Anionic sites in blood capillaries of the mouse cochlear duct.

The blood-cochlear barrier, which consists of the molecular size and ionic charge barriers, is known to play an important role in production and absorption of inner ear fluids. In this study, we employed poly-L-lysine colloidal gold conjugates (PL-CG) in combination with Lowicryl K4M resin to demonstrate anionic sites in blood capillaries of the cochlear duct. Male ICR mice weighing 30-40 g with a positive Preyer's reflex were used. The basement membranes of blood capillaries of the stria vascularis and the spiral ligament were successfully labeled with PL-CG pH 2.5. The luminal surface of capillaries in the stria vascularis and the spiral ligament intensely reacted with PL-CG pH 2.5. However, PL-CG pH 1.0 stained only the basement membrane of the spiral ligament. Predigestion with several glycosidases nearly eliminated PL-CG labeling. Anionic charge located on the luminal surface of the endothelial cell was mainly caused by the presence of sialic acid. On the contrary, anionic charge of the basement membrane was caused in a substantial degree by chondroitin and heparan sulfate-rich glycosaminoglycans. We obtained histochemical evidence that blood capillaries of the stria vascularis differ from those of the spiral ligament.

Extracellular matrix components regulating glandular differentiation and the formation of basal lamina of a human pancreatic cancer cell line in vitro.

The interaction between the extracellular matrix and human tumor-cell clones S2-013 and S2-020, derived from a pancreatic cancer cell line (SUIT-2), was examined in vitro, using various cell differentiation-promoting matrices in two- and three-dimensional cultures. S2-013 cells (well-differentiated tubular adenocarcinoma in xenografts in nude mice) cultured in Matrigel formed glandular structures. Ultrastructural observation revealed a morphological polarity of cells and a distinct basal lamina. On the other hand, S2-020 cells (poorly differentiated tubular adenocarcinoma in xenografts) cultured in Matrigel formed neither glandular structures nor a basal lamina, but only cell aggregates. The morphology of these two sublines cultured in Matrigel expressed the histological degree of differentiation which they presented in nude mice. In contrast, in type I collagen gel, S2-013 cells formed glandular structures without a basal lamina, and in soft agar, they were able to form neither glandular structures nor a basal lamina. S2-020 cells cultured in type I collagen gel or soft agar formed the same simple cell aggregates as in Matrigel. Matrices used in a three-dimensional culture influenced the degree of differentiation in S2-013 cells but had no effect on the morphological differentiation in S2-020 cells. To detect the factors which induce basal lamina formation, S2-013 cells were cultured on a microporous membrane coated with extra-cellular matrix components such as laminin, type IV collagen, and fibronectin. S2-013 cells formed a basal lamina only on the laminin. These cell lines may be useful in investigating the mechanisms regulating the formation of glandular structures and basal lamina.

A study of electrical promontory stimulation in tinnitus patients.

Electrical promontory stimulation relieved tinnitus in 74 (57.4%) of 129 ears (112 patients). There was no significant difference in etiology of tinnitus, age, average audiogram, or tinnitus frequency between patients who responded to electrical stimulation and those who did not. Most patients who did not respond to the initial stimulation trial did not respond to the subsequent trials, suggesting that the initial response to treatment predicts the subsequent response. Patients who did not respond to repetitive treatment were supposed to be under severe stress.

Purification and immunohistochemistry of Griffonia simplicifolia agglutinin-II-binding mucus glycoprotein in rat stomach.

Gastric mucus is thought to protect the gastric wall from mechanical trauma, desiccation, pathogenic microorganisms, acid and proteases. We purified Griffonia simplicifolia agglutinin-II (GSA-II)-binding mucus glycoprotein (GMG) from rat gastric mucosa by solubilization in a guanidine-containing buffer, gel permeation chromatography, Ricinus communis agglutinin-I (RCA-I)-affinity chromatography and GSA-II-affinity chromatography. Rat GMG showed high molecular weight on a Sephacryl S-1000 column, and a single band in 0.5% agarose-2% polyacrylamide composite gels and blots. A protein of approximately 60 kDa was contained in the GMG preparation. GMG was deglycosylated with trifluoromethanesulphonic acid treatment. An antibody was raised against deglycosylated GMG (deGMG). The antibody recognized deGMG, GMG, periodic acid-treated deGMG and O-glycanase-digested deGMG, but did not react to trypsin-digested deGMG. These results suggest that the antibody recognizes proteinase-sensitive region or peptide backbone of GMG. In immunohistochemistry, the mucous gel layer of the stomach luminal surface was stained with antibody. The antibody recognized not only gastric mucous neck cells and pyloric gland cells, but also gastric surface mucous cells, mucous cells in the duodenal gland, and goblet cells in the small intestine and colon. These results indicate that GMG is a component of rat gastric mucus, and that the antibody recognizes mucous-secreting cells in rat stomach and intestine.

Anti-bacterial antibodies in Epstein-Barr virus (EBV)-transformed oligoclonal B-cell lines established from normal persons and autoimmune disease patients.

We have established 950 and 430 oligoclonal B-lymphoblastoid cell lines (LCL) from two normal persons and eight autoimmune disease patients, respectively by using Epstein-Barr virus (EBV)-induced transformation. To re-evaluate the EBV technique for production of human monoclonal antibodies (mAb) related to infectious disease, we screened these oligoclonal LCLs for antibodies against 31 bacterial strains systematically. A total of 74 cultures out of 1380 were reactive to a total of 18 strains out of 31. Among these, eight cultures showed 10(-3) antibody (Ab) titers to Pseudomonas aeruginosa serotypes C, E, F and I, Staphylococcus aureus, Serratia marcescens and Bacillus cereus. Ten cultures showed 10(-2) Ab titers to Ps. aeruginosa serotypes D, E, F and I, Ps. maltophilia, Staph. epidermidis, Klebsiella ozaenae, Ser. marcescens and B. subtilis. The results reveal the further possibilities for the EBV technique to produce various infectious disease-related human mAbs.

A protein-specific monoclonal antibody to rat liver beta 1-->4 galactosyltransferase and its application to immunohistochemistry.

We have produced a new protein-specific monoclonal antibody (MAb) to rat liver beta 1-->4 galactosyltransferase. This MAb, GTL2, was selected as the most reactive IgG to a periodate-treated antigen. Antigen and protein specificities of GTL2 were verified by immunoblotting of a non-glycosylated recombinant protein of human galactosyltransferase and enzymatically deglycosylated rat galactosyltransferase. Using GTL2, an immunohistochemical study was done in rat liver, epididymis, and salivary glands. Intense staining was observed in Golgi areas of epididymal duct epithelial cells, and submandibular and sublingual acinar cells. Hepatocytes showed weaker staining. Immunoelectron microscopic observation revealed that the staining was exclusively localized in trans-Golgi membranes of these cells.

[Glomerular IgA deposits in an autopsy study].

One hundred kidneys from 100 non-selected autopsy cases without any overt renal disease were examined by immunofluorescence to reveal the incidences and features of cases with clinically latent glomerular IgA deposits. Glomerular IgA deposits were found in 10 cases (10.0%), consisting of 4 with liver cirrhosis and 6 with other diseases. IgA deposition was observed in 4 of 13 cirrhotic patients (30.8%), 3 of 15 patients with gastrointestinal carcinoma (20.0%), one of 11 patients with cardiovascular disease (9.1%), one of 3 patients with fulminant hepatitis (33.3%), and one of 21 patients with broncho-pulmonary disease (4.8%). Light microscopy showed minor glomerular abnormalities in all non-cirrhotic cases with IgA deposits except in one case. By contrast, variable significant glomerular lesions were found in the cirrhotic cases with IgA deposits, for example mesangial proliferation and circumferential mesangial inter-position. Excluding 13 cases with liver cirrhosis, the results of urinalysis at the time of admission were available for the study in 55 of 87 cases. Forty-four of 55 cases showed normal urinalysis. Glomerular IgA deposition was found in 4 cases (9.1%) of 44 with normal urinalysis. It may be said that IgA deposition without clinical evidence of nephropathy occurred even in a normal population with an incidence of about 10%.

Goblet cell mucins as the selective barrier for the intestinal helminths: T-cell-independent alteration of goblet cell mucins by immunologically 'damaged' Nippostrongylus brasiliensis worms and its significance on the challenge infection with homologous and heterologous parasites.

The aim of this study was to examine the role of T cells on the alteration of terminal sugars of goblet cell mucins in the small intestinal mucosa of parasitized rats and to clarify the biological significance of the altered mucins in the mucosal defence against intestinal helminths. For this purpose, Nippostrongylus brasiliensis adult worms obtained from donor rats at 7 ('normal' worms) or 13 days ('damaged' worms) post-infection were implanted intraduodenally into euthymic and hypothymic (rnu/rnu) rats. Expulsion of implanted normal worms and associated goblet cell changes were extremely delayed in hypothymic recipients compared with euthymic recipients. In contrast, intraduodenally implanted damaged worms were expelled by day 5 regardless of the strains. Around the time of expulsion of implanted damaged worms, euthymic recipients showed both goblet cell hyperplasia and alteration of mucins, whereas hypothymic rats showed only the latter. Dexamethasone treatment completely abolished goblet cell changes of both strains of recipients. To clarify the importance of the constitutional changes of goblet cell mucins in mucosal defence, euthymic rats were primed by implantation of damaged worms to induce goblet cell changes, and then 3 or 5 days later they were challenged by implantation with normal worms. The results show that when goblet cell changes were induced by priming with damaged worms, recipient rats could completely prevent the establishment of normal worms. When hypothymic rats were primed and challenged in the same manner, a similar but slightly less preventive effect was observed. Such a protective effect of altered mucins seems to be selective because priming of euthymic rats with damaged N. brasiliensis did not affect the establishment of Strongyloides venezuelensis. These results suggest that: (1) once N. brasiliensis adult worms are 'damaged' by the host's T-cell-dependent immune mechanisms, they can induce alteration of sugar residues of goblet cell mucins via host-mediated, T-cell-independent processes; (2) the expression of such altered mucins is highly effective not only in causing expulsion of established damaged worms but also in preventing establishment of normal worms; and (3) the preventive effect of altered mucins is selective against parasite species.

Histochemical demonstration of mucin in lymphatic vessels of human middle ear cholesteatoma.

We investigated the possible existence of mucin in lymphatic vessels in cholesteatoma perimatrix using the periodic acid-thiocarbohydrazide-silver proteinate (PA-TCH-SP) method. Histochemical staining distinguished two types of lymphatic vessels, one of which contained PA-TCH-SP reacting substance showing a loose mesh-like appearance. Connective tissue was edematous around this vessel and was infiltrated by abundant round cells. The second type of lymphatic vessel did not contain PA-TCH-SP reacting substance and few round cells were seen infiltrating tissue around this vessel. Gland-like structures of mucous epithelium in the perimatrix were heavily stained by the PA-TCH-SP method. Secretory granules of the mucous epithelium and its luminal content had a loose mesh-like appearance. Since contents of the gland-like structures may leak through the chinks of epithelial cells into subepidermal connective tissues, the resultant inflow of mucin into the lymphatic vessels may then cause inflammation of the cholesteatoma perimatrix.

Thyroplasty type I with ceramic shim.

To improve hoarseness or misswallowing caused by unilateral recurrent laryngeal nerve paralysis, medialization of paralyzed vocal cord has frequently been performed. This method includes such techniques as injection method, insertion method, and arytenoid adduction, each presenting its merits and demerits. The insertion method which can be done while monitoring the patient's voice seems advantageous in that the technique is easy to perform and generally guarantees the voice improvement. Among insertion methods, Isshiki thyroplasty type I is the one most representative as well as popularized. However, since a silicone shim is inserted in this operation, it may be accompanied by the risks of carcinogenicity, foreign body reaction, and induction of collagen disease of silicone. Therefore we planned to use a ceramic as a safe substitute instead of silicone. There has been no article reporting the use of ceramic in this type of surgery. We used a fibrin glue to fix the ceramic shim and for hemostasis, which was found very useful. Hitherto, 2 cases of unilateral recurrent laryngeal nerve paralysis underwent Isshiki thyroplasty type I using ceramic shim with satisfactory results.

Human colorectal carcinoma-specific glycoconjugates detected by pokeweed mitogen lectin.

Pokeweed mitogen (PWM) lectin, known to bind branched poly-N-acetyllactosamines, has a highly selective affinity for human colorectal carcinomas. We performed light microscopic (LM) histochemistry with PWM lectin on paraffin sections of human colorectal tissues. In histological sections, normal mucosae and adenomas with mild dysplasia exhibited negative reaction (0/10, 0/13, respectively) with or without neuraminidase pre-digestion, whereas adenomas with moderate dysplasia showed a small increase in PWM lectin reactivity after neuraminidase digestion (4/23). In contrast, we observed a high incidence of positive reactivity in colorectal carcinoma without neuraminidase pre-digestion (38/44). After digestion with neuraminidase, there was increased reactivity of colorectal carcinomas in situ (7/12) and invasive carcinomas (13/32). These results imply that human colorectal carcinomas consistently contain substantial amounts of PWM-reactive branched poly-N-acetyllactosamine glycoconjugates structures. We also compared the staining patterns of PWM lectin and monoclonal antibodies (MAb) directed to Lewis X (LeX) or Lewis Y (LeY) antigen. PWM lectin reactivity was largely confined to the apical membranes of carcinoma tissues. MAb-LeX or MAb-LeY immunoreactivity was seen on the apical membranes and in the cytoplasm of both adenomas and carcinomas. Therefore, histochemical studies with this lectin should be useful for identification of carcinoma tissues and analysis of glycoconjugates associated with colorectal carcinoma.

[A case of chest wall abscess due to Salmonella newport].

A 18-year-old female with a history of precordial pain was admitted. There was a mass (3.5 x 2.5 cm) in the right inframammary region which was suspected to be chest wall tumor on the basis of CT scan and Ga scintigraphy findings. Exploratory surgery, consisting of total resection of the tumor without removing the ribs, was performed and revealed an abscess. Culture of pus from the abscess grew Salmonella newport (0 antigen 8, H antigen eh; 1.2). Focal infection with Salmonellosis is discussed. Surgical treatment is necessary in addition to chemotherapy.

Bromodeoxyuridine-immunohistochemistry on cellular differentiation and migration in the fundic gland of Xenopus laevis during development.

Cellular differentiation and migration in the fundic glands of adult and larval Xenopus laevis have been examined using bromodeoxyuridine-immunohistochemistry. In the adult fundic gland, cumulative labeling with bromodeoxyuridine revealed a proliferative cell zone between the surface mucous cells and mucous neck cells, in what is referred to as the neck portion of the gland. The labeling-index of mucous neck cells had rapidly increased by week-5. The labeling-index of oxynticopeptic cells showed a more delayed increase until week-7, coincident with the decrease in the labeling of mucous neck cells. In the immature fundic glands of larvae, the labeled proliferating cells were randomly distributed throughout the developing gastric mucosa. During metamorphosis, the labeling-index of immature epithelial cells was highest at stage 63. Following administration of bromodeoxyuridine at this stage, there was no significant loss of labeled epithelial cells during the metamorphosing period. Furthermore, there was no significant difference in the labeling-indices among the epithelial cells, such as surface mucous cells/generative cells, mucous neck cells, and oxynticopeptic cells, 7 days after administration. Cellular differentiation and migration pathways of epithelial cells in the fundic gland of adult X. laevis and its larvae are discussed.

Recording eye movement during stepping in place with a CCD (charge-coupled device) imagesensor.

A new system for recording eye movements using a charge-coupled device (CCD) imagesensor in a video camera, in which less than 0.5 degree of horizontal and vertical eye movements can be recorded, was applied in 7 subjects. Eye movements were recorded while they stepped in place. We observed 5 normal subjects, a patient with horizontal and vertical oscillopsia during walking, and one with a chronic limp. In the normal adults, the amplitude of vertical eye movement was less than 0.5 degree, and the horizontal eye movement was less than 2 degrees. The mean amplitude of horizontal and vertical eye movements in the subject who limped was larger than that of normal subjects. The patient who complained of vertical oscillopsia during walking showed the largest amplitude of vertical eye movement among any subject tested. The vertical oscillopsia in the patient was due to the largest amplitude of vertical eye movement. This system for recording eye movements will be useful in analyzing the mechanism of gaze fixation during walking.

Characterization of beta 1----4 galactosyltransferase purified from rat liver microsomes.

beta 1----4 Galactosyltransferase was purified from rat liver microsomes. Catalytic properties of the enzyme resembled those of previously purified soluble and membrane-bound beta 1----4 galactosyltransferases. The enzyme purified in the present study showed a major band around a molecular weight of 53,000 on SDS-PAGE. The NH2-terminal sequence of the enzyme was determined up to the 20th residue. The sequence was identical to the amino acid sequence from Ala-13 to Lys-32 deduced from mouse beta 1----4 galactosyltransferase cDNA. These results suggest that most of the mature enzyme in rat liver microsomes is produced by removal of the NH2-terminal 12 amino acids from a precursor polypeptide.

Sialomucin in middle ear cholesteatoma perimatrix.

Mucosubstance histochemistry of human middle ear cholesteatoma revealed that sialomucins are abundant and sulfomucins present in small amounts in the glandlike structures of the cholesteatoma perimatrix. Based on the study, various glycoproteins were injected into the dermis of the external ear canal and infiltrated into the tympanic membranes of guinea pigs. Tympanic membranes were obtained 7 days later and light- and electron-microscopically studied. Injection of a sialomucin from bovine submaxillary gland resulted in marked proliferation of epidermis and degeneration of the lamina propria. Asialomucin prepared from sialomucin by hydrolysis produced mild thickening of the epidermis but the lamina propria was not degenerated. Fetuin and bovine serum albumin did not cause proliferation of the epidermis. Cholesterol granuloma formed in some of the specimens injected with sialomucin. The presence of sialomucin in cholesteatoma perimatrix and these experimental studies using tympanic membranes of guinea pigs suggests that sialomucins participate in the proliferation of epidermis and degeneration of subepidermal connective tissue in human middle ear cholesteatoma.

Immunohistochemical localization of dipeptidyl aminopeptidase IV in thyroid papillary carcinoma.

The localization of dipeptidyl aminopeptidase IV expressed aberrantly in thyroid carcinoma was studied by immunoelectron microscopy using a monoclonal antibody to the enzyme with special reference to enzyme-histochemical staining of the enzyme. Five thyroid papillary carcinomas were investigated including two lymph-node metastases. All cases showed the dense immunoreaction product on the apical membrane and only traces of the product on lateral membranes, endoplasmic reticulum and nuclear membranes. In one case only, the dense product was observed on basal tubular structures. Analysis, using immunogold labelling on pre-embedded cryosections, revealed that dipeptidyl aminopeptidase IV was localized on the luminal surface of cancer cells. Two different distribution patterns of dipeptidyl aminopeptidase IV activity staining, diffuse and apical patterns, reported previously were thought to be due to different amounts of dipeptidyl aminopeptidase IV in the cytoplasm of cancer cells. This enzyme-histochemical staining method is useful for pathological diagnosis of thyroid tumours and can be applied to clinical materials. The enzyme localization is revealed by the staining pattern.
