Serum concentrations of 25-hydroxyvitamin D and 24,25-dihydroxyvitamin D in patients with various types of renal disease.

Serum concentrations of 25-hydroxyvitamin D (25OHD) and 24,25-dihydroxyvitamin D (24,25(OH)2D) in patients with various types of renal disease were measured by a competitive protein binding assay. There was a significant (P less than 0.001) inverse correlation between serum levels of either 25OHD or 24,25(OH)2D and the degree of proteinuria in patients with chronic glomerulonephritis or idiopathic nephrotic syndrome. The ratio of 24,25(OH)2D to 25OHD was relatively low in patients with creatinine clearances (CCr) less than 30 ml/min/1.48 m2, while the ratio was higher in those with clearances greater than 85 ml/min/1.48 m2. There was a linear correlation (r = 0.783, P less than 0.001) between the ratio and the CCr in patients whose CCR ranged from 30 to 85 ml/min/1.48 m2. The 24,25(OH)2D/25OHD ratio also appeared to be correlated significantly (P less than 0.001) with the PSP-test. The serum levels of 25OHD and 24,25(OH)2D were lowered in nephrotic patients during treatment with prednisolone. The serum levels of 24,25(OH)2D were increased by 1 alpha-hydroxyvitamin D3 treatment in patients with chronic renal failure.

Enhancement of granulocytic colony formation of deletion of phagocytic cells in the bone marrow patients with idiopathic aplastic anemia.

To study the pathogenesis of aplastic anemia, phagocytic cells from the bone marrow of patients with aplastic anemia were investigated. From 10 of 10 patients with this disease the concentration of CFUc in the bone marrow cells significantly increased after the removal of phagocytic cells. On the other hand, in bone marrow from only 4 of 27 controls, 22 non-aplastic anemia patients and 5 normal volunteers, the concentration of CFUc increased after the same treatment. From these findings, it may be inferred that phagocytic cells from the bone marrow of patients with aplastic anemia suppress proliferation and differentiation of granulocytic progenitor cells, and that this phenomenon is related to the development of this disease.

Effect of peripheral blood mononuclear cells from aplastic anemia patients on the granulocyte-macrophage and erythroid colony formation in samples from normal human bone marrow in vitro--a cooperative work.

Thirty-six patients with aplastic anemia and three with aplastic anemia-PNH syndrome in eight institutes were studied for the presence of peripheral blood mononuclear cells having a suppressive effect on granulocyte-macrophage (G-M) and erythroid colony formation using a uniform protocol. In 11 cases of 29 (38%) and 9 cases of 29 (31%), the presence of mononuclear cells with a suppressive effect on G-M and erythroid colony formation, respectively, was demonstrated. However, the presence of mononuclear cells suppressive both to G-M and erythroid colony formation was demonstrated only in 3 of 19 cases (16%).

A new technique for the cytochemical examination of human hemopoietic cells grown in agar gel.

In this paper, a new technique for the cytochemical examination of human hemopoietic cells grown in agar gel is described. All colonies in the agar gel in a plastic dish can be observed as a whole plate preparation after being stained cytochemically. Granulocytes and monocytes/macrophages in a cluster or a colony were easily distinguished from each other by esterase activity in response to naphthol AS-D chloroacetate of alpha-naphthyl acetate. To illustrate this technique's possibilities a time course study on the formation of granulocyte and monocyte/macrophage clusters or colonies from normal marrow cells was performed.

Salmon calcitonin-induced stimulation of 1 alpha,25-dihydroxycholecalciferol synthesis in rats involving a mechanism independent of adenosine 3':5'-cyclic monophosphate.

The effect of natural salmon calcitonin on accumulation in plasma of 1 alpha,25-dihydroxy-[3H]cholecalciferol from 25-hydroxy[3H]cholecalciferol in vivo was investigated in vitamin D-deficient thyroparathyroidectomized rats into which graded doses of the hormone were continuously infused by use of a balance study system. A dose-dependent increase in plasma concentrations of 1 alpha,25-dihydroxy[3H]cholecalciferol was observed with calcitonin infusion for 6--30h at a rate greater than 20 M.R.C. m-units/h. Infusion of parathyrin or cyclic AMP produced a similar stimulation [Horiuchi, Suda, Takahashi, Shimazawa & Ogata (1977) Endocrinoly 101, 969--974], but the maximal effect of calcitonin was additive to that of either parathyrin or cyclic AMP. Furthermore concurrent infusion of theophylline (0.5 mumol/h) did not potentiate the effect of submaximal doses (3 and 20 M.R.C. m-units/h) of calcitonin. Plasma concentrations of calcium showed a decrease with calcitonin infusion for 30h, but those of Pi remained unchanged. These results strongly suggest that the rat kidney is endowed with a calcitonin-sensitive 1 alpha-hydroxylase system that is separate from the parathyrin/cyclic AMP system and is independent of changes in plasma Pi.

The decreased membrane fluidity of in vivo aged, human erythrocytes. A spin label study.

The decreased membrane fluidity of the in vivo aged, human erythrocytes is found, by monitoring the electron paramagnetic resonance (EPR) spectra of fatty acid spin labels incorporated into the membrane. In addition, the decreased cell sizes and the decreased cholesterol and phospholipids contents, without significant changes of the quantity of the membrane proteins, also the decrease of ATP and 2,3-diphosphoglycerate and the increase of ADP and AMP, in the aged cells, were observed. Further the functional impairments of the aged cells, i.e. the increased oxygen affinity and the decreased deformability, were shown. On the basis of these quantitative data, the alteration of the protein-lipid organization, due to decreased lipid/protein ratio, the modified protein-lipid interaction and/or the influences of the diminished ATP content, is suggested to contribute towards the decreased membrane fluidity of the in vivo aged erythrocytes.

Failure to demonstrate stimulatory effect of prolactin on vitamin D metabolism in vitamin-D-deficient rats.

The effect of administration of bovine prolactin in vivo on renal 1 alpha-hydroxylase (1 alpha-OHase) activity was investigated using thyroparathyroidectomized and sham-operated vitamin-D-deficient male rats. A high dose (1.5 IU/hr) and a low dose (0.3 IU/hr) of bovine prolactin was constantly infused into the cannulated femoral vein for 6 hr or 30 hr. The accumulation of 1 alpha, 25-hydroxyvitamin D3 [1 alpha, 25-(OH)2-D3] in plasma produced from 25-hydroxy-vitamin D3 (25-OH-D3) was calculated from the 6-hr conversion of intravenously injected [3H]-25-OH-D3 to [3H]-1 alpha, 25-(OH)2-D3. Both high dose and low dose of prolactin administration caused no significant stimulatory effect on 1 alpha-OHase activity in TPTX animals. Also, in sham-operated rats the prolactin treatment produced no significant change in the 1 alpha-OHase activity. Plasma calcium concentrations showed a tendency to increase slightly. These results suggest that prolactin has no direct stimulatory role on renal 1 alpha-OHase activity in rats. Its effect on calcium metabolism may be mediated by a different mechanism(s).

Lymphoma of the brain associated with polyposis of the colon--report of a case and review of Turcot's syndrome.

Pathological findings of a 15-year-old girl with polyposis of the colon who has subsequently developed primary lymphoma of the brain are reported. The authors consider this case to be a rare example of Turcot's syndrome although the histological typing of the brain tumor is not classical and familial background of the polyposis has not been demonstrated. Literatures on 14 previously reported cases of Turcot's syndrome are reviewed and clinical and pathological findings are summarized. The present case is the second report of Turcot's syndrome associated with lymphoma of the brain and is unique in regard to the age as cerebral lymphoma is extremely rare in the second decade.

ACTH, beta-LPH and beta-endorphin in pituitary adenomas of the patients with Cushing's disease: activation of beta-LPH conversion to beta-endorphin.

ACTH, beta-lipotropin (beta-LPH) and beta-endorphin concentrations were determined in pituitary adenomas of the patients with Cushing's disease. Immunoreactive ACTH and beta-endorphin were present in high concentrations and essentially equimolar amounts in pituitary adenomas. beta-LPH conversion to beta-endorphin was activated in pituitaries associated with ACTH/beta-LPH producing adenomas. Immunoreactive ACTH and beta-endorphin concentrations were markedly suppressed in the surrounding tissues.

A kinetic measurement of red cell deformability: a modified micropipette aspiration technique.

A kinetic measurement of the red cell deformability is developed, modifying the micropipette aspiration technique of BRAASCH (1971). The sedimented human red cells on a slide-glass are individually aspirated into a micropipette by negative pressure (delta P), and the decreasing "velocity (v)" of the applied electric current, due to the aspirating cell, is taken as a measure of the "easiness" of the cell to enter into the small orifice of the micropipette (inner diameter, 2 R congruent to 3 micrometer). An empirical relation, v infinity (pi r2) . (delta P) . ("deformability"), is obtained. In order to test the validity of the method, some factors influencing the deformability are studied: i.e., the "velocity" decreases as lowering temperature; the "velocity" of the glutaraldehyde-pretreated cells and of both in vivo and in vitro aged cells decreases. Therefore, the parameter, "velocity (v)," reflects the deformability well. In addition, if a few assumptions could be allowed, v would be related to the Young's modulus of the membrane.
