-glucan maltohydrolase, EC 3.2.1.2] has been purified from defatted soybean meal by fractional precipation with ammonium sulfate, ion-exchange chromatography on CM- and DEAE-Sephadex and gel filtration chromatography on Sephadex G-100. Two different components of beta-amylase were crystallized from ammonium sulfate solutions, and the homogeneity of each preparation was confirmed by sedimentation and disc electrophoretic analyses. Both components of soybean beta-amylase formed large single crystals (trigonal crystal system) from 40--50 per cent saturated ammonium sulfate solution buffered at pH 5.4 on dialyzing concentrated protein solution in the apparatus of Zeppezauer et al. Preliminary X-ray diffraction data gave a hexagonal lattice with unit cell dimensions a=86.1 A and c=144.4 A. The space group corresponds to P3121 or P3221, and one asymmetric unit contains one molecule of beta-amylase, assuming a crystal density of 1.25 g/ml and a molecular weight of the enzyme of 60,000 daltons. In this case, the crystal has a volume of 2.53 A-3 per atomic mass unit, and the percentage of protein in the crystal is about 52.

Approaches to the development of new poliovirus vaccines based on molecular genetics.

Complete nucleotide sequences of virion RNAs from immunologically distinct vaccine strains of poliovirus (i.e., Sabin strains 1, 2, and 3) were determined by use of molecular cloning and rapid sequence analysis techniques. When the nucleotide sequences were compared, extensive sequence homology was observed among the virion RNAs in the 5' and 3' terminal regions. This result strongly suggested that the serotypes of poliovirus were derived from a putative prototype poliovirus and that the homologous sequences conserved through a long evolutionary process may have essential roles for the replication and the viral functions of the virus. Many different amino acids, predicted from the nucleotide sequences, were located in polypeptides P1-1a, especially in P1-VP1, which is located on the surface of the virion and carries viral antigens. Locations of different amino acids, together with the calculated hydrophilicity values in P1-VP1, revealed possible loci for type-specific antigenic determinants. These data on functionally important conserved sequences and on possible locations of antigenic determinants will be useful in the construction of new polio vaccine strains.

Direct determination of diastereomeric carprofen glucuronides in human plasma and urine and preliminary measurements of stereoselective metabolic and renal elimination after oral administration of carprofen in man.

A direct stereospecific HPLC assay for carprofen glucuronides in biologic fluids was developed that makes use of a reverse phase (C 18) gradient system, in which the mobile phase consisted of a mixture of acetonitrile and tetrabutylammonium hydroxide buffer (pH 2.5). Reference diastereomeric glucuronides of carprofen were purified from human urine after oral administration of the single enantiomers. When 0.1 ml of sample was used, the limit of detection for carprofen glucuronides was 50 ng/ml in plasma and 200 ng/ml in urine. Coefficients of variation did not exceed 12% for both intra and interday variability. This HPLC method is applicable to pharmacokinetic analysis for carprofen glucuronides in humans. After oral administration of the single enantiomers there was little indication of metabolic inversion. For the two enantiomers the apparent total and metabolic clearances were similar. The limited data available suggest that renal clearance of the (S)-glucuronide was greater than that of the (R)-glucuronide.
